Abstract

Cancer is one of the leading cause of death in the world. Complicated and high cost treatments of cancer encourages people to look for alternative treatments among others using medicinal plants. Some medicinal plants that are empirically claimed to have anticancer effect are soursop (Annona muricata), takokak (Solanum torvum), and bidara upas (Merremia mammosa). However, these medicinal herbs have not been tested for cytotoxic activity against MCF-7 cancer cells. The purpose of this study was to determine the cytotoxic activity of the anticancer herbal formula consisting those three plants. The effect of each single extract and the combination formula were tested for their activity on cell viability of MCF-7 with MTT assay method. The herbal formula with the most active combination of medicinal plant extracts was further tested for its activity on the expression of Bcl-2 protein by immunocytochemistry methods. The results showed that each single extract and the combination formula gave cytotoxic effect on MCF-7 breast cancer cells. Formula A, consisted of a mixture of soursop leaves, takokak fruit, and bidas upas bulb with a ratio of 1:1:1, results in highest inhibition of the viability of MCF-7 cells with IC50 values of 48 µg/ml. The formula also enhances the apoptosis process in MCF-7 cells which is shown by decreasing the expression of antiapoptotic Bcl-2 proteins.

Abstract

Cancer is one of critical, chronic, and complex disease, also becoming the high cause of death in the world. Brazilin and brazilein in sappan wood (Caesalpinia sappan L.) and zerumbone in bitter ginger rhizome (Zingiber zerumbet L.) are known having cytotoxic activity with different mechanisms. This study aimed to examine combination effect of sappan wood and bitter ginger rhizome. Sappan wood and bitter ginger rhizomes macerated with ethanol 96% for 3x24 hours, filtered, and evaporated to obtain dried extract.Cytotoxic effect on MCF-7 cells was done using MTT assay. Combination Index (CI) was determined by CompuSyn based on the result of cytotoxic combination. Cell cycle profile and apoptosis induction was analyzed by flow cytometry. Sappan wood extracts and bitter ginger rhizome exhibited cytotoxic effects on MCF-7 cells with the IC50 values of 30 and 155 ?g/mL respectively. The combination of sappan wood 15 ?g/mL and bitter ginger 8, 12, 24, and 60 ?g/mL produced synergistic effect with the CI value of 0.57-0.85. Sappan wood 15 ?g/mL combined with bitter ginger 8 and 24 ?g/mL showed cell cycle inhibition at G2/M phase. The combination also increased apoptosis induction compared to untreated cells and its single treatment.The combination of sappan wood ethanolic extracts and bitter ginger rhizome showed synergistic cytotoxic effect. Its synergism effect revealed through cell cycle arrested at G2/M phase and acceleration of apoptotic induction.

Abstract

Latar Belakang : Laparatomi merupakan  pembedahan perut, membuka selaput perut dengan operasi yang dilakukan untuk memeriksa organ-organ perut dan membantu diagnosis masalah termasuk menyembuhkan penyakit-penyakit pada perut (Mansjoer, 2007). Pasca perawatan luka post op laparatomy hari ke 2 pasien merasakan nyeri hebat dan 75% penderita mempunyai pengalaman yang kurang menyenangkan akibat pengelolaan nyeri yang tidak adekuat. Hal tersebut merupakan stressor bagi pasien dan akan menambah kecemasan serta keteganggan yang berarti pula menambah rasa nyeri karena rasa nyeri menjadi pusat perhatiannya. Bila pasien mengeluh nyeri maka menjadi pengalaman yang kurang menyenangkan akibat pengelolaan nyeri yang tidak adekuat. Tingkat dan keparahan nyeri pasca perawatan luka tergantung pada fisiologis dan psikologis individu dan toleransi yang ditimbulkan nyeri. Mendengarkan musik dapat memproduksi zat endorphins (substansi sejenis morfin yang disuplai tubuh yang dapat mengurangi rasasakit/ nyeri) yang dapat menghambat transmisi impuls nyeri di sistem sarafpusat, sehingga sensasi nyeri menstruasi dapat berkurang. Tujuan : Mengetahui pengaruh terapi musik untuk mengurangi intensitas nyeri saat perawatan luka Post Op Laparatomy hari ke 2 di RSUD Dr. Moewardi. Metode Penelitian : Metode penelitian adalah metode quasi eksperimental design. Rancangan Penelitian ini menggunakan pendekatan one design prestest posttest only. Dalam penelitian ini teknik pengambilan  sampel yang digunakan adalah teknik accidental sampling yaitu Responden yang kebetulan ada atau tersedia di suatu tempat sesuai dengan konteks penelitian, jumlah responden 25 orang. Instrumen penelitian menggunakan kuesioner NRS. Menggunakan analisa bivariat dengan uji Willcoxon Hasil : Setelah dilakukan terapi music dari 25 responden 19 (76%) mengalami nyeri ringan, 6 (24%) mengalami nyeri sedang. Ada pengaruh dan perbedaan berbanding terbalik setelah dilakukan terapi musik, dengan nilai p <0,05 dan p = 0,000 pada signifikasi 95%. Kesimpulan : Ada pengaruh terapi musik terhadap intensitas nyeri pada perawatan luka pasien post operasi laparatomy hari ke dua di RS Dr. Moewardi

