Abstract

One of the treatments for diabetes mellitus disease is to control blood sugar level using an inhibitor of ?-glucosidase enzyme. The methanol extracts of the fruit, stem, and leaves of Rhodomyrtus tomentosa were found significant in inhibiting ?-glucosidase with an IC50 value of 20.57, 20.36 and 43.99 ?g/mL respectively. The ethyl acetate and n-butanol fractions from the methanol extract of R. tomentosa fruit exhibited the potent inhibition (IC50 13.49 and 19.29 ?g/mL) compare to acarbose and n-hexane fraction (IC50 383.68 and 1175.16 ?g/mL). A leptospermone derivative, rhodomyrtosone D, was isolated from the ethyl acetate fraction of R. tomentosa fruit. The structure of rhodomyrtosone D was identified based on spectroscopic analysis, as well as comparing with literature data. The ?-glucosidase inhibition of rhodomyrtosone D (IC50 110.45 ?g/mL) was 3.5 fold more potent than acarbose. Thus, R. tomentosa plant could be potential as a natural resource of ?-glucosidase inhibitor.

Abstract

Telah diekstrak senyawa Kuersetin-3-o-glikosida dari daun ubi karet (manihot glaziovii.m.a) dengan cara sokletasi menggunakan pelarut etanol teknis. Senyawa yang diperoleh berupa kristal kuning yang titik lelehnya 184,5° C -  186,5° C. Analisa terhadap kristal tersebut dengan alat Spektofotometri Ultra Violet dilakukan secara kualitatif dengan menggunakan pereaksi geser NaOCc 2,5%, NaOAc/H3BO3, AlCl3 5%/HCl 50%. Hasil penelitian menunjukkan gugus hidroksil (OH) yang terletak pada atom karbon C5, C7, C3, C4 dan glikosida pada atom karbon C3. Analisa data Spektroskopi Infra Merah menunjukkan adanya gugus-OH kuat pada 3417,0 cm-1, gugus –C=C- kuat pada 1651,5 cm-1, gugus –C=O kuat pada 1651,5 cm-1, gugus-C-H sedang pada 1293,3 cm-1 dan gugus –C-O sedang pada 1203,6 cm-1. Berdasarkan hasil analisa, kristal yang diperoleh adalah senyawa kuersetin-3-o-glikosida atau rutin (0,105%) berat kering.

Abstract

ABSTRAKKegiatan pengabdian masyarakat ini adalah membina Industri rumah tangga (IRT) IIn cake untuk memproduksi kue ringan yang cocok untuk penderita diabetes atau dapat mencegah diabetes. IIn cake adalah salah satu IRT yang mensuplai kue ringan (snack) di universitas Sriwijaya. Penyakit diabetes pada tahun terakhir meningkat dengan jumlah yang banyak. Penyakit ini merupakan penyakit yang memiliki komplikasi yang membahayakan. Baik diabetes tipe I maupun diabetes tipe II dapat menyerang pada semua umur. Untuk itu kami membina salah satu penyuplai kue ringan di UNSRI untuk memproduksi kue yang rendah gula dan rendah kalori untuk penderita diabetes dan konsumen yang menghindari diabetes. Kue yang dikembangkan diberi nama snack bar dibets dan snack chip dibets. Kedua jenis kue ini tidak memakai gula pasir, tetapi hanya brown sugar yang memilki indeks glikemik rendah. Tiap 100 gram snack bar dibets memiliki nilai gula 43,13±1,32; nilai  lemak 0,68± 1,32; nilai karbohidrat 45,25±1,27; serat 1,74±0,28. Kue khusus untuk penderita diabetes snack chip memiliki nilai gula 62,56±1,54; nilai lemak 0,58±0,65; nilai karbohidrat 60,42±1,53; nilai serat  1,73±0,18. Tim pengabdian juga membuatkan web site untuk penjualan dan sosialisasi kue snack bar dibets dan snack chips dibets.

Abstract

Telah dilakukan perbaikan visualisasi plasmid bermarka melanin (pIJ 702) pada Streptomyces lividans Rekombinan dengan modifikasi metode lisis alkali. Larutan netrium asetat yang digunakan untuk mengendapkan DNA dalam metode lisis alkali didapat pada pH 2 memberikan hasil yang terbaik dalam menvisualisasikan pita plasmid pIJ 702. Dengan perendaman etidium bromida (50 mg dalam 50 mL air) lebih dari waktu normal juga tidak memperlihatkan melanin pengganggu, sedangkan larutan buffer penyimpan yang di dapat pada pH di bawah 7 tidak memberikan pengaruh yang berarti (p = 0,05). Hasil visualisasi fragmen Bam H1 plasmid pIJ 702 juga jelas pertanda metoda bebas dari pengaruh melanin.

Abstract

Telah dilakukan uji daya hambat asap cair pirolisis tempurung kelapa terhadap pertumbuhan bakteri Bacillus cereus var mycoides. Bakteri Bacillus cereus var mycoides merupakan hasil isolasi bakteri kayu merawan (Hopea sp) yang ditanam dengan metode cawan tuang dan diuji pertumbuhannya dengan cara in vitro dengan menggunakan metode difusi kertas cakram. Hasil menunjukkan bahwa konsentrasi asap cair, yaitu 25%, 50%, 75% dan 100% v/v maka zona hambatannya yaitu sebesar 1,825; 2,425; 2,725 dan 3,10 cm. Senyawa antibakteri pada asap cair adalah senyawa fenol, 2-metoksi fenol, 2-metoksi-4-metil fenol, 4-etil-2-metoksi fenol dan 2,6-dimetoksi fenol.

