Abstract

Learning is one of the obligations of students to do in every school activity where they study. However, sometimes many students are less able to digest the subject matter delivered by the teacher. Therefore, the school held a scholarship program for outstanding students. In order to motivate students to study harder. Achievement scholarships are given with the aim of motivating students to study harder. Currently the scholarship is not right on target because it is still done manually and it is not clear the criteria for a student to get an achievement scholarship. To conduct an assessment in awarding scholarships to high achieving students use a decision support system to help solve a problem. For this reason, to conduct an assessment in the awarding of scholarships, a decision support system using the Profile Matching method is used. Profile Matching method is one of the methods used in decision making. In this study, there are several aspects of the assessment for awarding achievement scholarships, namely the KKM Aspect, the Attendance Aspect, the Behavior Aspect, the Craft Aspect or the Discipline, the Neatness Aspect.

Abstract

ABSTRAKPREPARASI RADIOFARMAKA 99mTc__DTPA_INH UNTUK DIAGNOSIS TUBERKULOSIS.Radiofarmaka telah menunjukkan manfaat yang nyata dan spesifik dalam pelayanan kesehatan terutama untuk diagnosis dan terapi antara lain untuk penyakit kanker dan infeksi. Salah satu cara. menangani banyaknya kasus tuberkulosis yang banyak terdapat di Indonesia, teknik kedokteran nuklir menggunakan radiofarmaka diethylenefriaminepenfaacetic acid-isonicotinic acid hydrazide (DTP A-INH) yang yang ditandai dengan teknesium-99m dapat dipakai salah satu metoda altematif untuk diagnosis tuberkulosis. Saat ini telah dilakukan preparasi 99mTc_DTP A-INH, 991~C_DTPA-INH dilakukan melalui beberapa tahapan yaitu: konyugasi DTPA-INH dan analisis hasil konyugasi dengan HPLC kolom C-18. Penandaan DTP A-INH dengan Tc-99m dilakukan dengan menambahkan SnClz sebagai reduktor dan larutan 99mTcperteknetat dan generator 99Mo;99mTc.Hasil penandaan dianalisis dengan menggunakan kromatografi kertas dan kromatografi lapis tipis, sedangkan stabilitas DTP A-INH dilakukan untuk menentukan waktu kadaluwarsanya. Hasil analisi dengan HPLC menggunakan kolom C-18 menunjukkan telah terjadi konyugasi antara DTPA dengan INH, sedangkan analisis dengan kromatografi kertas dan kromatografi lapis tipis menunjukkan kemurnian radiokimia lebih besar dari 90 % dan hasil pelabelan terbaik didapatkan pada penambahan SnClz sebanyak 200 ).lg. Hasil uji stabilitas untuk sediaan yang belum dilabel, menunjukkan sediaan masih stabil pada minggu ke 13. Pengujian kestabilan 99mTc_DTPA-INH pada suhu kamar menunjukkan kemurnian radiokimia masih stabil sampai 3 jam setelah penandaan. Kata kunci : radiofarmaka, Tc-99m, DTP A, INH, diagnosis, tuberkulosis. ABSTRACT PREPARATION OF 99mTc-DTPA-INH RADIO PHARMACEUTICALS FOR TUBERCULOSIS DIAGNOSIS. Radiopharmaceuticals have shown a real and spesifik usefulness in medical services, especially for diagnosis and therapy of several deadly diseases such as cancer and infection. To handle Tubeculosis which is common in Indonesia, nuclear medicine teclmiques which uses DTPA-INH radiophannaceutical labeled with technetium-99m offer an alternative method for tuberculosis 99m diagnosis. Preparation of Tc-DTP A-INH and its analysis have been carried out. The preparation consisted 99111 of several steps, conjugation of DTPA with INH and the conjugated DTPA-INH was labeled with Tc, 99m followed by C-18 HPLC analysis. Radiochemical purity of Tc-DTP A-INH was ana lysed using TLC/paper chromatography. Stability test of DTP A-INH to know the expiry date was carried out. The HPLC result showed that the conjugated DTP A-INH has been formed. The radiochemical purity of 99mTc_TPA-INH analysed with TLC/paper chromatography was obtained at higher than 90 %. Best result was obtained by addition of 200 ).lgram of SnClz. The stability test showed that DTP A-INH was stable until 13 weeks and thestability of labeled DTP A-INH at room temperature was stable after 3 hours incubation post labeling. Abstract

Information technology is very important in influencing organizations’ performance to win over the competition. The role of information technology and the high investment resulted in the increasing demands to value provided by information technology in businesses. Human’s need associated with technology development also gives impact to education, and this will cause the emergence of a new paradigm in the education sector. This study uses Luftman Method and conducted at SMK PUSTEK SERPONG. The study finds that there is no harmony between the strategy of information technology and the business strategy in SMK PUSTEK SERPONG. Therefore, it is important to restructure these strategies and systemize the priority steps to improve the performance of the strategy of information technology and business strategy.

