Abstract

Skin aging processes are divided into chronological aging and premature aging. Premature aging is generally caused by free radicals, from both air pollution and photoaging. Natural compounds from plant extracts are among sources of antioxidants and anti-hyaluronidase which have the ability to prevent antiaging. One of the potential fruits related to antioxidant and antiaging activities is Anana scomosus. A. comosus has a number of phenolic compounds with biological activities. One of the main phenolic compounds in A. comosus is luteolin. The aim of this study was to evaluate the antioxidant and antiaging potentials of Pineapple Core Extract (PCE). This study was conducted at the Biomolecular and Biomedicine Research Center, Aretha Medika Utama from August to November 2018. Analysis of antioxidants from PCE and luteolin was carried out using H2O2 scavenging activity assay. The antiaging assay was carried out through inhibition of hyaluronidase enzyme, one of the important enzymes in the aging process. Luteolin had lower IC50 value of H2O2 scavenging activity of around 24.12±3.13 ?g/ml, which was better than CPE with IC50 of 304.56±3.76?g/mL. The results of hyaluronidase inhibition activity assay showed that luteolin compound had a lower IC50 value of 67.38±3.99 ?g/mL when compared to PCE with an IC50 value of 161.15±1.05 ?g/mL. Hence, Luteolin has higher antioxidant and anti-hyaluronidase activities than PCEPerbandingan Aktivitas Antioksidan dan Antihialuronidase Ekstrak Bonggol Buah Nanas (EBN) dengan Senyawa LuteolinTerdapat dua jenis proses penuaan kulit, penuaan kronologis dan penuaan dini. Penuaan dini umumnya disebabkan oleh radikal bebas, baik dari polusi udara maupun photoaging. Penggunaan antioksidan dan antihialuronidase untuk mencegah proses penuaan telah banyak diketahui. Ananascomosus merupakan salah satu tanaman yang memiliki potensi sebagai antipenuaan dan antioksidan karena mengandung sejumlah senyawa fenolik yang memiliki berbagai aktivitas biologis. Salah satu senyawa fenolik utama dalam A. comosus adalah luteolin. Tujuan penelitian ini adalah untuk mengevaluasi potensi antioksidan dan anti-aging dari Ekstrak Bonggol Nanas (EBN) dan senyawa pembandingnya yaitu luteolin . Penelitian ini dilakukan di Biomolecular and Biomedicine Research Center, Aretha Medika Utama dari Agustus hingga November 2018. Pengujian antioksidan dari EBN dan luteolin  dilakukan menggunakan uji pemerangkapan H2O2. Uji antipenuaan dilakukan melalui uji penghambatan aktivitas hialuronidase, salah satu enzim penting dalam proses penuaan. Hasil penelitian menunjukkan bahwa Luteolin  memiliki nilai IC50 aktivitas pemerangkapan H2O2lebih rendah sekitar 24.12±3.13 ?g/ml lebih baik daripada EBN dengan IC50 sebesar 304.56±3.76?g/mL. Hasil uji aktivitas penghambatan hialuronidase, senyawa luteolin  memiliki nilai IC50 lebih rendah, yaitu sebesar 67,38±3,99 ?g/mL dibanding dengan EBN nilai IC50 161,15±1,05?g/mL. Hasil ini menunjukkan bahwa senyawa pembanding luteolin  memiliki aktivitas antioksidan dan antihialuronidase yang lebih tinggi daripada EBN.

