Abstract

Hatchery and culture technology of Humpback grouper has been developed. However,  sometimes it is still found constraint in sedd production and caused instability of production due to failure by many factors. Breeding program become necessary to provide seed with high quality and good genetic variation. Quantitative characteristic selection is one of breeding program to improve productivity in grouper aquaculture. The aim of this research was to produce good phenotyphic and genotypic quality of Humpback grouper candidate broodstock. Selection of Humpback grouper F1 and F3 was done based on quantitative characteristic (body length and weight) and genotyphic characteristic by using ramdom amplified polymorphism DNA (RAPD) method.  The results of conventional selection were found candidate broodstock of Humpback grouper F1 and F3 with body weight range from 170-210 g and 160-170 g consecutively.  Heterozygosity values of Humpback grouper analized by RAPD of F1 and F3 were 0.7940 and 0.7749 consecutively and it was not significantly different. This value emphasis that population of this F3 Humpback grouper was still good to grow for broodstock. Keywords: genetic variation, humpback grouper, F1, F3.

Abstract

This study aimed to determine the effect of LHRH-a hormone implantation on gonadal development of humpback grouper, Cromileptes altivelis 2nd generation (F-2). Individuals used were 20 female grouper fish with weight range of 600-800 gram/fish and total length range of 28.0-30.0 cm. The treatment used 5 fish each with tagging. Grouper fishes were stocked into the 75 m3 circular concrete tank. Hormone of LHRH-a with dosage of 50 ?g/kg body weight was used as implant treatment of (A) 1 time,, (B) 2 times, (C) 3 times, and (D) without implant. The results showed that implantation of LHRH-a hormone were significantly stimulate gonad development of humpback grouper second generation (F-2). Treatment of 3 times implant every month showed the best results produced  oocyte diameter up to 480 ?m, while the control of oocyte diameter only reached <400 ?m. Abstract

Grouper seeding technology still relies on the broodstock from the ocean, eventhough, the existence of the broodstock is rare and difficult to obtain. The broodstock takes a long time to mature and spawn. Therefore, to anticipate the scarcity humpback grouper from wild, it is important to produce broodfish from aquaculture. This study was aimed to know the reproduction performance of humpback grouper wild broodstock F-0, first generation broodstock (F-1) and second generation broodstock (F-2). The cultured of humback grouper broodstock used three tanks 75 m3 in volume (tank I, II and tank III). Tank I: reared 35 fishes broodstock F-0. Tank II and tank III for reared each 50 fishes F-1 and F-2 broodstock. Food for broodstock from the wild (F-0) was given from fresh trash fish, squid, and added vitamin mixture, vitamin C and vitamin E. Meanwhhile, food given to F-1 and F-2 broodstock was a commercial dry pellet (PG 9-10) that contains following nutrients: protein content of 43%, min. fat of 9%, ash content of 13%, fiber levels of 2%, and moisture content of 12%. Mixture of vitamin C and E were also added. The results showed that the humpback grouper F-0, F-1 and F-2 were cultured in 75 m3 concrete tank was successfully spawn with the fertilization rate of 87%, 78% and 45% respectively. This result proves that the broodstock from F-0 gaved the best results.Keywords: Humpback grouper reproduction, wild broodstock (F-0), first generation (F-1), seccond generation (F-2)

Abstract

The purpose of this research was to find the optimum color concentration of water for larval rearing of humbpback grouper (Cromileptes altivelis). The experiment conducted using 12 polycarbonat tanks with volume of 200 l. Initial density of larvae was 10 ind/l. Food grade coloring agent was used to result a green color water solution.  Four different water color concentration were tested as treatments i.e.: A 10 ppm; B 20 ppm ; C 30 ppm and D 40 ppm with three replications. The experiment was conducted for 30 days. The result showed that the highest survival rate was exhibited by C treatment (30 ppm) (P<0.05), the B (20 ppm) and D (40 ppm) treatments were significantly different (P<0.05) compare to A (10 ppm.) The survival rates of larvae at the end of experiment were A (8.2%); B (25.6%); C (40.4%) and D (29.3%).

Abstract

An experiment to evaluate the effectiveness immunostimulant and bacterin on humpback grouper have been conducted at the Disease Laboratory of Research Institute for Mariculture, Gondol, Bali. The experiment was designed in completely randomized design with four treatments in duplicates. Two hundred of humpback grouper juveniles (15-18 cm of total length, 55-65 g of body weight) were injected intraperitoneally with (A) bacterine at 107 cfu/kg body weight (BW), (B) peptidoglycan at 100 mg/kg BW, and (C) immuno star at 100 ml/kg BW, (D) control. The fish were then challenged with VNN by intramuscular injection at 10 days post treatment. Results showed that survival rates of juvenile after challenged with VNN were 60.00% (B & C), 53.34% (A), and 1.67% (control). Parameters of non-spesific immune respons showed that phagocytic activities were 17.56% (B), 17.55% (C), 13.11% (A), and 9.33% (control). In addition, lisozyme activities were 1.64 cm (B), 1.58 cm (C), 1.55 cm (A), and 1.46 cm (control). Immunostimulant and bacterin stimulated non-specific immune response, and increased survival rate of humpback grouper juvenile.

