Abstract

This research studied the application of chitosan on salted mackerel (Rastrelliger sp.) preservation during storage at room temperature. The research is aimed to study the effect chitosan concentration and storage time for Total Plate Count (TPC). The study was conducted in Laboratory of Microbiology, Faculty of Mathematics and Natural Sciences, Syiah Kuala University from April to August 2012. The research method was laboratory experimental.  The data was analyzed by Randomized Completely Design with two factorial. The first factor was chitosan concentration (three levels: 0%; 1%; 2%) while the second factor was storage time (four levels: 0; 2; 4; 8 weeks). The results of this study indicated that chitosan concentration was not significantly different for the Total Plate Count (p 0,05), however storage time was significantly for the Total Plate Count (p 0,05). Futhermore, the interaction chitosan concentration and storage time was significantly for the Total Plate Count (p 0,05).

Abstract

One of the alternatives to antimicrobials in disease control is probiotic bacteria as microbial control agents. This research was carried out to obtain indegenous isolates of probiotic bacteria from digestive tract Cyprinus carpio. Selection criteria were appointed based on the capacity to produce the antibacterial compound against the pathogen and grow at pH 2. The research was conducted from May to July 2014 at Microbiology Laboratory, Biology Department, UNSYIAH. The research was carried out by experiment laboratories. The planting of the bacteria was carried out with the pour-plate method in nutrient agar medium. The result show that there are six bacterial isolates varied morphological colony and cell and able to produce antibacterial compounds for active against A.hydrophila and E. tarda.

Abstract

The aims of this research were to determine the effectiveness of waste water processing in reducing content of phenol in RSUDZA. Research was conducted from June to September 2009. This research used 2 locations sampling which consist the tub before processing (inlet) and after processing (outlet), then data taken in 10 repetition consecutive days. The observed parameter ware phenol content and parameter like pH, BOD (Biochemical Oxygen Demand), COD (Chemical Oxygen Demand) dan TSS (Total Suspendid Solid) content. Data were analyzed by using paired t test (P 0,05). The result show that fenol content were significant different before and after the processing. Phenol content decreased from 32% to 96.4%. Phenol content (0.020 mg/L) outlet had fulfilled the quality standard already established. These refer as judged effective values the pH parameters show that there were increasing. The result also show that not only phenol content decrease but also BOD, COD and TSS content.

Abstract

This study was undertaken to investigate soil Actinomycetes from grand forest park of Pocut Meurah Intan as well as to evaluate their potential to produce antimicrobial compounds. The study was conducted in Laboratory of Microbiology, Faculty of Mathematics and Natural Sciences, Syiah Kuala University from February to July 2012. The research was carried out by using an explorative method with laboratorial evaluation. Three different soil samples were taken from a depth of 10-20 cm below the soil surface and isolated on YM agar medium. During the investigation, 24 isolates were recovered and exhibited various morphological characteristics. The results of the antimicrobial assay showed that 14 isolates (58.33%) were active against one or more of the test organisms. Among the 14 active isolates, one isolate (7.14%) showed antibacterial activity; 9 isolates (64.29%) showed antifungal activity; and 4 isolates (28.57%)  exhibited a broad-spectrum activity against both of the test bacteria and fungi. The inhibitory activity were also varied, ranged from weak ( 5 mm), moderate (5-19 mm), to strong ( 20 mm).  The highest antimicrobial activity was presented by ATH-17 against Candida albicans (34.50 mm), while the lowest was exhibited by ATH-15 against Escherichia coli (0.20 mm).

Abstract

Tsunami devastated many water infrastructures including the wells. Tsunami areas, land use (rural and urban areas) and land management (agriculture land and home garden) can be leading heavy metals in water quality. The purpose of this research is to investigate how the Tsunami rehabilitation efforts affected the water quality of shallow groundwater by measuring the count of heavy metals (mercury, cadmium and lead by Atomic Absorption Spectrophotometer during the rainy and dry season. Forty-eight water samples? of wells were collected to determine shallow groundwater from August 2007 to December 2007. The study includes interviews with the owners of the water facilities and sanitary inspection of shallow groundwater wells. The results show the presence Hg and most of Cd in well water are still below the regulatory acceptable limit by Ministry of Health R.I (2002). On the other hand, lead exceeds the regulatory acceptable limit

