Abstract

 Cinnamon has been used as a traditional treatment for thousands of years. Aqueous cinnamon extract namely cinnulin is known to have  positive effects on type-2 diabetes mellitus (T2DM). We aimed to study the effect of cinnulin enriched drink rich in polyphenols on plasma malondialdehyde (MDA), interleukin-6 (IL-6) and cyclooxygenase-2 (COX-2) in T2DM patients. 255 mg of cinnulin was mixed with tea extract into a 2.5 gram of instant drink with alumunium foil as the packaging material. A single blind study were performed on 33 respondents with T2DM from Dramaga District. Respondents were divided into three groups, i.e. group given cinnulin enriched drink (1), instant tea drink (2), and none of the products (control) (3). Group 1 and 2 were assigned to drink two sachetsof the products a day for 5 weeks. The MDA content of all groups remain unchanged after treatment although slightly increased (Wilcoxon, P>0.050) and no differences between groups was observed (P>0.050) suggesting that high phenolic content in the producst cause a reverse effect. Both inflammation marker (IL-6 and COX-2) decreased significantly for groups given cinnulin enriched drink and instant tea, whereas there is a significant increase for IL-6 and a significant decrease for COX-2 in control group. These results conclude that cinnulin enriched drink can be used as an antiinflammation drink for T2DM.

Abstract

The health benefits of cocoa both in vivo and in vitro have been reported in many studies. Cocoa is a rich source of flavonoids known to have antioxidant activity, such as catechin, epicatechin and procyanidin. The aim of this research was to evaluate the effect of fat free cocoa powder drink consumption on antioxidative properties and proliferation activities of woman lymphocyte. Healthy woman subjects were divided into cocoa group (n = 9) and control group (n = 9). Cocoa powder drink containing skim milk and sugar was given to the cocoa groups every morning for 25 days. The control group received only water containing skim milk and sugar. Both cocoa and control group received physical medical checkup at the beginning and at the end of the intervention. Their peripheral blood was taken for lymphocyte antioxidant analysis. The measured antioxidant properties consisted of antiradical activity by DPPH method, malonaldehyde (MDA) and glutathione levels. The data of cocoa group showed that there was a significant increase (p ? 0.05) in antiradical level from 31+11.2 to 40.19+7.42% and glutathione from 48.2±10.5 to 66.7±15.9 ?mol/land a decrease in MDA level in the lymphocyte (p < 0.05) from 2.98±2.21 to 1.29±0.33 ?mol/las compared to the control group (from 25.77±6.9 to 26.79±6.12%; 34.7±20.7 to 37.8±19.2 ?mol/land 3.01±1.53 to 2.069±0.707 ?mol/l respectively) after consumption of the cocoa powder drink. The results of this research revealed that fat free cocoa powder has a strong antioxidant activity which was manifested up to the blood cells.

Abstract

    Antioxidant activity of various ginger extracts are different.  The aims of this work are to determine the antioxidant activity of ginger extracts on linoleic acid. The antioxidant activities were tested on  linoleic acid supplemented with water and dichloromethane extract of ginger.   The antioxidant  activity was tested by measuring malonaldehide absorbance.  Total phenol and iron content  of the extracts were analyzed by spectrometry.  This research showed that antioxidant activity of water and dichloromethane extract of ginger was stronger than a-tocopherol, and antioxidant activity of dichloromethane extract is stronger than water extract of ginger.  Antioxidant activity of ginger extracts, seems to be correlated with  the  total phenol and iron contents.

Abstract

Ikan selar kuning (Seloroides leptoleptis) merupakan salah satu jenis ikan berukuran kecil yang pemanfaatannya masih terbatas dalam bentuk segar dan pengeringan secara tradisional. Penelitian untuk mengembangkan teknologi pengolahan akan dapat meningkatkan nilai guna ikan tersebut. Salah satu teknologi pengolahan yang patut mendapatkan perhatian adalah teknologi pengolahan bubuk ikan.