Abstract

Folklore was born as the product of the culture in society; it can be in the form of myth, legend, or tale. In folklore, there are cultural value and wisdom that can be used as the guidance of life for the society, such as, democratic value, the value of hard-working, the value of honesty, moral, and love to their country. Those values lie in Cilacap’s folklore. Their existence takes part in the increase of tourism visit, which, at the end, hopefully, can increase the society’s economic level.

Abstract

Immunoradiometric assay (IRMA) is a method of analysis based on immunological reactions of antigens-antibodies binding. This highly specific and sensitive method was used for in vitro diagnosis in small quantity of sample. Center for Radioisotope and Radiopharmaceutical Technology, BATAN has developed Thyroglobulin IRMA Kit using coated tube method that can determine thyroglobulin levels in microgram quantities. Coated tube was made with immobilisation of anti thyroglobulin into polistyrene tube. Development of IRMA kit performed through several steps including: optimization component of kit, optimization assay and kit validation. Optimization of coated tube involved selection and volume of solvent, using blocking and non-blocking agent, and volume of blocking agent. The optimum condition for coated tubes was found to be using 0.1M phosphate buffer pH 7.4 with coating volume of 500 μL, 3% BSA in 500 μL blocking agent 0.1M phosphate buffer pH 7.4, with maximum binding and non-specific binding (NSB) of 60.58 and 1.40%, respectively. The optimized coated tube was found to be stable up to 4 weeks.

Abstract

TSH IRMA kit is a kit used for the determination of TSH (Thyroid Stimulating Hormone) levels in human blood serum. Thyroid hormone is a hormone that our bodies need for growth of the brain, bone and other tissues and regulate the metabolism in the body. TSH normal range for adult is in the range of 0.4-4.5 mIU/L, whereas for baby is about 3.0-18.0 mIU/L. Thyroid would affect the quality of optimal growth of children if disturbed. Therefore, TSH assay in the blood needs to be determined to know whether the function of the thyroid gland works normally or not. Detection of TSH in blood can be performed by Immunoradiometricassay (IRMA) method. IRMA method is one of the immunoassay techniques based on immunological reactions (antigen-antibody binding) using radionuclide 125I as a tracer, that sample in small quantity can be detected.  IRMA method was developed locally by replacing TSH IRMA kit which is costly since imported from commercial companies. Center for Radioisotope and Radiopharmaceutical Technology (PTRR) BATAN has successfully developed the TSH IRMA kit that can be used to determine the levels of TSH in human blood. TSH IRMA kit must be validated to know the limit of detection, sensitivity, accuracy, precision and the assay parameters, such as Non-Specific Binding (NSB) and Maximum Binding (MB). Validation of TSH IRMA kit had been carried out resulting in the limit of detection of 0.115 ng/mL, accuracy with a recovery of 93.6-108.0 %, intra-assay precision (% CV) QC L = 1.9848, QC M = 3.6360 % and QC H = 2.2085 % while the inter-assay precision (% CV) QC L = 11.0055, QC M = 5.6768 %  and  QC H = 5.4181 %.  It was concluded that this TSH IRMA kit showed good performance based on the % NSB and % B/T of 0.68 and 34.64 %, respectively.

