Abstract

The aim of this research was to determine the copepod daily feeding to Dunaliella salina and Chlorella sp because the research of copepod daily consumption to the phytoplankton in the sea was a part of discovering the water productivity. The determination was needed in the laboratory experimental method, in order to qualifying the biomass and energy transfer of sea food web / chain. The observation based on the laboratory experimental method and conducted in the Hatcehery and Laboratory of Alga, Marine Station, Teluk Awur Jepara. Four treatment were applied during the observation, i.e.: A. D.salina 20 litre; B. D.salina  2 litre; C. Chlorella sp 20 litre; D. Chlorella sp 2 liter. The copepod grazing rate was determined base on parameter of cell phytoplankton/ ml/ hour/ copepode. There were 4 series observation. The observations were done for 36 hours with 3 hours interval. The data collected was arranged as copepod daily consumption and analyzed using MAXIMS Analysis Models. The highest copepod daily grazing rate was 88.02 ± 44.18 cell phytoplankton/ ml/ hour/ copepod for D.salina on 20 liter volume of media. While the lowest one 50.16 ± 43.99 cell phytoplankton/ ml/ hour / copepod for Chlorella sp on 2 liters volume media. The Copepod daily grazing model tends to form a constant model for D.salina on 20 liters of media and proportional model for 2 liters volume of media. The Chlorella sp on 2 liters volume of media graze by the copepod showed the same model for constant and proportional model. The water quality of the media remained in tolerance range to support the grazing rate of copepod to the phytoplankton during the research.

Abstract

The pond area of Bedono waters become a shallow water, due to the influence of high tide (rob). This condition affect the macrozoobenthos, one of the animals that live in the area. The objective of this research is to look for the macrozoobenthos community structure of in the Region Morosari Waters, Bedono District of Sayung, Demak. Twenty five polychaeta species were found in this research, 7 species of bivalves, 6 species of gastropods and 1 species of crustacean, with abundances ranging between 36-1331 ind/m2 with diversity index value (H ') in each research ranged from 2.95 (moderate diversity level) to 0.83 (low diversity level), with evennes index (e) ranged from 0.46 (moderate evennes index) to 1.00 (high evennes index). Domination index (C) ranged from 0.15 (this value shows that no domination index) to 0.69 (this value shows that domination index).

Abstract

Dunaliella salina and Chlorella vulgaris is a natural feed microalgae with high carotenoid content that can be increased using protoplast fusion technique. Protoplast fusion as one of the application fields of genetic engineering is a method for obtaining recombinant with the desired properties and profitable in a short time. This study aimed to see the effect of the addition of carotenoid-rich feed results from protoplast fusion recombinant D. salina and C. vulgaris on the survival rate and weight of shrimp post larvae. Mixed fusion results feed and artificial feed needed for the growth of post-larval shrimp, moulting and skin pigmentation. The results showed that the recombinant from protoplast fusion intergenera D. salina and C. vulgaris contains carotenoid pigments higher, reaching 124.6 mg / g bks from the second parent, namely D. salina reached 101.83 mg / g bks, while C. vulgaris 97.18 ug / g bks. Feed manufacturing is done by mixing pellets and 80-100x103 cells per 0.0225 g of feed. Application of feed carried on Penaeus monodon F. (tiger prawn) stadia PL-20 for a month. The results of weight measurements showed the highest prawn post larvae reached at artificial feeding plus recombinant protoplast fusion results intergenera D. salina and C. vulgaris and was able to raise the level of post-larval shrimp survival rate reached 88%.   Keywords: D. salina, C. vulgaris, Protoplast Fusion, Carotenoid, Penaeus monodon F.

