Abstract

Penelitian ini bertujuan mengamati perbandingan teknik kastrasi pada caudal dan vas deferent terhadap bobot badan, panjang dan volume skrotum pada sapi Bali. Penelitian ini dilaksanakan pada bulan Juni sampai Agustus 2016 dilaksanakan dikandang percobaan Jurusan Peternakan, Fakultas Pertanian, Universitas Sriwijaya. Metode penelitian menggunakan 6 ekor sapi Bali dibagi menjadi  dua  teknik,  Pertama  3  ekor  sapi   yang  dikastrasi  dengan  teknik pemotongan pada  caudal (P0) dan kedua 3  ekor sapi  yang dikastrasi dengan teknik pemotongan pada vas deferent (P1) metode statistik yang digunakan adalah uji  T-Test.  Hasil  penelitian  menunjukan  adanya  perbedaan  bobot  badan  pada kedua metode teknik kastrasi pada caudal maupun pada vas deferent, sedangkan pada panjang dan volume skrotum tidak memberikan perbedaan yang secara signifikan terhadap perlakuan kedua metode kastrasi baik pada caudal maupun vas deferent.

Abstract

The purpose of this study was to investigate the relationship between spermatozoa DNA damages with spermatozoa protein profiles of after freezing. The rate of spermatozoa DNA damages was measured by Sperm-Bos-Halomax from two Brahman bull straw samples and the spermatozoa protein was isolated by separating the upper and lower fractions of the centrifugation results. The protein profiles were then analyzed using SDS PAGE with a 12.5% separating gel concentration. The data obtained were analyzed descriptively by comparing level of spermatozoa DNA damages with spermatozoa protein profiles of upper and lower fractions. The results of the analysis showed that in the upper fractions at 37.11% level of spermatozoa damages, one protein band (29 kDa) and at 10.66% level of DNA spermatozoa damages 9 protein bands (128 kDa, 110, 91, 55, 44, 29, 27, 25 and 20 kDa) were found, respectively. Meanwhile, in the lower fractions of frozen semen, at 37.11% level of spermatozoa DNA damages 4 protein bands (105, 82, 56 and 25 kDa), and at 10.66% level of spermatozoa DNA damages 8 protein bands (109, 95, 79, 69, 50, 44, 24 and 18 kDa) were found, respectively. It can be concluded that there are differences in the spermatozoa protein profiles between different levels of spermatozoa damages.

Abstract

The aim of this research was to study the effects on different supplementation legumes in the digestibility of fermented kumpai grass (Hymenachne acutigluma) based rations through in vitro technique. This study was conducted in Animal Feed and Nutrition Laboratory of Agriculture Faculty, Sriwijaya University. This study was done in 3 months. A completely randomized design with four treatments and four replicates was used in this study. The treatments were R0= 70% fermented kumpai grass + 30% concentrate + 0% legume, R1= 55% fermented kumpai grass + 7,5% lamtoro leaves + 7,5% water mimmosa + 30% concentrate, R2= 55% fermented kumpai grass + 7,5% acacia leaves + 30% concentrate , and R3=55% fermented kumpai grass + 5% lamtoro leaves + 5% acacia leaves + 5 % water mimmosa + 30% concentrate. Variables measured were dry matter digestibility, organic matter digestibility, volatile fatty acid (VFA), N-ammonia and pH. The result indicated that The adding of different legumes in the ration significantly (P<0.05) affected the dry matter digestibility, organic matter digestibility, N-ammonia and VFA. Duncan Multirange Range Test showed that treatment of control (R0) had the highest digestibility in dry matter (65,88%) and organic matter (65,34 %). The highest content of N-Ammonia, VFA was obtained in the treatment of adding lamtoro, acacia and water mimmosa (R3), namely 11 mM N-ammonia, and 158 mM VFA. It was concluded that the treatment R3 with adding lamtoro, acacia and water mimosa had the lowest digestibility but increased the ammonia and VFA of ration. ABSTRAK Tujuan penelitian ini adalah untuk mempelajari pengaruh suplementasi legum yang berbeda pada ransum berbasis rumput kumpai terhadap nilai kecernaan in vitro. Penelitian dilaksanakan selama tiga bulan di Laboratorium Nutrisi dan Makanan Ternak Fakultas Pertanian, Universitas Sriwijaya. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan empat perlakuan dan empat ulangan. Masing-masing perlakuan adalah Perlakuan R0 = Ransum Kontrol (70% rumput kumpai fermentasi + 30% Konsentrat + 0% leguminosa (kontrol), R1 = 55% rumput kumpai fermentasi + 7,5% lamtoro + 7,5% kemon air + 30 % Konsentrat, R2 = 55% rumput kumpai fermentasi + 7,5% daun akasia + 7,5% kemon air + 30 % Konsentrat, R3 = 55% rumput kumpai fermentasi + 5% lamtoro + 5% kemon air + 5% daun akasia + 30 % konsentrat. Peubah yang diamati adalah koefisien cerna bahan kering (KCBK), koefisien cerna bahan organik (KCBO), N-Amonia,Volatile Fatty Acid (VFA), dan pH. Hasil penelitian memperlihatkan bahwa perlakuan berpengaruh nyata (P<0,05) terhadap KCBK, KCBO), N-Amonia dan VFA, sedangkan pH tidak nyata (P>0,05). Hasil uji lanjut menunjukkan bahwa perlakuan R0 memiliki nilai kecernaan bahan kering dan bahan organik tertinggi; KCBK 65,88 %, KCBO = 65,34 %. Kandungan N-Amonia dan VFA tertinggi terdapat pada perlakuan R3 yaitu N-Amonia: 11,00 mM dan VFA: 158 mM. Simpulan dari penelitian ini adalah penambahan kombinasi daun lamtoro, kemon air, dan akasia dapat menurunkan nilai KCBK, KCBO, sedangkan kadar N-Amonia dan VFA terjadi peningkatan.