Abstract

Aflatoksin merupakan senyawa mikotoksin yang bersifat sangat toksik sehingga dapat menjadi penyebab terjadinya kanker pada manusia. Aflatoksin berpotensi karsinogenik, mutagenik, teratogenik, dan bersifat imunosupresif oleh karena itu kandungan aflatoksin B1 dalam bahan dan prod uk pangan harus dibatasi. Salah satu teknik penentuan kadar aflatoksin B1 adalah radioimmunoassay (RIA) yang didasarkan pada reaksi immunologi antara antigen dan antibodi yang spesifik hanya untuk antigen tertentu saja, serta menggunakan antigen yang ditandai zat radioaktif sebagai peru nut. Pusat Teknologi Radioisotop danRadiofarmaka BATAN telah berhasil mengembangkan kit RIA Aflatoksin B1 yang dapat digunakan untuk penentuan kandungan Aflatoksin B1dalam bahan dan produk pangan. Sebelum digunakan di lapangan kit aflatoksin B1 harus divalidasi meliputi penentuan batas deteksi, kepekaan (sensitivitas), ketelitian (presisi) dan parameter assay (Non Spesific Binding, NSB dan Maximum Binding, MB) sehingga dapat digunakan untuk menentukan kadar aflatoksin B1? Penelitian ini bertujuan untuk menentukan batas deteksi, ketelitian intra assay dan inter assay serta parameter assay. Telah dilakukan validasi kit RIA aflatoksin yang menghasilkan batas deteksi 0,35 ng/mL dengan ketelitian intra assay memberikan koefisien variasi (%CV) QC 9,80% sedangkan ketelitian inter assay untuk QC 12,39%. Kit RIA aflatoksin B1 ini disimpulkan memberikan unjuk kerja yang baik karena menghasilkan %NSB sebesar 6,6 dan B/T sebesar 47,18. Aflatoxins are mycotoxins compounds that are highly toxic and carcinogenic. Aflatoxins are potentially carcinogenic, mutagenic, teratogenic, and immunosuppressive so that the content of aflatoxin B1 in food products should be limited. One technique of determining the level of aflatoxin B1 is a radioimmunoassay (RIA) which is based on immunological reactions between antigens and antibodies, and using radioactive substances as a tracer. Center for Radioisotopes and Radiopharmaceuticals Technology (PTRR) has successfully developed Aflatoxin B1 RIA kit that can be used to determine the aflatoxin B1 in food products. Aflatoxin B1 RIA kit must be validated, which includes determining the limits of detection, sensitivity, accuracy (precision) and assay parameters (Non Specific Binding, NSB and Maximum Binding, MB) that can be used to determine the level of aflatoxin B1. This study aims to determine the limit of detection, accuracy intra-assay and inter-assay and assay parameters. The Aflatoxin B1 RIA kit validation results in the detection limit of 0.35 ng / mL with coefficient of variation (% CV) QC 9.80%, while the inter-assay precision for QC 12.39%. RIA Kit Aflatoxin B1 is inferred provide good performance because it produces 6.6% for NSBand 47.18 for B/T.

Abstract

ABSTRAK OPTIMALISASI PEMBUATAN KIT IRMA CA-125. Telah dilakukan penelitian optimalisasi pembuatan kit  IRMA  CA-125 yang meliputi pembuatan perunut monoklonal anti CA-125 bertanda 125I dan  pembuatan  "coated tube". Pada pembuatan perunut digunakan 2 macam oksidator yang divariasikan jumlahnya, yaitu kloramin-T dan N- bromosuksinimid.  Optimalisasi pembuatan "coated tube" dilakukan dengan memvariasikan volume larutan "coating", pengaruh pencucian, pengaruh penambahan BSA, pengaruh   bufer  pelarut dan uji pemasangan monoklo-nal anti CA-l25. Hasil percobaan menunjukkan bahwa oksidator  kloramin-T dapat digunakan untuk pembuatan perunut mononoklonl  anti CA-l25 dengan rendemen cukup tinggi yaitu 96.5% dan kemumian radiokimia 93,2%. Jumlah oksidator yang optimal adalah 10 µg. Pembuatan "coated tube  untuk  monoklonal anti CA-l25 dengan menggunakan volume larutan "coating" yang bervariasi antara 300 µI sampai dengan 600 uL tidak menunjukkan perbedaan nyata pada nilai %NSB dan %BT. Proses pencuci "coated tube" menggunakan BSA 0, I % memberikan pengaruh nyata pada %BT. walaupun tidak berpengaruh terhadap nilai %NSB. Penggunaan bufer bikarbonat untuk pembuatan "coated tube" memberikan hasil yang lebih baik dari pada bufer fosfat. Untuk pemasangan monoklonal anti CA-125 diketahui bahwa pasangan monoklonal anti CA-125. monoklonal anti CA-125  M37203M sebagai  perunut dan monoklonal anti CA-125  M86924M sebagai "coating solution" memberikan hasil yang optimal. Optimalization on thepreparation of CA-125 IRMA-kit has been studied involving preparation of 125I  labeled monoclonalanti CA-125 as a tracer and preparation of coated tubes. Chloramine-T and N-bromosuccinimide were used as oxidators by varying them in their quantity. Optimalization on preparation of coated tube was carried out by variation in volume of  coating solution, effect of washing, addition of BSA 0.1%, buffer and pairing test for the monoclonal anti CA-125. The use of chloramine-T as oxidator for 125I labeling of monoclonal anti CA-15 was found to giv labeling-yield of about 96.5% and radiochemical purity of 93.2%. Optiimum amount of the oxidizing agent was found to be 10µg. There were not any significant difference in utilization of coating solution volume ranging at 300 to 600µL. for preparing monoclonal anti CA-125 coated tube. The non specific binding (NSB) and the %B/T values were found to be unaffected  by washing step, while a blocking of coated tube using of BSA 0.1% decreased the %B/T value. Utilization of bicarbonate buffer for the preparation  of coated tube was found to be more satisfactory than that of phosphate buffer. It was shown that the monoclonal anti CA-125 M37203 used for tracer being paired with monoclonal anti CA-125 M866924M used for coating solution gave an optimal result based on the values of %NSB  and %B/T. Abstract