Abstract

Pandansari estuary waters is an area which flood by high tide or known as ?rob?and became a shallow water area. Macrozoobenthos is used as one of the biological component in monitoring of water quality. The purpose of this study was to determine the macrozoobenthos community structure in Pandansari waters, District of Sayung, Demak. There were 4 stations earned 48 genera consist of 7 class, including in the 6 phyla of Molluscs (35 species), Polychaeta (17 species), Crustaceans (5 species), Sipuncula (1 species), nematodes (1 species) and Nemertina (1 species). Macrozoobenthos abundance ranged from 1.910-21.442 ind/m3, with a diversity index of 0.45-2.67, which indicated that the carrying capacity of Pandansari waters to support macrozoobenthos live is low to moderate. The evenness index ranged from 0.23-0.97 are in the category of small to high which indicated the proportion is not the same type of macrozoobenthos, and dominance index of 0.08-0.82, which indicated there was species dominating macrozoobenthos in the waters of Pandansari, district of Sayung, Demak.

Abstract

Morosari waters located in Sayung District, Demak Regency is waters area where there is a mixture of land and sea water. Settlement activity, aquaculture, industrial land and existing at the entry and exit activities of fishing boats around the estuary can contribute to the dynamics of water quality fluctuations and productivity. The dynamics of water quality that occur greatly affect community structure and distribution of phytoplankton in the waters. The results of this study showed 28 genera for the vertical method, while for the method of horizontally obtained 31 genera. Average abundance obtained by the observation station was 1087.09 cells/l with vertical method, while for the horizontal method obtained 884.69 cells/l. While the abundances obtained by times of sampling average of 1087.09 cells/l for the vertical method, while for the horizontal method was 831.38 cells/l. Index phytoplankton diversity showed medium and obtained the average gain by the station is 2.02 for method of vertical and horizontally method to obtain 1.85. While based on the sampling time obtained an average of 2.09 for the vertical method, and for the horizontal method is 1.58. Uniformity index indicates the type of high uniformity and obtained the average by the station is 0.68 for the vertical method, while the horizontal method is obtained 0.4. While the sampling time is obtained based average of 0.64 for the vertical method, while for the horizontal method is 0.51. Dominance index obtained average based on the observation station is 0.3 for the vertical method, while the horizontal method obtained 0.6. While the sampling time is obtained based on an average of 0.35 for the vertical method, where for the horizontal method is 0.48 which indicates there is no dominating genus.

Abstract

Energi matahari dibutuhkan oleh phytoplankton dilaut dalam proses fotosintesis. Laju fotosintesis akan meningkat bila intensitas cahaya meningkat dan menurun bila intensitas cahaya berkurang, sehingga cahaya berperan sebagai faktor pembatas utama dalam fotosintesis atau produktvitas primer. Penelitian ini dilakukan untuk pembuatan dan pengujian pengaturan intensitas cahaya lampu buatan untuk proses fotosintesis fitoplankton Dunaliella sp. Cahaya lampu diatur intensitasnya mengikuti pola cahaya matahari dengan panjang gelombang 0.4?m- 0.7?m menggunakan rangkaian konverter AC-AC. Konverter AC-AC tersebut menggunakan rangkaian TRIAC yang pemicuannya dikontrol dengan mikrokontroler AT89S52. Hasil pengujian menunjukkan bahwadari perlakuan siklus on-off (A), siklus 3 jam (B) dan siklus matahari buatan (C), menunjukkan bahwa respon fotosintesis yang paling bagus dan efisien adalah perlakuan dengan siklus matahari buatan (C)Kata kunci : Intensitas cahaya buatan, fitoplankton Dunaliella sp., microcontroller, AC-AC converterSun light intensity plays an important role to photosynthesis process in the sea water. The photosynthesis rates will be high when the light intensity is high and it will be down when the light intensity decreases.Therefore light play significant role as the limiting factor in primary productivity of photosynthesis. The research purpose is to provide the artificial lighting lamp for phytoplankton. (Dunaliella sp.) photosynthesis process. The lighting lamp intensity is controlled in order to be similar with sunlight intensity pattern that has wavelength 0.4 mm 0.8mm by using AC - AC converter circuit. The AC-AC converter uses TRIAC triggered by AT89S52 microcontroller. The experimental result indicates that the artificial lighting controller can be used to support phytoplankton photosynthesis process. This is indicated by the dissolved oxygen (DO) and the density of phytoplankton which tend to follow the pattern of the given lighting intensity.Key words: artificial lighting, phytoplankton Dunaliella sp., microcontroller, AC-AC Converter