Abstract

Aging involves progressive physiological changes of the body, including skin dysfunction and death which can be accelerated by the presence of free radicals. Compounds with potential antioxidants and antiaging are contained in various fruits, including dragon fruit (Hyloceureus polyrhizus (F.A.C. Weber) Britton & Rose). This study aimed to compare the antioxidant and antiaging activities of dragon fruit rind extract (DFRE) and Kaempferol. The antioxidant and antiaging activities were measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenger and inhibitory activity assay of tyrosinase, respectively and was conducted at the Biomolecular and Biomedicine Research Center, Aretha Medika Utama, from August to November 2018. Data acquired were then analyzed using ANOVA, followed by Post Hoc Test using Turkey HSD test with a confidence level of 95% (?=0.05). Phytochemical assay was also conducted to determine the content of active ingredients in thedragon fruit rind extract. Phytochemical test result showed that DFRE contained flavonoids, tannins, phenols, triterpenoids, and alkaloids compounds. Both DPPH scavenging and tyrosine inhibition assay indicated that Kaempferol compound also had antioxidant and antiaging activities. The IC50 values of DFRE and Kaempferol antioxidant activities were 164.98 ?g/mL and 83.30 ?g/mL, respectively, while the IC50 values of tyrosinase inhibition activity of DFRE and Kaemferol were 88.46 ?g/mL and 59.34 ?g/mL, respectively. DFRE had antioxidant activities and tyrosinase inhibition activities thus can be used as an alternative antiaging and antioxidant agents. Perbandingan Aktivitas Antioksidan dengan Antitirosinase Ekstrak Etanol Kulit Buah Naga (Hyloceureus polyrhizus (F.A.C. Weber) Britton & Rose)Aging atau penuaan adalah proses kompleks yang ditandai dengan penurunan progresif fungsi fisiologis tubuh, termasuk kulit yang diikuti oleh disfungsi dan kematian kulit. Faktor-faktor yang dapat mempercepat proses penuaan dini di antaranya radikal bebas dan gaya hidup yang tidak sehat. Buah-buahan banyak mengandung senyawa yang berpotensi sebagai antioksidan dan antiaging, salah satu di antaranya adalah buah naga. Penelitian ini bertujuan mengetahui potensi antioksidan dan antiaging ekstrak kulit buah naga (EKBN) yang dibandingkan dengan senyawa kaempferol (SK) sebagai pembanding. Penelitian yang dilakukan meliputi uji fitokimia untuk mengetahui kandungan bahan aktif pada kulit buah naga, uji pemerangkapan DPPH, dan uji penghambatan tirosinase dan dilakukan di Pusat Penelitian Biomolekul dan Biomedis, Aretha Medika Utama, dari Agustus hingga November 2018. Data lalu dianalisis menggunakan analisis varians (ANOVA) satu arah (one way) Hasil penelitian uji fitokimia menunjukkan EKBN mengandung golongan senyawa, flavonoid, tanin, fenol, triterpenoid, dan alkaloid serta tidak mengandung golongan senyawa saponin, steroid, dan terpenoid. Senyawa kaempferol memiliki aktivitas antioksidan melalui pemerangkapan DPPH dan aktivitas antiaging melalui penghambatan tirosinase yang lebih baik dibanding dengan EKBN. Aktivitas antioksidan dilihat dari nilai IC50 EKBN sebesar 164.98 ?g/ml dan SK sebesar 83.30 ?g/mL. Aktivitas penghambatan tirosinase memiliki nilai IC50 EKBN masing-masing sebesar 88.46 ?g/mL dan IC50 SK sebesar 59.34 ?g/mL. EKBN memiliki aktivitas antioksidan dan penghambatan tirosinase sehigga dapat dimanfaatkan sebagai alternatif bahan antiaging dan antioksidan.

Abstract

Tapak dara or Madagascar Periwinkle (Catharanthus roseus [L] G.Don), a natural plant, is empirically reported to have promising anticancer activity. To elucidate its mechanism, a research was conducted to investigate the possible ethanol extract of  C.  roseus in inducing apoptosis on breast cancer cell line (T47D). Antioxidant activity of C. roseus was investigated as well. Sub-G1 flowcytometric apoptotic analysis result showed that extract of C. roseus at 6.25 μg/mL induced apoptosis for 26.365%. Increasing extract concentration resulted an increasing apoptotic level as well, extract at concentration of 12.5 μg/mL induced apoptosis for 22.235%.  Meanwhile doxorubicin at concentration of 10  μg/mL induced apoptosis for 36.055%. The antioxidant activity was determined by using  in vitro assay: inhibition of  2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging  activity. Antioxidant activity of  C. roseus extract were compared to quercetin and butylated hydroxyanisol (BHA), as positive controls.  The results showed that DPPH IC50 of C. roseus extract, quercetin and BHA were 358.411 μg/mL, 19.200 μg/mL  and  94.178  μg/mL, respectively. We suggest that  C. roseus extract had a potential anticancer activity by inducing apoptosis.

