Abstract

Parasit yang sering menyerang ikan koi adalah Myxobolus sp. Ikan yang terinfeksi Myxobolus sp. akan mengalami gejala klinis berupa nodul kemerahan dan pembengkakan pada organ insang. Penelitian bertujuan untuk mengetahui pengaruh pemberian deltamethrin terhadap ekspresi CD4 pada ikan koi (Cyprinus carpio) yang terinfeksi Myxobolus sp. Pada penelitian ini dibagi menjadi 4 perlakuan (A) Kontrol, perlakuan (B) ikan koi terinfeksi Myxobolus sp., perlakuan (C) ikan koi terinfeksi Myxobolus sp. dengan pemberian deltamethrin dosis 0,5 µl/g dan perlakuan (D) ikan koi terinfeksi Myxobolus sp. dengan pemberian deltamethrin dosis 1 µl/g. Kemudian dilakukan pengamatan eritrosit, leukosit dan immunohistokimia untuk mengetahui ekspresi CD4 pada usus. Berdasarkan hasil penelitian didapatkan eritrosit pada perlakuan (A) sebesar 500.000 sel/mm3, (B) sebesar 1.850.000 sel/mm3, (C) sebesar 1.110.000 sel/mm3 dan (D) sebesar 1.670.000 sel/mm3. Hasil pengamatan leukosit perlakuan (A) didapatkan sebesar 121.600 sel/mm3, (B) sebesar 333.550 sel/mm3, (C) sebesar 265.770 sel/mm3 dan (D) sebesar 145.620 sel/mm3. Berdasarkan hasil Imunohistokimia  didapatkan hasil pada perlakuan (A) nilai DAB sebesar 15,0%, (B) DAB sebesar 30,2%, (C) DAB sebesar 24,3% dan (D) DAB sebesar 19,2 %. Dari penelitian ini dapat disimpulkan bahwa pemberian deltamethrin berpengaruh terhadap eritrosit dan leukosit serta dapat menurunkan ekspresi CD4 sebagai respon imun pada ikan koi (Cyprinus carpio) yang terinfeksi Myxobolus sp. Parasites that often attack koi are Myxobolus sp. Fish infected with Myxobolus sp. will experience clinical symptoms of red nodules and swelling in the gill organs. The study aimed to determine the effect of deltamethrin administration on CD4 expression in koi fish (Cyprinus carpio) infected with Myxobolus sp. In this study divided into 4 treatments (A) Control, treatment (B) koi fish infected with Myxobolus sp., Treatment (C) koi fish infected with Myxobolus sp. by treatment a dose of 0.5 µl / g deltamethrin and treatment (D) koi fish infected with Myxobolus sp. with the treatment of 1 µl / g of deltamethrin. Then erythrocytes, leukocytes and immunohistochemistry were observed to determine CD4 expression in the intestine. Based on the results of the study obtained erythrocytes in treatment (A) of 500,000 cells / mm3, (B) of 1,850,000 cells / mm3, (C) of 1,110,000 cells / mm3 and (D) of 1,670,000 cells / mm3. Observation of leukocyte treatment (A) was obtained at 121,600 cells / mm3, (B) of 333,550 cells / mm3, (C) of 265,770 cells / mm3 and (D) of 145,620 cells / mm3. Based on the results of immunohistochemistry the results obtained in treatment (A) DAB value of 15.0%, (B) DAB of 30.2%, (C) DAB of 24.3% and (D) DAB of 19.2%. From this study it can be concluded that the administration of deltamethrin affects erythrocytes and leukocytes and can reduce CD4 expression as an immune response in koi fish (Cyprinus carpio) infected with Myxobolus sp.

Abstract

This research has the objectives to determine LC50 96 hours value and observation toward hematology, gills and kidney histology of goldfish (Cyprinus carpio) exposed to organophosphate pesticide with Poksim active compound (trade brand FOKKER 500 EC). Method used in this paper is experiment to determine LC50 96 hour value with probit analysis. Descriptive method was used for gills and kidney tissues microanatomy observation with hematoxilin eosin (HE) coloring and hematology. Toxicity result of Fokker 500 EC pesticide toward goldfish obtained LC50 96 hours with 41,7 ppm. Histological result showed that increasing exposure doses in real test had caused increase gills and kidney tissues damage. Hematology observation result during research obtained that along with the increasing exposure doses of pesticide in the real test, acute toxicity test would caused reduction in total erythrocyte, leukocyte and hemoglobin of goldfish. Result also showed that Fokker pesticide usage was allowed until 1,8 ppm dose based on histology and hematology evaluation. Keywords: acute toxicity, goldfish, histology, hematology, pesticide

