Abstract

Telah dilakukan penelitian tentang efek ekstrak etanol rimpang temugiring terhadap kadar trigliserida serum darah tikus. Penelitian ini menggunakan 28 ekor tikus jantan galur Wistar dengan berat badan antara 150-200 gram yang berumur 2 -3 bulan.Tikus dibagi menjadi 7 kelompok perlakuan dan setiap kelompok terdiri atas 4 ekor tikus. Kelompok I (Kontrol normal) mendapat perlakuan diet standar (BR II) dan air minum, kelompok II (Kontrol negatif) yang diberi perlakuan diet standar, diet lemak tinggi, dan air minum, kelompok III (Kontrol pensuspensi) yang diberi perlakuan diet standar, diet lemak tinggi, larutan CMC Na dan air minum, kelompok IV (Kontrol positif) yang diberi diet standar, diet lemak tinggi, suspensi gemfibrosil dosis 0,9 mg./kgBB dan air minum, kelompok V, VI, VII, diberikan perlakuan peroral dengan ekstrak etanol temugiring dosis 10 mg /kgBB, 20 mg /kgBB, dan 40 mg/kgBB.. Peningkatan kadar trigliserida diberikan dengan diet lemak tinggi (5 gram lemak sapi dan 10 gram kuning telur) dalam sediaan emulsi pada periode ke-2. Perlakuan ekstrak etanol temugiring diberikan setelah keadaan hipertrigliserida..Penetapan kadar trigliserida dengan metode GPO (Glycerol-3-Phosphate Oxidase). Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol temugiring selama 2 minggu berturut-turut dapat menurunkan kadar trigiserida

Abstract

flavonoid yang digunakan masyarakat untuk pengobatan tradisional. Dari penelitian terdahulu dilaporkan bahwa senyawa flavonoid dapat berpotensi menurunkan kadar asam urat darah dengan cara menghambat aktivitas xanthine oxidase. Oleh karena adanya flavonoid yang terkandung dalam akar Sambiloto maka dilakukan penelitian apakah ekstrak etanol akar Sambiloto dapat menghambat aktivitas xanthine oxidase. Sebagai pembanding digunakan Allopurinol. Ekstrak etanol dibuat dari serbuk akar Sambiloto diekstraksi dengan etanol menggunakan metode penyarian dengan alat Soxhlet, sebelum penyarian dilakukan pengawalemakan menggunakan petroleum eter. Penghambatan aktivitas xanthine oxidase oleh ekstrak etanol akar Sambiloto ditentukan melalui penurunan produksi asam urat yang dimonitor dengan spektrofotometer pada 295 nm dengan xanthine sebagai substrat. Nilai kecepatan yang diperoleh selanjutnya digunakan untuk menghitung nilai aktivitas. Kemudian ditentukan konsentrasi ekstrak etanol yang mampu menghambat aktivitas xanthine oxidase sebesar 50% (IC50). Hasil dianalisis secara kuantitatif, dengan menggunakan uji Kruskal Wallis dengan taraf kepercayaan 95%, kemudian dilanjutkan dengan uji Mann Withney. Flavonoid dalam ekstrak etanol dipisah dengan cara kromatografi kertas dan perubahan bercak ditentukan dengan UV 366 dengan dan tanpa pemberian uap amoniak. Hasil penelitian menunjukkan bahwa ekstrak etanol akar Sambiloto menghambat aktivitas Xanthine Oxidase dengan IC50 16,82 μg/ml sedangkan IC50 Allopurinol adalah 4,29 μg/ml. Ekstrak etanol akar Sambiloto diduga mengandung flavonoid golongan flavon atau flavonol.