Abstract

The relationship among of sperm DNA damage in cows with pregnancy rates has not been widely studied. The purpose of this study to determine the relationship of sperm DNA damage with pregnancy rate on Brahman cows. The sperm DNA damage rate was measured by Sperm-BosHalomax® from 2 samples of male Brahman bull straw (40002 and 40885) and pregnancy rate was measured from the success rate of artificial insemination. In 14 female Brahman cows divided into two groups. One group of 7 in the artificial insemination with 40002 males with 37.11% sperm DNA damage and one in artificial insemination with 40885 with 10.65% sperm DNA damage. The data obtained were analyzed descriptively by comparing sperm DNA damage with pregnancy rate. The results showed that at 37.11% sperm DNA damage level was found pregnancy rate 57.11% with ultrasound on 30 day and pregnancy rate 42.80% with ultrasound to 45 day. Result of research on sperm DNA damage level of 10.66% found pregnancy rate 57.11% with ultrasound to 30 day and level pregnancy 57.11% with ultrasound 45 days. The results of this study have concluded that there is a difference in the rate of sperm DNA damage with pregnancy rate in Brahman cows. The sperm DNA damage has an effect on pregnancy rate on Brahman cows.

Abstract

ABSTRAKHubungan antara tingkat kerusakan DNA sperma dengan keguguran pada sapi belum banyak dilaporkan. Tujuan penelitian ini untuk mengetahui hubungan antara kerusakan DNA sperma dengan tingkat keguguran pada sapi Brahman dan sapi Bali, tingkat keguguran pada sapi Brahman dan sapi Bali belum banyak dilaporkan. Tingkat kerusakan DNA sperma diukur dengan Sperm-Bos-Halomax® dari dua straw sampel semen beku sapi Brahman (40002,40885) dan tingkat kebuntingan diukur dari tingkat keberhasilan inseminasi buatan pada 14 ekor sapi Brahman dan 14 ekor sapi Bali yang masing-masing dibagi menjadi dua kelompok. Satu kelompok sebanyak tujuh ekor diinseminasi buatan dengan semen pejantan 40002 dengan tingkat kerusakan DNA sperma 37,11 % dan satu kelompok lagi diinseminasi buatan dengan semen pejantan 40885 dengan tingkat kerusakan DNA sperma 10,66%. Data yang diperoleh dianalisis secara deskriptif dengan membandingkan kerusakan DNA sperma dengan tingkat keguguran. Hasil penelitian menunjukkan bahwa pada tingkat kerusakan DNA sperma 37,11% ditemukan tingkat keguguran14,31 % pada sapi Brahman dan 28,60 % pada sapi Bali, dengan pemeriksaan ultrasonografi (USG) pada hari ke-30 dan hari ke-45. Hasil penelitian pada tingkat kerusakan DNA sperma 10,66% tidak ditemukan keguguran pada pemeriksaan USG pada hari ke-30 dan USG pada hari ke-45. Berdasarkan hasil penelitian yang dilakukan dapat disimpulkan bahwa kerusakan DNA sperma 37,11 % memengaruhi keguguran pada sapi Brahman dan sapi Bali. Kerusakan DNA sperma 10,66 % tidak berpengaruh terhadap keguguran pada sapi Brahman dan sapi Bali.