Abstract

Ekinase (Echinacea purpurea (L.) Moench) a medicinal plant that has immunostimulatory activity. This plant has been cultivating in Tawangmangu region by Medicinal Plant and Traditional Medicine Research and Development Office since 2002. Ten accessions of E. purpurea were found based on their morphological variation, three accessions of them are selected as promising accessions namely BH2, BHU3 dan BHU5. The objective of this research was to observe the genetic diversity of  those  promising accessions and 8 variants accession from mass selection year I using RAPD analysis. Those accessions were amplified using 10 RAPD primers. A total of 64 scorable fragments were generated from 9 RAPD primers, among which 48 fragments (75%) were polymorphic. The Dice coefficient was used to calculated the genetic similarity and UPGMA was used to generate the dendogram. The genetic similarity index among accessions evaluated ranged from 75,49-84,21% thus indicating that low level of genetic diversity. RAPD analysis proved to be efficient for genetic diversity of ekinase accessions from mass selection year I.

Abstract

Dringo (Acorus calamus L.) used as medicinal plant in Indonesian ethnic groups. Those information based on Ristoja 2012 research. The objective of Ristoja was to provide a database of local ethnomedicine knowledge, herbal formula and medicinal plant in Indonesia. Inter Simple Sequence Repeats (ISSR) markers were used to assess the genetic diversity of Dringo from 20 selected ethnic groups in Indonesia. Ten selected ISSR primers generated 82 amplified fragments with 51,2% were polymorphic. Dice coefficient was used to calculate similarity index and UPGMA was used to construct a dendogram. The genetic similarity index among accessions ranged from 76,7-100% thus indicating that low level of genetic diversity in dringo. Genetic diversity database can be useful for medicinal plant mapping and conservation especially for in situ conservation.

Abstract

Gerrardanthus macrorhizus (GM) caudex, is traditionally used in cancer therapy by the Tetun people in Belu District, East Nusa Tenggara Province, Indonesia, where it is known as “akar batu”. This study aimed to explore the cytotoxic effects of G. macrorhizus caudex aqueous extract, as well as its combination with doxorubicin, on MCF-7 cells. Also investigated were the possible mechanisms of interaction through cell cycle progression and apoptosis induction. Single treatments of 5–320 µg/mL of the extract showed morphological alterations in MCF-7 cells, but did not show any cytotoxic effect. Combining the extract with doxorubicin resulted in a synergistic cytotoxic effect. Doxorubicin concentrations equivalent to 1/12, 1/8, and 1/5 fold of the IC50 combined with 20 µg/mL decreased viability to 48%. We then explored the combination effect of doxorubicin 0.4 µM with GM 5 and 20 µg/mL using a flow cytometer. A low concentration of the extract (5 µg/mL) combined with 0.4 µM of doxorubicin resulted in slight cell cycle modulation by G1, G2M arrested and apoptosis induction. The combination of doxorubicin and a higher concentration of the extract (20 µg/mL) did not show cell cycle modulation, and led to necrosis. Therefore, G. macrorhizus caudex at low concentrations has the potential to be developed further as a co-chemotherapeutic agent.