Abstract

Cholesterol was a steroidal compounds contained in the human body. Cholesterol imbalance in the body caused hypercholesterol disease. Acharan sulfate is a compound found in snail body and potentially lowering cholesterol level. Acharan sulfate was extracted from snail body with maceration method using ethanol solvents. Research was done by using animals model, male rats (Rattus norwegicus) Wistar strain which was two months old and weighing 180 to 200 grams. The animals model were divided into six groups. One groups used as normal control. Five groups were treated with high cholesterol diet with suspense of cow fat and yellow egg for ten days. After that, three groups were treated with the ethanolic extract of snail body with doses of 40 mg/kgbw, 80 mg/kgbw, and 160 mg/kgbw. The two other groups were treated with Simvastatin and BR II feed (disease control). Total cholesterol, HDL and LDL concentrations were determined using enzymatic photometric test CHOD-PAP method. Results showed that snail body the ethenolic extract of decreased total cholesterol concentration by 6,33 mg/dl at doses 160 mg/kgbw and 4,90 mg/dl at doses 80 mg/kgbw. HDL concentration increased by 0,85 and 8,06 mg/dl respectively at doses 80 mg/kgbw and 160 mg/kgbw. LDL concentration decreased by 1.96 mg/dl, 2.22 mg/dl and 3.52 mg/dl respectively at doses 40, 80, and 160 mg/kgbw.

Abstract

The inhibitory activity of heme polymerization of (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide have been done. This study aims to analyse the (1)-N-(2-nitrobenzyl)-1,10-phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide as inhibitory of polimerization heme. Analysis of heme inhibtory polimerization activity used the experimental in vitro method. The activity showed by IC50 (the capable concentration of extract to inhibiting polymerization heme by 50% ). The IC50 value acquired by probit analysis. Assess IC50 of (1)-N-(2-nitrobenzyl)- 1,10-phenantrolinium iodide not to be identified, (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide and chloroquine by successively are 0,571±0,071; 25,498±1,876 mg/mL. The result showed the (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide had the highest value of the heme polymerization inhibitory activity than chloroquin, (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide hadn’t the heme polymerization inhibitory activity.

Abstract

Kerusakan pada hepar dapat ditunjukkan oleh aktivitas enzim seluler yang semakin meningkat. SGOT dan SGPT adalah dua macam enzim yang paling sering dihubungkan dengan kerusakan sel hepar. Salah satu penyebab kerusakan hepar adalah senyawa radikal bebas yaitu CCl4. Ganggang hijau (Ulva lactuca L.) memiliki senyawa melatonin yang bersifat anti oksidan. Penelitian ini bertujuan mengetahui kemampuan ekstrak etanol ganggang hijau (Ulva lactuca L.) sebagai hepatoprotektor dengan menurunkan kadar SGOT dan SGPT akibat senyawa radikal bebas yaitu CCl4. Hewan uji dibagi menjadi 5 kelompok. Kelompok I (kontrol normal) hanya diberi makan dan minum, kelompok II  diberi suspensi CMC-Na 1% sebagai kontrol pelarut, kelompok III diberi suspensi ekstrak etanol ganggang hijau dengan dosis 100mg/kgBB/hari, kelompok IV diberi suspensi ekstrak etanol ganggang hijau dengan dosis 200mg/kgBB/hari, dan kelompok V larutan pembanding tablet curcuma sebagai kontrol pembanding dengan dosis 200mg/kgBB/hari. Perlakuan dilakukan selama 21 hari secara per oral, pada hari ke-22 diberi suntikan CCl4 1,0mL/kgBB secara intraperitoneal kecuali kelompok I. Pengambilan darah melalui sinus orbitalis untuk diukur aktivitas SGOT dan SGPT. Hasil yang diperoleh dianalisis statistik menggunakan uji One Way ANOVA dan dilanjutkan dengan uji LSD dengan taraf kepercayaan 95 %. Hasil penelitian dapat disimpulkan bahwa ekstrak etanol ganggang hijau (Ulva lactuca L.) dapat menurunkan aktivitas SGOT dan SGPT pada tikus jantan putih yang diinduksi CCl4.

