Abstract

The purpose of this research was to induce polyploidy on Impatiens balsamina L. Seedlings of I. balsamina was immersed in 0,01% colchicines solution for 4, 6, 8, and 24 hours. Chromosome of I. balsamina was visualized using squash method stained with aceto-orcein. Morphological observations were conducted on plant height, stem circumference, leaf length and width, number of branches flowering time and size of flower. The result revealed that 0,01% solution of colchicine was able to induce polyploidy on I. balsamina. Obsevation on morphological characteristic showed that colchicine treatment increased plant height, stem circumference, leaf length and number of branches. Colchicine treatment induced earlier flowering time, but flower size was unaffected. Twelve chromosomes (2n = 2x = 12) were observed in the diploid seedlings while 25 chromosomes were observed in the tetraploid (2n = 4x = 24 seedlings.

Abstract

A field experiment has been conducted at Pesinggahan up-land regency of Klungkung Bali, to know the interaction effect between planted space and bio-urine dosage on the optimal production of Panicum maximum. Randomized Completely Block Design was arranged in factorial model. The planted space (J) treatments were: J1 = 10 cm ? 20 cm, J2 = 20 cm ? 20 cm, and J3 = 40 cm ? 20 cm, and the bio-urine (B) treatments were: B0 = without bio-urine, B1 = 3,750 l ha-1 (1.44 l plot-1), B2 = 7,500 l ha-1 (2.88 l plot -1), B3 = 11,250 l ha-1 (4.32 l plot -1) and each treatment replicated three times. Result of these experiment showed that a significant interaction (P<0.05) between bio-urine fertilizer and planted space on dry weight leaf and dry weight of total forage in the second regrowth. The highest yield of dry weight forage was on the bio-urine 7,500 l ha-1 (B2) dosage and 10 cm ? 20 cm (J2) space of planting. The optimal dosage was on 8,007.9 l ha-1 with 15,05 ton ha-1 maximal production.

Abstract

Impatiens balsamina L. is a plant that is widely cultivated in Bali. The flower of this plant is used as a component of offering in Hindu religious ceremonies. The flower petals are thin and wilt easily, therefore genetic modification is needed to get better quality. This research aimed to study the effect of 0.01% colchicine with soaking periods of 4 hours, 8 hours, 12 hours and 24 hours to seedlings of I. balsamina. Seeds of I. balsamina were germinated and sprouted seeds were treated with 0.01% colchicine for 4 hours, 8 hours, 12 hours and 24 hours. The sprouted seeds were then transfer to polybag with soil media. The experiment was designed with randomized blok design with five replicates. Observations were done on plant height, length and width of leaf, stem diameter, number of stomata as well as observation on plant stem cortex cells. Results showed that colchicine reduced plant height at three weeks after planting, while plant stem diameter increased. Control plants had the smallest leaf length and width. The number of stomata decreased at plants derived from colchicine treated sprouted seeds and the lowest stomatal number observed was at soaking duration of 24 hours.
