Abstract

Teripang hitam Holothuria atra yang memiliki nilai potensial sebagai bahan baku pada industri farmasi telah dibudidayakan di Balai Bio Industri Laut ? LIPI sejak tahun 2016. Infeksi penyakit dan penurunan sintasan hidup menjadi salah satu kendala dalam kegiatan budidaya teripang jenis ini terutama pada saat pemeliharaan induk. Salah satu upaya untuk mengatasi hal tersebut adalah penggunaan bakteri probiotik. Penelitian ini bertujuan untuk melakukan isolasi dan seleksi bakteri yang menghasilkan enzim protease, selulase, dan amilase sebagai kandidat bakteri probiotik pada sedimen dan saluran pencernaan teripang H. atra. Penumbuhan bakteri dari sedimen dan saluran pencernaan pada media selektif yang ditambahkan substrat selulosa, pati, dan kasein dilakukan untuk melihat potensi bakteri yang mampu menghasilkan enzim selulase, amilase, dan protease melalui proses hidrolisis. Dua isolat bakteri yang memiliki aktivitas enzim selulase dan amilase tertinggi telah diperoleh, yaitu isolat SSe9 dengan indeks Hydrolysis Capacity 7 mm dan isolat SAm6 dengan indeks Hydrolysis Capacity 6,6 mm. Hasil tersebut menunjukkan bahwa kedua isolat bakteri tersebut berpotensi untuk dapat dikembangkan lebih lanjut menjadi bakteri probiotik.

Abstract

ABSTRACT Sandfish (Holothuria scabra) is one of endangered species, which has high economical value but has not been commonly cultivated. One factor that affects the success of sandfish culture is survival of larvae during attachment phase . In this phase the larvae of H. scabra need a substrate as a place to live. This study investigated the effect of seagrass and macroalgae as the settlement substrates on the survival of H. scabra larvae. The experiment trial was conducted in UPT LPBIL LIPI Mataram using completely randomized design with five replications for each substrate treatment. The settlement substrates used were Ulva sp. (P1), Gracilaria gigas (P2), Enhalus acoroides (P3), and Eucheuma cottoni (P4). Surface area was 14x19 cm2 in each treatment. At the beginning of experiment trial, one thousand larvae were distributed into plastic tank containing 10 L seawater for each tank. After 13 days experimental period, the survival of H. scabra larvae showed significantly different among substrates treatment. The highest survival was achieved by E. acoroides (15.53±2.23%), and followed by Ulva sp. (5.07±0.74%), E. cottoni (2.57±0.25%), and G. gigas (1.96±0.17%). Keywords: settlement, Holothuria scabra, substrate, seagrass, macroalgae  ABSTRAK Teripang pasir (Holothuria scabra) adalah salah satu spesies berekonomi tinggi yang terancam punah, tetapi belum banyak dibudidayakan. Salah satu faktor yang mempengaruhi tingkat keberhasilan budidaya teripang pasir adalah sintasan pada fase penempelan. Pada fase tersebut larva H. scabra memerlukan substrat sebagai tempat hidup. Penelitian ini bertujuan untuk mengevaluasi pengaruh jenis substrat lamun dan makroalga yang berbeda terhadap sintasan (STS) larva teripang pasir pada fase penempelan. Penelitian dilaksanakan di Laboratorium Budidaya UPT LPBIL LIPI Mataram menggunakan metode eksperimental rancangan acak lengkap dengan lima ulangan. Perlakuan jenis substrat, yaitu: Ulva sp. (P1), Gracilaria gigas (P2), Enhalus acoroides (P3), dan Eucheuma cottonii (P4). Substrat dirangkai dengan luasan yang sama sebesar 14x19 cm2. Wadah pemeliharaan berupa wadah plastik dengan volume air laut 10 L, kepadatan awal 1.000 individu/wadah. Perhitungan akhir STS dilakukan pada hari ke-13 masa pemeliharaan. Hasil penelitian menunjukkan bahwa jenis substrat yang berbeda memberikan pengaruh yang berbeda nyata terhadap STS larva teripang pasir. Substrat E. acoroides memberikan sintasan tertinggi sebesar 15,53±2,23%. STS perlakuan substrat Ulva sp. sebesar 5,07±0,74%, perlakuan E. cottonii sebesar 2,57±0,25%, dan perlakuan G. gigas sebesar 1,96±0,17%. Kata kunci: penempelan, Holothuria scabra, substrat, lamun, makroalga