Abstract

The research was conducted to determine the quality of the kanji pedah by counting total number and percentage of Aspergillus flavus and Aspergillus niger. It had been done since March from to October 2010 in Microbiology Laboratory of Biology Departement FMIPA Unsyiah. Kanji pedah as the sample was taken from Meunasah Mesjid Syamtalira village, Aron Subs-district of North Aceh. This research used sample from variety storage time groups range 3-7 days; 5-7 months; 9-12 months; and 15-18 months. Data were analyzed in descriptive and compared with the quality standard from Directorate General of National Agency of Drug and Food Control (Dirjen BPOM RI). The result showed the entire sample had fungi with ranged 1.45-63.7x102. The amount of fungi exceeds the quality standard already established by the National Agency of Drug and Food Control No. 05018/KBPOM/2001: 50 colony/g. Also Aspergillus flavus was found with range from 3%-27% meanwhile Aspergillus niger was found with range from 21,1%-40,9%.

Abstract

Abstract. Saga (Abrus precatorius L.) have been used by Indonesian community as a traditional medicine. The research aims to determine the activity of the methanol extract of saga leaves to inhibit the growth of clinical Streptococcus pneumoniae growth. Inhibition tests was done with casting cup method. The results showed that the methanol extract of saga leaves at a concentration of 37,5; 50; 62,5; 75 and 87,5 mg/mL formed growth inhibition zone of clinical S. pneumoniae with consecutive average of 0,5; 1,75; 2,82; 4,00; and 4,82 mm. Abstract

Bakteri Asam Laktat (BAL) merupakan kelompok bakteri yang mampu mengubah karbohidrat  (glukosa) menjadi asam laktat. Penelitian ini bertujuan untuk mengisolasi dan mengetahui karakteristik BAL yang berperan dalam aktivitas fermentasi asam laktat pada biji kakao (Theobroma cacao L.).  Parameter yang diamati adalah morfologi koloni (bentuk, tepian, elevasi dan warna isolat), morfologi sel (bentuk sel dan pewarnaan Gram), uji biokimia (TSIA, indol, motilitas, MR-VP, tipe fermentasi, ketahanan suhu 15oC dan 37oC, ketahanan garam 5%, 6.5% dan 10%, serta katalase,) dan uji aktivitas antimikroba.  Dari hasil penelitian ini diperoleh tiga isolat BAL yang merupakan kelompok bakteri Gram positif berbentuk basil dan kokus. Ketiga isolat secara umum memiliki morfologi koloni yang berbeda yaitu berbentuk bundar, tepian licin, elevasi cembung dan datar, berwarna krem dan putih susu. Hasil uji biokimia menunjukkan bahwa ketiga isolat termasuk famili Lactobacillaceae, genus Lactobacillus (isolat FBK 1 dan FBK 3) dan genus Enterococcus (isolat FBK2).  Pada uji daya hambat (aktivitas antimikroba), ketiga isolat mampu menghambat pertumbuhan bakteri Escherichia coli dan Staphylococcus aureus.