Abstract

Perairan Selat Badung memiliki karakteristik yang sebagian besar dipengaruhi oleh aktivitas Arus Lintas Indonesia (ARLINDO). Pencampuran massa air akibat pergerakan massa air vertikal menjadikan kondisi lapisan yang homogen, dimana nilai suhu, salinitas dan densitas berada pada nilai yang hampir sama di lapisan tertentu akan membentuk Mixed Layer Depth (MLD). Penelitian dilakukan untuk mengetahui karakteristik MLD dan pengaruhnya terhadap konsentrasi klorofil-a di Perairan Selat Badung, Bali. Metode pengukuran klorofil-a menggunakan spektrofotometri. Hasil penelitian menunjukkan bahwa suhu dan salinitas sebagai parameter MLD, membentuk lapisan yang homogen pada kedalaman yang bervariasi. Sebaran kandungan klorofil-a pada kedalaman MLD 12-23 m, menunjukkan nilai klorofil-a yang cenderung lebih tinggi pada permukaan perairan dibandingkan di perairan yang lebih dalam. Pada MLD kedalaman 12-60 m, menunjukkan bahwa kecenderungan kandungan klorofil-a lebih tinggi pada lapisan di kedalaman tersebut. Namun pada beberapa stasiun menunjukkan bahwa meskipun terdapat lapisan homogen yang cukup dalam, kandungan klorofil-a lebih tinggi di lapisan permukaan dibandingkan pada perairan yang lebih dalam. Kata Kunci: mixed layer depth, klorofil-a, perairan selat Badung Badung Strait characteristics is largely influenced by the ARLINDO (Indonesian Throughflow) current activity. The mixing of water masses due to the vertical movement of water masses, homogenized some range of layer (Mixed Layer Depth), whereas the value of temperature, salinity and density were about on the same range. The study was conducted to determine the characteristics of MLD and its influence on the concentration of chlorophyll-a of Badung Strait, Bali. Chlorophyll-a content was measured by using spectrophotometry method. The results showed that temperature and salinity as the MLD parameters, formed homogeneous layer (MLD) at varying depths. Distribution of MLD at depth of 12-23 m, indicating that chlorophyll-a consentration tends to be higher on the surface than at depth. In conditions at depth of 12-60 m, showed that chlorophyll-a higher on the depth, where a lot of MLD formed on the layer. However, in some stations showed that although there were quite a lot of homogeneous layer, chlorophyll-a consentration was higher on the surface than in the depth. Abstract