Abstract

Various diseases including cancer and cardiovascular diseasesare induced by free radicals via lipid peroxidation, protein peroxidation, DNA damage, and cellular degeneration in thecells.Piper betle L.,commonly called betel,belongs to thePiperaceae family.It originated from South and South Eas tAsia,and has been used as a traditional medicine by people theredue to its medicinal properties.In the present study, free radicalscavenging activity of ethanolic leaves extract of Piper betle L.together with its different solvent fractions(hexane,ethylacetate,butanol,and water)were evaluated by DPPH freeradical scavenging assay.The ethanolic extract and the ethylacetate fraction had shown very significant DPPH (1,1)diphenyl)2)picryl)hydrazyl) radical scavenging activitycompared to the other fractions.In DPPH free radical scavengingassay,the IC50 value of the ethanolic extract and the ethyl acetate fraction were found to be 17.431g/mL and 11.53 1g/mL, respectively. The free radical scavenging activity was assumed due to the total phenolic content.The total phenoliccontent was found to be highest in the ethanol extract(277.68µgEGCG/mg)and the ethyl acetate fraction(559.38µgEGCG/mg).The results concluded that the ethanolic leaves extract of Piper  betle L. and its ethyl acetate fraction have a consider ably freeradical scavenging activity.

Abstract

Stem cells are cells that became the beginning of the growth  to others cell that constract the whole body of organisms, including human. Stem cells are from embryonic, fetal, or adult cells, which are capable to proliferate to cells in a long time, which this cell doesn’t have specific function, stem cells are able to differentiate  into specific cell that build tissues and organ systems. Stem cells have charateristic including :  undifferentiated cells, this cell is self-renewal, cells can differentiate into more than one type of cells (multipotent/pluripotent). Types of stem cells include embryonic stem cells derived from embryo, or inner cell mass (ICM) and adult stem cells found in bone marrow, cord blood, umbilical cord, peripheral blood, liver, skin, brain, dental pulp, fatty tissue. Abstract

Aging is a natural process in human that can be characterized by the appearance of black spot on the skin due to hyperpigmentation. Aging may occur due to an excessive amount of free radicals in the body. Antioxidants possess ability to capture free radicals and inhibit tyrosinase which induces skin aging. Aloe vera has been used in traditional medicine because it contains several bioactive compounds that act as antioxidant and prevent aging process. This study aims to determine phytochemical content, antioxidant activity and tyrosinase inhibition activity of Aloe vera rind (AVRE) and gel (AVGE) extract. This research was carried out at Aretha Medika Utama-Biomolecular and Biomedical Research Center in Bandung city in September-November 2018. Phytochemical assay was determined using modified Farnsworth method. Antioxidant assay was determined using DPPH scavenging activity and antiaging assay was obtained using tyrosinase inhibition assay. AVRE contains flavonoid, phenol, steroid, and alkaloid. Meanwhile, AVGE contains steroid and alkaloid. IC50 DPPH scavenging activity of AVRE was 113.18 µg/mL followed by AVGE was 291.96 µg/mL. IC50 tyrosinase inhibition activity of AVRE was 65.04 µg/mL followed by AVGE was 111.89 µg/mL. AVRE had more active DPPH scavenging activity and tyrosinase inhibition activity than AVGE.

Abstract

Free radicals and UV exposure can cause aging. Aging prevention needs substances that can prevent molecular oxidation reactions in cells and inhibit the activity of enzymes that trigger aging. Research on pineapple skin and flesh extract (Ananas comosus (L.) Merr.) reported the presence of luteolin compound which functions as antioxidants and antityrosinase. However, in this study, the object used was pineapple core extract (PCE), which has not been widely known for its antioxidant and antityrosinase activity. Therefore, the purpose of this study was to determine the content of phytochemical compounds, antioxidant activity, and inhibitory activity of tyrosinase enzymes by PCE and then compared with the luteolin (LT) compound test results using Fansworth method, DPPH scavenging activity assay and tyrosinase enzyme inhibition assay. Based on the study results, the phytochemical compounds contained in PCE were tannins and triterpenoids.  PCE and LT produced the highest DPPH scavenging activity, which was 64.86% and 59.32% (final concentration 200?g/ml and 6.25?g/ml) and the highest tyrosinase inhibition activity, which was 60.52% and 85.02% (final concentration 100 ?g/ml). Antioxidant activity was determined based on IC50 of 87.46µg/mL and 4.17 µg/ml respectively. IC50 tyrosinase enzyme inhibition EBN and LT respectively at 62.27µg/ml and 5.25 µg/ml. Antioxidant activity through DPPH free radical scavenging test and tyrosinase enzyme inhibition activity by PCE was lower than LT.