Abstract

Sorgum merupakan tanaman serealia yang sangat berguna sebagai sumber karbohidrat alternatif dalam program diversifikasi pangan. Sorgum memiliki kandungan protein yang lebih tinggi dibandingkan beras dan berprospek baik untuk dikembangkan di Indonesia. Banyak penelitian telah menunjukkan bahwa sorgum sangat baik untuk kesehatan, antara lain untuk mengurangi resiko penyakit degeneratif. Penelitian secara in vitro sebelumnya, menunjukkan bahwa serealia ini mampu meningkatkan proliferasi limfosit manusia, yang menunjukkan perbaikan sistem imun. Tujuan penelitian ini adalah untuk mempelajari efek sorgum terhadap sistem imun dan kapasitas antioksidan secara in vivo pada tikus. Tiga kelompok tikus diberi pakan kontrol, pakan mengandung 50 persen atau 100 persen sorgum sebagai sumber karbohidrat selama 7 minggu. Hasil penelitian menunjukkan bahwa kelompok tikus yang diberi pakan mengandung 50 persen atau 100 persen sorgum mengalami peningkatan aktivitas proliferasi mencapai berturut-turut 70 persen dan 63 persen, aktivitas antioksidan hati (DPPH) mencapai 38 persen dan 29 persen, aktivitas enzim superoksida dismutase (SOD) mencapai 98 persen dan 91 persen, aktivitas enzim katalase (CAT) mencapai 28 persen dan 21 persen, dan aktivitas glutation peroksida (GPx) mencapai 57 persen dan 33 persen, akan tetapi mengalami penurunan kandungan malondialdehid (MDA) hingga 22 persen dan 16 persen. Penelitian ini menunjukkan bahwa sorgum mempunyai aktifitas imunostimulani dan aktivitas perbaikan antioksidan sehingga baik bagi kesehatan.Sorghum is a cereal that would be useful as alternative carbohydrate source in food diversification program. It has higher protein content than rice and good prospect to be developed in Indonesia. Researches have shown that sorghum has functions in health, such as to decrease degenerative disease risk. Previous in vitro study of sorghum showed that this cereal could increase human lymphocyte cell proliferation in vitro, indicating immune system improvement. The objectives of this research were to study the effects of sorghum on the in vivo immune system and liver antioxidant capacity in rats. Three groups of rats were fed control diet, diet containing 50 percent or 100 percent sorghum as sources of carbohydrate. The results showed that the rats fed with 50 percent or 100 percent sorghum displayed increase in, respectively, proliferation activity by 70 percent and 63 percent; liver antioxidant activity (DPPH) by 38 percent and 29 percent, super dioxide dismutase enzyme activity (SOD) by 98 percent and 91, catalyst enzyme activity (CAT) by 28 percent and 21 percent, and glutathione peroxides enzyme activity (GPx) by 57 percent and 33 percent; but decreased in malondialdehyde (MDA) by 22 percent and 16 percent. This research showed that sorghum has immunostimulation and antioxidant improvement activities and will be very good as source of carbohydrate diet.

Abstract

Natural killer (NK) cell consitutes white blood cells which specifically functions in lysing tumor and virus invected cells. In this research, a commercial ?Jamu?  was tested to observe its effect on NK cells activity against leukemic cell lines (K562) in vitro. Jamu was extracted with hot water, diluted and added into cell cultures consisted of a mixture of human peripheric limphocyte cells, as the source of the effector NK cells, and K562 cell line i.e., the target cells which were cell line derived from human leukemia and had been labelled with H3-thymidine. The mixture of the cells were made by culturing the two cells at the ratio of 50:1 and 100 : 1, respectively. The results showed that lysing activity of NK cells in the presence of ?Jamu? water extract measured as lysing percentage and lysing index increased only slightly, which were not statiscally significant. It should be considered that the test used in this research represents only a part of the lysing mechanism by NK cells against the target cells.  An in vivo test  for a period of time will be recessary to elucidate ffurther this NK cell activity.