Abstract

This aims of this research were to determine the optimum combination of papain and bromelin enzymes in feed to increase the growth performance of nilem fish (Osteochillus vitattus). The research was conducted in three places, namely the Laboratory of Faculty Agriculture Syiah Kuala University, Brackish Water Aquaculture Center, Aceh Besar, and Laboratory of Faculty Marine and Fisheries Syiah Kuala University in December 2016 until February 2017. The research consisted of 6 treatments and 4 replications. The tested treatments were the differences of the combination of papain and bromelin enzymes in the feed i.e, 100% papain + 0% bromelin, 75% papain + 25% bromelin, 50% papain + 50% bromelin, 25% papain + 75% Bromelin, and 0% Papain + 100% bromelin.  Fish was fed three times for at feeding level of 5% biomass for 70 days. The result showed that the absolute growth was 4,26 g to 6,65 g, specific growth rate was 0.05%/ day – 0.75%/ day, daily growth rate was 8.51-10.88 g/day, survival rate was 90% -100%, feed conversion ratio was 4.60-5.32, and feed efficiency was 18.94% - 24.70%. The highest values all parameters were found at 75% papain + 25% bromelin treatment. The ANOVA test showed that of the combination of papain and bromelin enzymes in feeding had the significant effect on weight gain, specific growth rate, daily growth rate, feed conversion ratio, and feed efficiency of nilem fish. The best combinations of papain and bromelin enzymes in this study were 75% papain + 25% bromelin.       Penelitian ini bertujuan untuk menentukan konsentrasi optimum kombinasi enzim papain dan enzim bromelin dalam pakan untuk meningkatkan pertumbuhan ikan nilem (Osteochillus vitattus) . penelitian ini dilakukan di tiga tempat yaitu, Laboratorium Fakultas Pertanian Universitas Syiah Kuala, Balai Budidaya Air payau, Aceh Besar, dan Laboratorium Fakultas Kelautan dan Perikanan Universitas Syiah Kuala pada bulan Desember 2016 sampai Februari 2017. Penelitian ini terdiri dari 6 taraf perlakuan dan 4 ulangan. Perlakuan yang diuji adalah perbedaan konsentrasi kombinasi enzim papain dan enzim bromelin dalam pakan yaitu, pakan tanpa uji, 100% papain + 0% Bromelin, 75% papain + 25% bromelin, 50% papain+ 50% bromelin, 25% papain + 75% bromelin, , dan 0% Papain + 100% bromelin. . Ikan diberi pakan tiga kali sehari sebanyak 5% dari biomassa selama 70 hari penelitian. Hasil penelitian diperoleh pertambuhan bobot berkisar 4,26 g-6,56g, laju pertumbuhan spesifik 0,05% per hari- 0,75% per hari, laju pertumbuhan harian 8,51 g per hari-10,88g per hari, tingkat kelangsungan hidup 90%-100%, rasio konversi pakan 4,60-5,32, dan efesiensi pakan 18,94%-24,70%. Nilai tertinggi pada setiap parameter yang diuji terdapat pada perlakuan 75% papain + 25% bromelin. Hasil uji ANOVA menunjukkan bahwa penambahan kombinasi enzim papain dan enzim bromelin dalam pakan berpengaruh nyata terhadap pertambuhan bobot mutlak, laju pertumbuhan spesifik, laju pertumbuhan harian, ratio konversi pakan, nilai efisiensi pakan, dan koefisiensi keragaman ikan nilem akan tetapi tidak berpengaruh nyata terhadap nilai kelangsungan hidup. Hasil penelitian menunjukkan bahwa penambahan kombinasi enzim papain dan enzim bromelin dalam pakan meningkatkan pertumbuhan ikan nilem (Osteochillus vitattus). Konsentrasi kombinasi enzim papain dan enzim bromelin terbaik pada penelitian ini adalah 75% papain + 25% bromelin

Abstract

ABSTRACTTurmeric is a herb and potential as antimicrobial. The objective of this study was to tigme out the clinical symptoms of catfish (Pangasius sp.) after infection  by Aeromonas hydrophila, to determine the effect of turmeric extract (Curcumin domestica) on the survival rate of catfish that was infected by A. hydrophila and the optimum concentration of the extract. This study was carried out experimentally using a Completely Randomized Design (CRD) with five treatments of extract concentration (0; 0.4 ; 0.5 ; 0.6 ; and 0.7 ppt) and four repetitions. It was observed that the catfish  had been cured using turmeric extract since12 days after infection. The results of ANOVA showed that the extract turmeric (Curcuma domestica) gave the significant effect on the survival rate of catfish (Pangasius sp). The higher survival rate was obtained at 0.7 ppt (77.5%).Keywords : Pangasius sp, Curcuma domestica, Aeromonas hydrophila.  ABSTRAKKunyit adalah jenis rempah-rempah yang mengandung metabolit sekunder yang berperan sebagai antimikroba. Penelitian ini bertujuan untuk melihat perubahan gejala klinis ikan patin (Pangasius sp.) pasca infeksi bakteri Aeromonas hydrophila, mengetahui pengaruh ekstrak kunyit (Curcuma domestica) terhadap kelangsungan hidup ikan patin yang terinfeksi bakteri A. hydrophila serta konsentrasi optimum ekstrak kunyit. Penelitian ini dilakukan secara eksperimental menggunakan rancangan acak lengkap (RAL) dengan lima taraf perlakuan dan empat kali pengulangan. Perlakuan yang diberikan meliputi konsentrasi ekstrak kunyit dengan konsentrasi 0 (kontrol), 0,4; 0,5; 0,6; dan 0,7 ppt. Hasil pengamatan menunjukkan bahwa ekstrak kunyit telah menyembuhkan ikan patin pada hari ke-12 pasca infeksi bakteri.  Hasil uji ANOVA menunjukkan bahwa ekstrak kunyit (C. domestica) berpengaruh nyata (P < 0,05) terhadap kelangsungan hidup ikan patin. Nilai kelangsungan hidup ikan patin terbaik diperoleh pada perlakuan 0,7 ppt yaitu (77,5%).Kata kunci: Pangasius sp, Curcuma domestica, Ae romonas hydrophila