Abstract

It has been isolated identified anti bacterium of antibiotic compunds produced by streptomyces sp. The isolation of sterptomyces sp was done from three kinds of soil on Soak area Km . 6, Palembang Municipallity using Plating Technic Spread Method produced 4 streptomyces isolate,. The test of the blocked capacity againts the whole isolates (4 isolates) with the bactery terstor showed that TL.J2 isolate from corn plant land has biggest activities. The isolates of antibiotics produced using fermentation method for TL.J2 isolates by using solvent extraction method and continued with HPLC produced 4 pure compunds. The test of the blocked capacity againts the 4 pure compunds, just STP1 which still antibacteria activities. Furthermore, the identification of STP1 using IR Spectrophotometer and UV showed that the results can be clasified into Alifaitic antibiotic consisting of unsaturated ketod alifatic function with the primer amine and hydroxy as a substiutent.

Abstract

Aims of this study to concentration test of methane gas from swamp forage silage with in vitro methods.This study used Completely Randomized Design with 3 treatments and each treatment consisted of 5 replications. The treatments used are as follows: P1 (100% kumpai tembaga grass (Hymenachne acutigluma)), P2 (50% kumpai tembaga grass (Hymenachne acutigluma) + 50% kemon air (Neptunia oleracea lour)), P3 (100% kemon air ( Neptunia oleracea lour)). The parameters observed N-Ammonia, total gas production, methane gas concentrations in vitro and VFA partially. The results of variance showed that treatment significantly different (P <0.05) with methane concentration  in vitro with highest gas concentration treatment of silage made from 50%  kumpai tembaga grass (Hymenachne acutigluma) and  50% kemon air (Neptunia oleracea lour).Keywords: Concentration, Methane, Silage Forage Swamp, In Vitro.

Abstract

An interesting natural product chemistry aspect of Fragraea fragrans is that their fruits are richness with ursolic acid and its isomer oleanolic acid (3.05% of dried powder). As our continuous work on these inseparable structural isomeric triterpenes, this paper reports that 51.0% of inseparable 3-oxo-ursolic[3-oxo-oleanolic] acids and 48.6% of inseparable 3-acethyl-ursolic [3-acethyl-oleanolic] acids have already been made from those triterpenes as starting materials of the oxidized and acetylated compounds and evaluated their activity against P388 leukemia cells. The activity of 3-oxo-ursolic [3-oxo-oleanolic] acids with IC50 = 18.6 µg/mL exhibited three-fold more potent against P388 leukemia cell proliferations compared to ursolic [oleanolic] acids with IC50 = 53.5 µg/mL; while the 3-acethyl-ursolic [3-acethyl-oleanolic] acids with IC50 = 37.9 µg/mL showed two-fold more potent then their parent triterpenes (IC50 = 53.5 µg/mL) in the inhibition of P388 leukemia cell growth.

Abstract

Mindi’s leaves (Melia azedarach Linn.) is an Indonesian medicinal plant that used as traditional medicine. Mindi leaves contains some secondary metabolites which have potency to decreased total cholesterol and LDL level. The study purposed to know antihyperlipidemic effect of ethanol extract of mindi’s leaves (Melia azedarach Linn.) seen from total cholesterol, and LDL in male albino rats. Male wistar albino rats were divided into 5 groups, negative control group (sodium CMC 0.5%), positive control group (simvastatin 0.193 mg/200 gBW), group I (ethanol extract mindi’s leaves with dosage 300 mg/200 gBW), group II (ethanol extract mindi’s leaves with dosage 600 mg/200 gBW), and group III (ethanol extract mindi’s leaves with dosage 1200 mg/200 gBW). The rats were given high-fat supplement and propylthiouracil for 15 days to increase cholesterol, and the extract was given for the next 15 days. Average cholesterol level, LDL, and body weight after induction was 90.28 mg/dL, 31.09 mg/dL, and 222.32 g. The result showed ethanol extract mindi’s leaves could decreased total cholesterol level and LDL level with % decreased in total cholesterol (%PDTC) and % decreased in LDL (%PDLDL) of group I is 37.78% and 35.57%, group II is 45.99% and 40.39%, and for group III is 56.29% and 52.42%. The result showed that ethanol extract of mindi’s leaves has antihyperlipidemic activity and significantly different from negative control (p&gt;0.05). Based on the percentage relation of decreased total cholesterol and LDL levels to dose, then the effective dose 50 (ED50) value of ethanol extract mindi’s leave is 869 mg/200 gBW for total cholesterol and for LDL reduction level is 1086.84 mg/200 gBW

Abstract

Research on physical interaction of chitosan-alginate entrapping extract of papaya leaf (Carica papaya L) into submicron particles formation has been performed. Preparation of papaya leaf extract into submicron particle dosage form of chitosan and sodium alginate polymer using ionic gelation method aimed to increase the solubility of extract. Submicron particles consisting of papaya leaf extract, chitosan, sodium alginate and CaCl2 were combined using variation of stirrer speed of 500, 750, and 1000 RPM. The optimum formula obtained has a speed of 1000 RPM with the percent EE value of 71.90%. The results of submicron particles characterization such as diameter and particle size distribution (PDI) using particle size analyzer (PSA) tools were 189.2 nm and 0.330. The results of XRD revealed the changes of type of crystalinity form to amorphous on submicron particles. The results of FTIR revealed the physical interaction without shifting of wave number of carbonyl, amine, and hydroxyl group which indicated that there were no chemical interactions occurred. These data indicated that papaya extract can be formulated into submicron particles of chitosan-alginate polymer.