Abstract

Sperm DNA integrity is very important in the success of fertilization and embryo development.Toluidine blue (TB) is a sensitive staining to examine the chromatin structure of sperm. The aim of thisstudy was to detect the sperm DNA damage before and after freezing using TB. Semen was obtained fromeight superior bulls (two Brahman, two Ongole, two Simental and two Limosin) belong to Lembang ArtificialInsemination Centre. Semen was collected twice a week using artificial vagina, then was evaluatedmacrocopically and microscopically after collected, including sperm motility, viability, plasma membraneintegrity (MI) and acrosome intact (AI), sperm concentration, abnormality and sperm DNA integrity. Thesemen that been used in this study showed the total motility was more than 70%, sperm concentrationwas more than 1000x106, and the sperm abnormality was below 20%. The result showed that the qualityof semen after freezing processed was decreased significantly (P<0.05) on percentage of sperm motility,viability, MI and AI, whereas there was no different on DNA integrity (P>0.05). Sperm DNA integrity offresh and frozen semen were 93.91±4.77% and 92.06 ±2.41% respectively. The decrease of DNA integritywas low (1.84%) compared to motility (28.3%), viability (21.6%), MI (14.1%), and AI (11.8%). In conclusion,toluidine blue can be used to detect of DNA damage and the freezing process will not decrease the DNAintegrity.

Abstract

This research was done to identifiy Brahman cross (BX) cow as candidate of twinning embryo transfer recipient. However, BX cow oftenly reported low in reproduction performance especially from cattle farmer. The conditionwas assumed related to its management and feeding system. Body score condition (BCS) was one of quality parameters in cow. Therefore, enhancement of BCS through management improvement and probiotic BIOPLUS suplementation on BX cows would increase quality of twinning embryo transfer recipient candidate.Identification of production system, body score condition and reproduction organ of twenty BX cow were held in SidomulyoVillage, Banyuasin I District and Banyuasin Regency. Improvement on management production and suplementation of Probiotic BIOPLUS were done for 2 months. The result shows that only 13 of 20 cows were fulfill the requirement of twinning embryo transfer recipient candidate with body score condition were 3-4, 21-24 days of estrous cycle and improvement on reproduction organ performance.Penelitian ini bertujuan untuk mengidentifikasi calon induk resipien transfer embrio kembar. Namun, induk sapi BX sering kali dilaporkan memiliki performans reproduksi yang rendah, terutama yang dipelihara oleh peternak rakyat. Hal ini diduga berkaitan dengan system pemeliharaan dan pemberian pakan. Nilai Kondisi Tubuh (NKT) adalah salah satu parameter kualitas induk. Peningkatan NKT melalui perbaikan manajemen dan suplementasi probiotik BIOPLUS pada induk BX diharapkan akan meningkatkan kualitas calon induk resipien transfer embrio kembar. Dua puluh ekor induk BX yang ada di Desa Sidomulyo, Kecamatan Banyuasin I Kabupaten Banyuasin diidentifikasi sistem produksi, NKT dan organ reproduksinya. Perlakuan perbaikan manajemen produksi dan suplementasi probiotik BIOPLUS dilaksanakan selama 2 bulan. Hasil penelitian menunjukkan bahwa hanya 13 dari 20 induk BX yang memenuhi persyaratan sebagai calon resipien transfer embrio kembar dengan rataan NKT 3-4, siklus berahi 21- 24 hari dan perbaikan pada kesehatan organ reproduksi.Keywords: BX cow, twinning cattle, embryo transfer, probiotic.