Abstract

Antenatal Care adalah uapaya preventif program pelayanan kesehatan obstetric untuk optimalisasi luaran maternal dan neonatal. Menurut WHO pada tahun 2012 sebanyak 536/100.000 persalinan hidup, di Indonesia pada tahun 2012 sebesar 228/100.000 kelahiranhidup,sedangkan di Provinsi Lampung padatahun 2012sebanyak 105/100.000 kelahiran, di kabupaten Lampung Tengah pada tahun 2012 mencapai 17 kasus. Berdasarkan pra survey di BPS Uswatun Khasanah pada tanggal 13 Februari 2014 data tentang kunjungan K4 tahun 2011 sebesar 77,8%menurun ditahun 2012 menjadi 63,4%. Maka, Tujuan dari penelitian ini adalah diketahuinya hubungan dukungan suami pada ibu hamil dengan kunjungan Antenatal Care di BPS Uswatun Khasanah Liman Benawi Kecamatan Trimurjo Lampung Tengah Tahun 2014.Jenis penelitian ini menggunakan jenis penelitian kualitatif dengan rancangan analitik dan menggunakan pendekatan croos sectional. Populasi dalam penelitian ini adalah seluruh ibu hamil  yang melakukan kunjungan antenatal care (ANC) pada saat penelitian sebanyak 48 responden dengan sampel accidental sampling. Teknik pengumpulan data menggunakan lembaran kuesioner. Analisa data univariat dengan distribusi frekuensi dan analisa bivariat dengan uji chi  square.Hasil penelitian dengan analisa univariat terdapat 28 responden (58,3%) yang kurang mendapatkan dukungan dari suami dan terdapat 26 (54,2%) responden yang kurang teratur melakukan kunjungan antenatal care (ANC), hasil analisa bivariat, Adanya hubungan yang signifikan dukungan suami dengan kunjungan antenatal care (ANC) pada ibu hamil dimana p-value = 0,000. Dari hasil penelitian ini menunjukkan bahwa dukungan suami mempunyai pengaruh positif terhadap kunjungan Antenatal Care ibu yang berarti semakin ibu mendapatkan dukungan suami akan mempengaruhi meningkatnya keteraturan kunjungan antenatal care (ANC). Maka, saran dari responden bagi suami untuk ikut aktif dalam melakukan kunjungan antenatal agar informasi dapat diterima oleh suami istri dan target kunjungan antenatal care (ANC) minimal 4x selama kehamilan dapat tercapai. Kata Kunci :Dukungan Suami, Kunjungan Antenatal  Care (ANC)

Abstract

Avocado Persea Americana Mill. is a commercially important crop and studies have shown that the pulp may have benefits to cardiovascular health, dermatological health and possibly anti-cancer activity. Avocado seeds have several medicinalproperties such as anti-hyperglycemic, antimicrobial, antioxidant and anti-inflammation. This study aim to evaluate the effect of avocado seed extract on viability and apoptosis of breast cancer cell line MCF-7. The anticancer effect was evaluated by cytotoxic test using MTT assay and the effect on apoptosis and cell cycle was examined by flow cytometry method. The cytotoxic test showed that chloroform extract had strong cytotoxic activity against MCF-7 cell lines with IC50 value of 94.87 μg mL-1. Furthermore, the chloroform extract was partitioned with methanol and yield of soluble methanol fraction (FLM) and non soluble methanol fraction (FTLM). The cytotoxic activity of the methanol soluble fraction (FLM) and non soluble methanol fraction (FTLM) against MCF-7 cell lines was increased with IC50 of 34.52 and 66.03 μg mL-1, respectively. Flow cytometry analysis using annexin-V and propidium iodide staining revealed that methanol soluble fraction could induce apoptosis and modulating the cell cycle arrest in MCF-7 cell. This research indicated that avocado seed has a potency to induce apoptosis and as anti-proliferative to MCF-7 cells lines.

Abstract

Research on the growth and oleanolic acid content of pearl grass (Hedyotis corymbosa) in experimentall field of Medicinal Plants and Traditional Medicines Research and Development Center (B2P2TOOT), Tanjungsari Village, Tegal Gede Village, Karanganyar and B2P2TOOT laboratory, Tawangmangu, Karanganyar. The research has been conducted in August to November 2017. The purpose of this research is to know the dosage of cow manure and liquid organic manure which has the most influence to the growth and the oleanolic acid content of pearl grass. The experiment was designed in a randomized complete block design with twelve treatments and three repication. The observed data  analyzed using  variance analysis  with 5% level then if there was a significant difference in the continued test with Duncan’s Multiple Range Test (DMRT). The results showed that the use of cow manure at doses of 0 kg-1m2, 4,6 kg-1m2, 6,1 kg-1m2 and 7,6 kg-1m2 did not give increase to all observation variables. The use of POC dose 0 ml-1liter, 5 ml-1liter and 10 ml-1liter did not give increase to all observation variables. All given treatments may show that oleanolic acid is present in pearl grasses.