Abstract

Kanker adalah sekelompok penyakit yang timbul apabila sebuah sel atau sekelompok sel lepas dari kontrol yang mengatur pertumbuhan. Ganggang hijau (Spirogyra sp.) merupakan salah satu tanaman obat yang digunakan sebagai obat tradisional untuk pengobatan kanker. Ganggang hijau (Spirogyra sp.) mempunyai kandungan zat aktif berupa melatonin dimana melatonin merupakan senyawa yang sudah diteliti para peneliti dunia sebagai obat antikanker dan antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol ganggang hijau (Spirogyra sp.) terhadap ekspresi COX-2 pada sel T47D. Ganggang hijau diekstraksi dengan menggunakan alat Soxhlet dengan pelarut etanol 96%. Penelitian ini menggunakan 3 kelompok yaitu kontrol sel, pemberian ekstrak etanol ganggang hijau konsentrasi 247,668 μg/ml dan 123,834 μg/ml. Untuk memastikan adanya ekspresi COX-2 dilakukan uji imunositokimia secara tidak langsung. Pengamatan dilakukan menggunakan mikroskop cahaya untuk melihat dan menghitung persen ekspresi COX-2. Hasil yang didapatkan menunjukkan terdapat perbedaan yang bermakna penurunan ekspresi COX-2 pada tiap kelompok.. Jadi dapat disimpulkan bahwa ekstrak etanol ganggang hijau (Spirogyra sp.) mampu menurunkan ekspresi COX-2 pada sel T47D.

Abstract

Green Algae, an organism with active substance such as phytomelatonin, has potential to be developed as Indonesian traditional medicine. As the long term addition of Green Algae ethanol extract (Ekstrak etanol ganggang hijau, EEGH) influences the hematology system, in this paper, the safety test was done to ensure the safety of its use through subchronic toxicity test of EEGH on the hematology parameters of Wistar rats. The test group consisted of three groups treated with EEGH 100 mg/kg, 200 mg/kg, and 400 mg/kg, while the control group was given by 0.5% CMC-Na, with 8 rats each respectively. By using blood samples taken from orbital sinus on the 29th day, common hematologic parameters (erythrocytes, leukocytes, and hemoglobin level), the parameters of hemostasis (platelets, pT, aPTT, BT) and immune parameters (Differential Leukocytes Counts include neutrophils segment, lymphocytes, monocytes, and eosinophils) were finally observed and showed that the 28 days-addition of EEGH increase the hematological parameters of Wistar rats.

Abstract

Previous studies showed the potential of melatonin of green algae ( Ulva lactuca L ) to reduce the risk of coronary heart disease with the activity of antihiperlipidemia. Obesity and hiperlipidemia were the risk factor of degenerative diseases such as heart disease. This research aims was explore the effect of ethanol extract of green algae against weight, consumption of rat feed test white male rat were given high fat diet . Research was started with extraction green algae with ethanol 96 % to obtained concentrated extract. The extract was tested to wistar rats age of 2 months. Animals test divided into 6 group. Group I was control group, treated with high fat diet of lard 2 ml / 200g BW, of group II given high fat diet and simvastatin, group III, IV and V given high fat diet and green algae extract doses 50 mg/ Kg BW, 100 mg/Kg BW and 200 mg/ Kg BW. Group VI is the control group without diet fat high. Treatment was conducted over 28 days. Measuring normal of his weight every five day for 28 days and in measuring consumption feed with measure weight feed the rest of feed early 20 g. The difference of data weight rats per 5 days and consumption fodder for 28 days counted area under curve ( AUC ) from curves time versus weight. The measurement of triglycerides levels. The result showed the high fat diet with lard 2 ml / 200g BW for 28 days reduce weight and consumption feed mice in a significantly than control without high fat diet. The treatment with extract ethanol dose of 50 , 100 and 200 mg / Kg BW did not reduce weight, feed consumption, triglyceride levelsof  mice were given high fat diet.