Abstract

Objective : This study was conducted to determine the antioxidant activities of spice extractsincluding clove (Syzygum aromaticum L.), Indonesian cassia (Cinnamomum burmanni (C. Nees& T.Ness)), coriander (Coriandrum sativum L), nutmeg (Myristica fragrans Houtt), java cardamom(Amomum compactum Soland. Ex maton).Methods : This research was to evaluate antioxidant activities including 1,1-diphenyl-2-picrylhydrazyl(DPPH) scavenging activity, the value of superoxide dismutase (SOD), and total phenoliccontent.Result : The highest DPPH activity is clove and Indonesian cassia extracts with IC50 value 4.16μg/ml and 5.46 μg/ml respectively. The highest SOD value are Indonesian cassia extract (9.1432U/ml) at 500 μg/ml, 7.0045 U/ml at 125 μg/ml and 4.6751 U/ml at 31,25 μg/ml. Clove extract was thehighest of phenolic content (188.35 μg/mg eugenol equivalent).Conclusion : Indonesian cassia extract have high antioxidant activities both DPPH scavenging andSOD activities. Clove extract contain the highest eugenol compared with Indonesian cassia,coriander, nutmeg and java cardamom.

Abstract

Aging is a natural process in human life and is triggered by the presence of free radicals (ROS). The use of antioxidants from natural ingredients is one of the breakthroughs to overcome aging and counteract the harmful effects caused by the free radicals. This study aimed to determine and compare the antioxidant activity of H2O2 scavenging and hyaluronidase inhibition of red dragon fruit peel extract (DFPE) and kaempferol-3-o-rutinoside (KOR) compounds. Dragon fruit peel extract (DFPE) is obtained through extraction by maceration method using 70% ethanol solvent. The design of this study included antioxidant and anti-aging activity assay of EKBN and KOR at the series concentration of 15.63; 31.25; 62.50; 125; 250; 500 µg/mL through H2O2 scavenging, as well as the DFPE and KOR hyaluronidase inhibition assay at the series concentration of 5.21; 10.42; 20.83; 41.7; 83.33; 166.67 µg/mL. EKBN shows that the average activity of H2O2 scavenging is lower than KOR. In addition, the IC50 values of KOR for H2O2 scavenging is lower (351.46±2.30ug/mL) than DFPE (409.64±23.17ug/mL). While, KOR also has higher values of inhibitory activity than of the DFPE. However, the IC50 value of KOR for hyaluronidase inhibition activity was 84.07±10.46µg/mL, equivalent to the IC50 value of DFPE (85.32±10.24µg/mL). The presence of antioxidant and anti-aging activity in the EKBN is probably caused by betalain and the KOR compound itself contained in red dragon fruit. The results of the paired-samples T-test on antioxidant activity and anti-aging of DFPE and KOR showed non-significant difference. Thus, DFPE has an equivalent antioxidant and anti-aging through H2O2 scavenging and hyaluronidase activity as possessed by the KOR compound.

Abstract

Degenerative diseasesandchronicdiseasesare oftencausedbyoxidativestress. Oxidative stress caused by free radicals. Antixodant as inhibitor are needed to prevent it which is one of antioxidant sources is tea. Tea processinggenerally produce various kinds of teas such as black, green and oolong tea. Tea processing  affect thecontent ofphenolic compounds. The aim of the research is to evaluate phytochemical content, totalphenolic content of blacktea, green tea andoolongtea extracts using catechin, quercetin, kaempferol, myricetin as standard, and to evaluate the antioxidative potency of black tea, green tea and oolong tea extracts compared to catechin, quercetin, kaempferol, myricetin. Phytochemical assay using modified Farnsworth method, the antioxidant activity were measured by  by its 1,1-diphenyl-2-picryl-hydrazyl (DPPH) scavenging activity. Green tea extract contained highest phenolic and flavonoid.  The highest antioxidant activity was green tea extract with IC50=0,487μg/mL. Green tea extract content phenol and flavonoid are higher compared to the other extracts,  green tea extract has the highest antioxidant activity.