Triningsih , 2017  Peningkatan Hasil Belajar IPA tentang penyebab perubahan benda melalui Penggunaan Metode Inkuiri di Kelas VI SD Negeri Bedahan 01. Penelitian Tindakan Kelas (PTK). Rumusan masalah yang disusun adalah “Apakah penggunaan metode inkuiri dapat meningkatkan hasil belajar siswa dalam pembelajaran IPA terhadap konsep penyebab perubahan benda?” Penelitian Tindakan Kelas ini bertujuan untuk meningkatkan hasil belajar siswa dalam pembelajaran IPA kelas VI melalui penggunaan metode inkuiri dan meningkatkan motivasi belajar siswa dalam pembelajaran IPA kelas VI melalui penggunaan metode inkuiri. Prosedur Penelitian Tindakan Kelas dilaksanakan melalui proses beralur terdiri dari 4 tahap, yaitu: 1) perencanaan; 2) pelaksanaan; 3) Observasi; dan 4) refleksi. Dari analisis data diketahui bahwa pada setiap siklus terjadi peningkatan ketuntasan hasil belajar siswa. Pada kondisi awal, siswa yang mencapai ketuntasan baru 44,44%. Pada siklus I siswa yang mencapai ketuntasan mengalami kenaikan 50% dari kondisi awal menjadi 66,67%. Pada siklus II mengalami kenaikan 50% menjadi 100%. Hal yang sama juga terjadi pada kesungguhan belajar siswa. Berdasarkan hasil analisis data penelitian ini dapat disimpulkan bahwa: Penggunaan metode inkuiri mampu mempermudah siswa dalam memahami materi pembelajaran; Penggunaan metode inkuiri mampu meningkatkan kesungguhan siswa dalam belajar; Penggunaan metode inkuiri mampu meningkatkan hasil belajar siswa.

Abstract

UJJ BANDING KIT RIA T3 PRODUK PRR-BATAN SISTEM "COATED TUBE" DENGAN PRODUK IZOTOP-HUNGARlA. Radioimunoassay (RIA) merupakan teknik analisis yang didasarkan pada prinsip immunologi dan menggunakan perunut radioaktif sehingga cuplikan dalam jumlah kecil mudah dideteksi serta spesifik karena didasarkan pada reaksi imunologi yaitu ketika terjadi ikatan antigen antibodi yang spesilik hanya untuk antigen tertentu saja. Teknik ini dapat digunakan untuk penemuan kadar T3 dalam serum yang mempunyai matriks yang komplek dan kadar yang sangat bervariasi. Triiodotironin (T3) adalah salah satu hormon yang diekskresikan oleh kelenjar tiroid. Telah dilakukan penelitian tentang uji banding Kit RIA T3 produk PRR-BATAN sistem "coated tube? dengan produk lzotop-Hungaria sebagai Gold Standard, tujuannya untuk melihat kesesuaian pengukuran kadar T3 dengan cara membandingkan hasil analisis menggunakan kit RIA T3 produk PRR-BATAN sistem ?coated tube? dengan produk lzotop Hungaria terhadap 273 sampel yang berasal dari poliklinik PTKMR-BATAN Pasar Jum?at. Hasil uji assay dari kedua kit tersebut diperoleh 185 sampel negatip hipotiroid (true negative), 30 sampel positip hipotiroid (true positive), 17 sampel false positip hipotiroid dan 41 sampcl false negatip hipotiroid. Sedang untuk sampel hipetiroid diperoleh hasil 207 sampel negatip hipertiroid (true negative), 41 sampel positif hipertiroid (true positive), 20 sampel false positip hipertiroid, dan 5 sampel false negatip hipertiroid. Hasil uji banding kit RIA T3 didapatkan nilai diagnostic sensitivity sebesar 42.25% dan diagnostic specivicity sebesar 91.58 % untuk pengukuran sampel hipotiroid, didapatkan diagnostic sensitivity sebesar 89,13% dan diagnostic specivicity sebesar 91,18 % untuk pengukuran sampel hipertiroid sehingga kualitas Kit RIA T3 produk PRR-BATAN sistem "coated tube" belum sama dengan produk dari Izotop Hungaria.Kaw Kunci: Kit, T3, RIA, uji banding