Abstract

Penelitian tentang aktivitas perlindungan sel oleh ekstrak rimpang jahe segar dan tunas jahe telah dilakukan secara in vitro terhadap sel limfosit mencit. Komponen fenol yang mempunyai aktivitas antioksidan diperkirakan dapat melindungi sel dari kerusakan dan kematian. Penelitian lain mengungkapkan bahwa komponen fenol terbukti memiliki aktifitas antiproliferatif dan efek toksik terhadap sel kanker. Sel kanker adalah: sel normal yang bermutasi menjadi ganas, Tujuan penelitian ini udalah untuk mengetahui pengaruh ekstrak rimpang jahe segar dan tunas jahe terhadap aktivitas proliferasi: sel kanker K562 (kanker darah) dan A549 (kanker paru-paru) yang berasal dari manusia. Terdapat enam jenis ekstrak juhe yang digunukun sebagi perlakuan, yaitu (1) ekstrak air rimpang jahe segar, (2) ekstrak air tunas jahe, (3) ekstrak air rimpang jahce segar dengan berat molekul kurang dari 8000 dalton, (4) ekstrak air tunas jahe dengan berat moJekul kurang dari 8000 dalton, (5) ekstrak etanol rimpang jahe segar dan (6) ekstrak etanol tunas jahe. Setiap jenis ekstrak jahe terscbut diencerkan, 0 kali (tanpa pengenceran), 2,3 dan 4 kali pengenceran. Setiap jenis ekstrak jahe ditambahkan ke dalam kultur sel  kanker K562 dan A549 yang kemudian diinkubusi selama 4 hari. Pada hari ke tiga di lakukun penambahan 3H timidin untuk menandai DNA sel kanker yang berproliferasi. Hasil penghitungan sel menunjukkan bahwa ekstrak air rimpung jahe segar cenderung menghambat proliferasi sel kanker K562 lebih baik daripada ekstrak lainnya, dengan indeks penghambatan mencapai 80,57%, Sementara itu, ekstrak air tunas jahe dengan berat molekul kurang dari 8000 dalton (8 k Da)ternyata cenderung Iebih menghambat proliferasi sel kanker A549 dibandingkan jenis ekstrak jahe lainnya, dengan bcsar indcks pcnghambatan 51.72%. The Effect of Fresh and Ginger Shoot (Zingiber officinale Roscoe) Extracts on Cancer Cell Line ProliferationCell protection activity of fresh ginger rhizome and ginger shoot on mouse spleen limphocyre had been reported Phenolic compound which has antioxidative activity is expected could protect cell from damage and death. Other research result indicated that, some of phenolics compound had been proven that it had antiproliferative activity and toxic effect to cancer cell. Cancer cell comes from normal cell which mutated becomes malignant sel. The aim of this research was to find out the effect of fresh ginger rhizome and ginger shoot extracts on proliferative activity or K562 (crythroleukemic) and A549 (lung carcinma) human cancer cell lines. There were six kinds of ginger extracts were applied as treatments, They were, (1) water extract fresh ginger rhizome, (2) water extract ginger shoot, (3) Water extract fresh ginger rhizome which had molecule weight less than 8000 daltons (8 kDa), (4) water extract ginger shoot which had molecule weight less than 8000 daltons, (5) ethanol extract fresh ginger rhizome and (6) ethanol extract ginger shoot. Each extract was diluted with RPMI-1640 (0,2,3 and 4 times dilution). Ginger extracts were added into K562 and A549 Cancer cell culture and incubated for four days, AI the third day, 3H thymidine was added to sign the DNA of cancer cell which proliferated. The result of cell counting showed that water extract fresh ginger rhizome lent to inhibit cancer cell line K562 (human erythroleukemic cell) proliferation better than the others, with 80,57% index inhibition. Water extract ginger shoot which had molecule weight less than 8000 daltons tent to inhibit cancer cell line A549 (human lung carcinoma) better than the others with 51,72% index inhibition.

Abstract

Type 2 diabetes mellitus (T2DM) is a non-insulin dependent diabetes, because insulin is not secreted by beta cell pancreas or dysfunction of insulin reseptors. The purposes of this research were to evaluate the effect of dietary fiber rich- tofu from black soybean on the antioxidant plasma status, malonaldehide (MDA) and hepatic marker in T2DM patients. One group (n=11) of patients was given 80 g of tofu for 30 days while the control group (n=11) was not. No significant differences was observed pertaining the characteristics of both groups before interventions (age, body mass index, body weight, body high, systolic blood pressure, diastolic blood pressure and fasting blood glucose). The antioxidant activity, MDA and AST/ALT of both groups were analyzed from plasma at day-0 and day-30. Antioxidant activity was analyzed by DPPH method, MDA level with TBARS method, and AST/ALT by International Federation of Clinical Chemistry (IFCC) method. The result showed that the plasma antioxidant increased (4.77±9.49%), but the MDA (2.11±1.73 nmol/mL) and AST (7.73±16.72 U/L)/ ALT (3.55±12.79 U/L) level decreased. Rich dietary fiber tofu from black soybean may have potential use for improving the health status of T2DM patients.

Abstract

Type 2 diabetes mellitus (T2DM) subjects have a hyperglycemia condition. Hyperglycemia can cause oxidative stress that trigger glucose autooxidation and reactive oxygen species (ROS). ROS also triggers the inflammation and induces the cytokine signalling from cells and body tissues, such as interleukin and prostaglandin enzyme (COX-2). The purpose of this research was to analyze the effect of dietary fiber rich-tofu from black soybean (DFT) on bloods glucose and inflammations of T2DM patients. Intervention group (n=9) was given 80 grams DFT during 30 days and control group (n=9) was not. Blood collections were conducted before and after intervention by certified nurses for all groups for analysis in the laboratory. Fasting Blood Glucose (FBG) was measured by glucometer, while the plasma was analyzed with ELISA to measure the optical density value of IL-6 and COX-2 enzyme. Consumption of DFT resulted in the decline of the optical density of IL-6 value (p=0.00), but DFT could not reduce the FBG and optical density of COX-2 enzyme (p=0.22 and p=0.51). Therefore DFT can be considered as an effective food for reduction of T2DM inflammatory.