Nelayan yang tinggal di sekitar perairan pantai Kabupaten Batang adalah pihak yang secara langsung memiliki hubungan dengan perairan dan menggunakan jaring arad sebagai alat yang digunakan untuk memanfaatkan produknya. Ketergantungan nelayan di sekitar perairan pantai Kabupaten Batang dalam memanfaatkan sumber daya alam masih tinggi sehingga memerlukan usaha alternatif untuk meningkatkan perekonomian mereka melalui program pemberdayaan nelayan. Daya nelayan adalah elemen yang memungkinkan nelayan bertahan hidup dan dapat mengembangkan diri dalam mencapai kemajuan dan kekayaan. Penelitian ini bertujuan untuk mengetahui tingkat daya sosial dan ekonomi nelayan jaring arad yang berada di perairan pantai Kabupaten Batang dan peran pemangku kepentingan terkait dalam pemberdayaan nelayan. Untuk mendapatkan data primer, survei dilakukan untuk 145 responden (±30% dari nelayan yang ada di perairan pantai Kabupaten Batang) pada bulan Maret tahun 2014. Kemudian dilakukan analisis deskriptif dan crosstab. Tingkat daya nelayan jaring arad di perairan pantai Kabupaten Batang masih dikategorikan ke dalam berdaya, baik dari aspek ekonomi dan non. Hal ini dapat dilihat dari rendahnya akses teknologi, keputusan bisnis, dan keterampilan lobi. Para pemangku kepentingan yang membantu bisnis nelayan sebagian besar berasal dari pemerintah dan masyarakat dengan skor 7 (cukup). Oleh karena itu, strategi pemberdayaan diperlukan untuk meningkatkan daya nelayan sehingga peningkatan kesejahteraan dapat terwujud.Kata kunci : Nelayan jaring arad, tingkat keberdayaan, perairan pantai kabupaten BatangFishermen who live around the coastal waters of the Batang which is directly linked to water and use nets arad as a tool that is used to exploit its products. Dependence inshore fishermen around Batang inutilizing natural resourcesis still high so it take seffort to improve their economical ternatives through empowerment programs fishermen. Power fishing is the element that allows fishermen to survive and be able to develop them selves in achieving progress and wealth. This study aims to determine the level of social and economic power arad fishing nets which are in Batang coastal waters and the role of the stakeholders involved in the empowerment of fishermen. To obtain primary data, a survey was conducted for145 respondents (±30% of the fishermen in the coastal waters Batang) in March 2014 and descriptive analysis was then performed cross tab. The power level fishing arad nets in coastal waters Batang still categorized into powerless, both from the economic aspects. It can be seen from low technology access, business decisions, and lobbying skills. The stakeholders who help businesses fishermen mostly from government and society with a score of 7(enough). Therefore, empowerment strategy is required to icrease the welfare of fishermen so that improvement can be realized.Keywords : Fishermen arad nets, level empowerment, coastal waters of the Batang district

Abstract

Prupuh village is a village in the district Panceng by having a hamlet that Hamlet Prohibition. 2000more residents in the village Prupuh with 575 heads of household, and 400 homes were built. In generallivelihoods of the majority of the villagers are farmers Prupuh. In the rainy season corn is one of the cropsgrown by the residents both in the fields and in the yard. Corn Transformation Machine addressed to thevillagers of Prupuh that planting corn with manual harvesting. People have become less productive becauseof the time needed to separate the corn from bongkolnya very long. From the experimental results usingCorn Transformation Machine tool time needed to separate the corn seed from bongkolnya ie 30 secondsper 1 piece of corn, while if using a hand (manual) takes 5 minutes per 1 piece of corn. The tool can savetime for 4.5 minutes and simultaneously increase productivity the amount of corn kernels are separatedfrom bongkol.

Abstract

Diversification of feed rich in natural carotenoids is needed in aquaculture because they will increase survival and body weight of animal farming. Different types of important carotenoids such as β-carotene, zeaxanthine can be combined through a process of protoplast fusion making it cheaper and faster. Crustaceans can not synthetize carotenoid de novo and they need it to provide nutrition and possibly disease resistance, pigmentation and esthetic value. Green microalgae produce carotenoids and can be manipulated easily by protoplast fusion. The research was conducted to obtain some fusants from interspesies protoplast fusion of D. salina and C. vulgaris. Interspecies protoplas fusion was carried out by protoplast isolation, protoplast fusion and protoplast regeneration. Microscopic and cell analysis will used to confirm positive regenerate protoplast. Analysis of the obtained fusants is limited to morphological description due to the complexity and variability of fusant. The stabilities of fusants obtained were examined by successive subcultures. The result revealed that the fusant from D. salina and C. vulgaris maintain their stability in growth and increased carotenoid production during four periods of subculturing. This result suggesting the potency of fusant to be used as food supplement.   Key words: Carotenoid, Protoplast fusion, Dunaliella, Chlorella