Abstract

Techniques for producing Holothuria scabra or sandfish sea cucumbers from aquaculture have been developed rapidly in the recent years to address the issues of decreasing sandfish population and increasing market demand. During the process of culturing H. scabra, the early stages of sandfish (i.e. larvae and juveniles) are the most critical of the development. Therefore, the Research and Development Division of the Marine Bio Industry – LIPI conducted a study to assess the growth performance of juvenile H. scabra  ultured in four different rearing conditions: (i) indoor concrete tank, (ii) indoor fiberglass tank, (iii) outdoor concrete tank with 1 mm hapa mesh, and (iv) outdoor concrete tank with 3 mm hapa mesh. Each of the rearing condition was set with an initial juvenile density of 15 juveniles per tank with a mean initial weight of 14 g, and fed ad libitum with a mixed feed consisting of Enhalus acoroides sea grass, Pennisetum purpureum napier and cow manure. The experiments were conducted for 20 weeks between April 26 - September 23, 2013. After the experimental period, juveniles reared in the outdoor concrete tank with 1 mm mesh show the best performance with respect to their survival rate (48.89 ± 3.85 %),mean final weight (22.07 ± 1.01 g), final biomass (162.05 ± 18.17 g) and specific growth rate (0.15 ± 0.03% per day). Further studies that combine rearing conditions with different bottom substrates as well as investigating the effect of light are needed to optimize the production of H. scabra from aquaculture.

Abstract

Holothuria scabra is a commercial tropical sea cucumber species, which has been exploited severely in recent years. This study was conducted at the Mataram Marine Bio Industry Technical Implementation Unit for 20 days aiming to determine the relationship between wet and dry weight of sea cucumbers with and without colon. Results of regression analysis between wet and dry weight show a significant positive correlation. The relationship between wet and dry weight without the colon is y = 0.1685x – 0.2319 (R2 = 0.9236) whereas the wet and dry wet of the intestine is y = 0.075x + 0033 (R2 = 0.791).

Abstract

Sea cucumbers belonged to the species of Holothuria atra are one of the holothurians that could reproduce asexually through fission particularly when their ambient environments are not optimum to reproduce sexually. The reproductive pattern of H. atra has been shown to correlate with to the condition of H. atra population as well as their habitats and environments. This study aims to investigate the asexual reproduction and fission rate of H. atra in Tanjung, North Lombok, Indonesia. We conducted surveys in February, May, July, August, September and December 2016 by applying the free collection method. A total of 1,383 individual H. atra were collected consisting of 931 intact or normal individuals, 174 anterior individuals, 261 posterior individuals and 17 uncategorized individuals. Fissioned H. atra were found in every sampling conducted in the six different months with fission rates range from 5.53 to 30.40%. This research shows that the population of H. atra in Tanjung, North Lombok, are susceptible to asexual reproduction (fission). Fission rate value of > 5% is associated with an adaptation mechanism of H. atra to maintain their population in the wild. Further investigation on the environmental conditions stimulating H. atra fission is needed to understand the reproduction and population dynamic of this species in Tanjung waters.

Abstract

Teripang pasir, Holothuria scabra merupakan komoditas hasil laut yang bernilai ekonomis tinggi. Penangkapan berlebihan stok di alam mendorong berkembangnya kegiatan budidaya. Penempelan merupakan fase kritis pada larva teripang karena terjadi peralihan sifat planktonis ke bentik yang memerlukan substrat. Penelitian ini bertujuan untuk mengetahui preferensi penempelan, pertumbuhan, dan sintasan larva H. scabra fase penempelan pada substrat lamun yang berbeda. Penelitian menggunakan metode rancangan acak lengkap dengan perlakuan empat jenis daun lamun berbeda dan lima ulangan. Perlakuan terdiri atas Enhalus acoroides (L-1), Syringodium isoetifolium (L-2), Cymodocea serrulata (L-3), dan Cymodocea rotundata (L-4). Jumlah awal larva sebanyak 1.000 individu dan substrat dirangkai dengan luasan yang sama sebesar 12 cm x 17 cm untuk setiap unit penelitian. Hasil penelitian menunjukkan jenis lamun yang digunakan sebagai substrat berpengaruh secara nyata terhadap preferensi penempelan dan sintasan larva teripang pasir, namun tidak berpengaruh terhadap pertumbuhan. E. acoroides menunjukkan hasil terbaik dengan preferensi penempelan 0,26 ind. cm-2 dan sintasan 10,66%; sehingga layak digunakan sebagai substrat penempelan dalam pembenihan teripang pasir, H. scabra.Holothuria scabra larvae on different seagrass substrates. Overfishing of natural stocks has compelled an interest to begin aquaculture practice. Settlement is a critical phase for the planktonic larvae as they will transform to benthic form in the presence of substrate. This study aims to evaluate the settlement preferences, growth, and survival rate of H. scabra larvae settled on different seagrass leaves. The research was conducted using the Completely randomized design with four different species of seagrass leaves and five replications. The treatments consist of Enhalus acoroides (L-1), Syringodium isoetifolium (L-2), Cymodocea serrulata (L-3), and Cymodocea rotundata (L-4). Initial number of larvae was 1,000 individuals and the substrate was set with same widthof 12 cm x 17 cm for each unit. Results of the experiment indicated that settlement preference and survival rate of H. scabra larvae was significantly affected by seagrass used as substrate while no significantly differences was observed for growth of larvae. E. acoroides showed the best result with 0.26 ind. cm-2 settelement preference and 10.66% survival rate, so that suitable to be used as settlement substrate in H. scabra hatchery.