Abstract

The purpose of this experiment was to know the effect of implantation LHRH-a hormone on gonade maturation of grouper Cromileptes altivelis. The weight of female broodfishes were used between 1.40 and 1.65 kg/pcs and total lenght between 41.0 and 48.0 cm. Some of 5 fishes were stocked into the 75 m3 circular concrete tank. Abstract

Observation on parasitic diseases had been conducted in Gondol Research Institute for Marine culture on mild fish (Chanos chanos), seabass (Lates calcarifer), tiger grouper (Epinephelus fuscoguttatus), humpback grouper (Cromileptes altivelis), mangrove grouper (E. coioides), camouflage grouper (E. microdon), coral trout (Plectropoma leopardus, P. maculatus), snappers (Lutjanus argentimaculatus, L. seabae) and napoleon wrasse (Cheilinus undulatus) rearing in captivity. The result showed that Cryptocaryon irritans was found on napoleon wrasse and humpback grouper; Amyloodinium ocellatum was found only on napoleon wrasse; Gill flukes (Haliothrema sp. dan Diplectanum sp. ) was found on humpback grouper and mangrove grouper; skin flukes Neobenedenia sp. was found only on napoleon wrasse and Benedenia was found on all grouper, snapper and seabass; Crustacea (Rhexanella sp.) and copepod (Caligus sp. and Lepeophtheirus sp.) was found on all grouper, snapper and seabass; Hirudinae (Zeylanicobdella arugamensis ) was found on all grouper, snapper and seabass.

Abstract

The purpose of this experiment was to know gonad maturation and spawning frequency of the first generation in humpback grouper, Cromileptes altivelis. This experiment had two groups of feed types (A and B). Both groups were put in circular concrete tanks with water volume of 75 m3 each. Each tank was stocked 40 fishes with body weight of 500—1,000 g/fish and total length of 28—38 cm. Group (A) was fed with trash fish, squid, and vitamin mix, while group (B) was fed with artificial diet. Both groups were fed once a day, in the morning. The water was exchange continuously, of 300%—500% per day. The result showed the fish spawn after 31 months rearing. During the experiment, the range of water temperature and salinity were 28.4°C—31.7°C and 30.2—34.4 ppt, respectively.

Abstract

Dengan keberhasilan produksi benih kerapu bebek, masih banyak kendalakendala yang harus diperbaiki di antaranya kualitas benih, yaitu morfologi benih kerapu bebek dari hasil produksi perbenihan masih ada yang cacat seperti insang terbuka, mulut bengkok, tulang belakang bengkok, dan sebagainya yang akan mengganggu pertumbuhan bila dibudidayakan, selain mempunyai harga yang murah dan rentan terhadap penyakit. Diduga salah satu penyebab cacat adalah tidak berkembangnya gelembung renang pada larva yang diakibatkan ketidakmampuan larva mengambil udara di permukaan air yang tertutup oleh lapisan minyak. Karena itu untuk membersihkan lapisan minyak di permukaan air perlu adanya penelitian penggunaan skimmer dengan waktu yang berbeda yaitu: saat larva berumur 6 hari; 15 hari, dan tanpa skimmer. Hasil yang diperoleh adalah penggunaan skimmer pada larva berumur 6 hari menghasilkan sintasan dan pertumbuhan lebih baik di samping mempunyai persentase perkembangan gelembung renang yang lebih tinggi dan persentase benih yang cacat lebih rendah dibanding dengan perlakuan yang lain.

Abstract

Kerapu bebek/kerapu tikus/humpback grouper (Cromileptes altivelis) merupakan salah satu komoditas perikanan budidaya laut yang memiliki nilai jual tinggi. Sebelum berkembangnya kerapu hibrid, Balai Besar Penelitian dan Pengembangan Budidaya Laut Gondol, sudah berhasil mengembangkan pembenihan ikan kerapu bebek meskipun sintasan yang diperoleh masih bervariasi. Pemantauan benih hasil budidaya sebagian besar terfokus pada pertumbuhan dan jika sudah mencapai ukuran konsumsi bisa dijual atau diekspor ke Hongkong. Sebagai institusi penelitian nampaknya masih perlu mengupas lebih detail komoditas yang dikembangkan. Beberapa catatan yang dapat diperhatikan, untuk lebih mengenal komoditas kerapu bebek, sebelum dijadikan sebagai ikon jenis ikan yang pernah memiliki masa kejayaan, karena saat ini popularitasnya relatif menurun karena ada larangan ekspor dan masuk dalam satwa CITES. Catatan tersebut antara lain; kerapu bebek selama fase yuwana (2-3 bulan) dapat dijadikan sebagai kandidat ikan hias dengan performasi bintik bintik hitam pada permukaan tubuh yang merupakan keunikan tersendiri, karena jumlah dan polanya berbeda antara satu individu dengan yang lainnya serta akan bertambah jumlahnya seiring dengan bertambahnya umur dan bobot badan. Spot/bintik hitam kemungkinan identik dengan sidik jari manusia, yang bisa digunakan untuk membedakan antara satu individu dengan individu lainnya dalam satu populasi. Gonad kerapu bebek turunan pertama (F1) dengan umur dan kisaran bobot badan yang kurang lebih sama (600-900 g), menunjukkan 95% berada pada fase betina. Sementara pematangan dan pembentukan gonad jantan pada kerapu bebek hasil budidaya dapat dipacu dengan rangsangan hormonal dan pemberian pakan segar dengan kualitas yang baik. Dari hasil pengamatan diperoleh informasi bahwa kerapu bebek hasil budidaya dapat memijah dengan baik dan menghasilkan turunan generasi kedua (F2), akan tetapi kualitas telur dan larva yang dihasilkan masih lebih rendah bila dibandingkan dengan larva pemijahan induk alam (F0).