Abstract

Kijing Taiwan (Anodonta woodiana) is one type of shellfish that live in freshwater, are filter feeders, have high endurance life, and exist in abundant number making Kijing Taiwan can be used as indicators of water pollution that can assist in purifying water. The study is descriptive qualitative, and data is obtained by measuring public perceptions using Likert scale. The later stage begins with determining the respondents. Respondents consist of 27 people from 76 citizen members of the UPR Source Mina fish seed producers and are over 30 years old. The results show that people tend to behave negatively towards Kijing Taiwan by removing and exterminating Kijing Taiwan en masse, as it has been assumed to reduce the cultivation of fish seed. They do not have enough knowledge about the role of Kijing Taiwan leading them to have the wrong perception on the role of Kijing Taiwan in aquaculture ponds. Formulating the strategic management of freshwater aquaculture waters must be done by proposing stakeholders to conduct public education and to convey the results of research on Kijing Taiwan, among other on the role of Kijing Taiwan to help early detection of pollution and as water purifier in aquaculture ponds naturally. Keywords: Benefits of Kijing Taiwan, Public Perception

Abstract

Centella asiatica is a herbal medicine has been used for many studies in some animals as antioxidant properties. This study was aimed to enhance the level of SOD in Cyprinus carpio after oral administration of C. asiatica extract. In the present study, phytochemical screening of bioactive constituents, antioxidant assay, and their effect on SOD in C. carpio liver were investigated. C. asiatica extract was obtained by maseration for 24 h using ethanol as a solvent. C. asiatica extract with 50, 100, 150 and 300 mg kg-1 body weight were administered every 3 days for two weeks. On the last day of experiment, fishes were killed and liver were removed from the body. The results showed that C. asiatica extract had some bioactive compounds such as flavonoid, alkaloid, terpenoid, tanin, saponin and DPPH scavenging activity with an IC50 value of 125 μg mL-1. C. asiatica extract also enhanced the level of SOD in fish liver. The SOD level was significantly difference compared to the control group with P<005. These results indicate that the ethanolic extract of C. asiatica had a potential antioxidant properties by scavenging free radical and enhancing the SOD level as antioxidant deffense inside the body.

Abstract

Merkuri digunakan sebagai pengikat emas dan dilepaskan ke lingkungan perairan yang menyebabkan pencemaran. Konsentrasi merkuri pada limbah pencucian emas harus diminimalkan sehingga tidak berbahaya bagi perairan. Salah satu caranya adalah dengan mengendapkan limbah. Penelitian ini bertujuan untuk mendeteksi konsentrasi merkuri dan kualitas air pada setiap kolam penampungan secelum limbah dibuang ke perairan. Kolam penampungan limbah sebanyak enam kolam (P1, P2, P3, P4, P5, P6) dengan satu kolam air bersih (P0). Merkuri dideteksi  menggunakan  Atomic Absorption Spectrophotometry (AAS),  kualitas air yang diukur adalah suhu, pH, dan warna air limbah. Data hasil penelitian dianalisis secara deskriptif. Konsentrasi merkuri di semua kolam pengendapan antara 0,021 ± 0,002 mg / L hingga 0,051 ± 0,005 mg / L). Penurunan rata-rata konsentrasi merkuri dari kolam P1 ke P6 adalah 16,7%. Konsentrasi merkuri telah melebihi batas maksimum yang diperbolehkan untuk merkuri di air. Suhu pada setiap kolam mengalami penurunan dari kolam P1 sampai P6, namun pH meningkat.

Abstract

Human and industrial activities in the area of Manyar Subdistrict Mangrove Waters, Gresik District can increase pollution and the presence of heavy metals in aquatic environments and aquatic organisms. This triggers input of heavy metals, especially Pb and Cu. Mangroves can accumulate and have a high tolerance for heavy metals so they can be used as plants for phytoremidiation purposes (phytostabilization). The mangrove ecosystem can be used as a control of heavy metal pollution and can also be used as a pollutant trap. This study was conducted to determine the potential of phytoremidiation based on the accumulation and translocation of heavy metals in Avicennia marina mangroves in absorbing heavy metals Pb and Cu from their environment. Based on research results MacFarlane et al., (2007) mangrove Avicennia marina is one of the plants that can effectively accumulate heavy metals due to its root system. The sample was analyzed using AAS (Atomic Absorption Spectrophotometer). The research parameters include levels of Pb and Cu metals in sediments, mangrove roots and leaves. The results of Pb heavy metal concentrations ranged from 0.30 ppm - 4.84 ppm and Cu ranged from 0.27 ppm - 11.42 ppm with the highest values found in sediment stations 2. Avicennia marina at the study site was excluder because the BCF value < 1 which ranged from 0.02 - 0.18 ppm and phyto-extraction because the value of TF > 1 ranged from 0.35 to 1.69 ppm. The results of this study indicate that Avicennia marina mangroves can be developed into phytoremidiation agents because they are able to absorb and transfer heavy metals from the environment to other body tissues.
