Abstract

Callus induction determines the successfulness of micropropagation through somatic embryogenesis. This research was conducted to determine the effect of 2,4-D and BAP on callus induction of peanut (Kelinci strain). Callus induction was done on MS medium, with the observed variables were of initiation time, diameter, texture and color callus. Statistical analysis showed that 2,4-D and BAP affect the initiation time, diameter, texture and color callus. The earliest initiation of callus formation was obtained from MS medium with the addition of 4 mg/L 2,4-D. The diameter of the largest callus obtained from MS medium with the addition of 3 mg/L 2,4-D + 1 mg/L BAP. Callus color were affected by the treatment of 2,4-D and BAP, while callus texture was not affected by these treatmen, in which compact callus texture was not affected by these treatment, in which compact callus texture was observed in all treatments.Keywords: Induction of callus, 2,4-D, BAP, peanuts

Abstract

The objective of this research was to test antibacterial activity ethanol and ethyl acetate of lime peel (Citrus aurantifolia Swingle) extract by Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) against the growth of clinical isolate bacteria. The type of this study used laboratory experimental using sensitivity test, dilution of tube and spreading plate. The experimental design used Complete Random Design using three repetitions on three concentrations i.g 25, 50, and 75%. Data was analyzed by qualitative with measuring inhibit diameter zone bacterial and qualitative using Two Way Anova followed by BNT with α= 5%. The statistical analysis showed that the solvent factor and concentration factor was significantly against growing bacterial and there was an interaction affect of both factors on S. aureus growth. The result presented that the ethanol and ethyl acetate of lime peel extract strongly inhibited Staphylococcus aureus, Staphylococcus epidermidis, and Pseudomonas aeruginosa; and midly inhibited the growth of Klebsiella pneumoniae. The MIC ethanol and ethyl acetate of lime peel extract against S. aureus, S. epidermidis and P. aeruginosa was 25% and K. pneumoniae was 50%. The MBC showed that ethanol and ethyl acetate of lime peel  extract killed bacteria P. aeruginosa at 15%, S. aureus, S. epidermidis at 20% and K. pneumoniae at 25%. Taken together, it can be concluded that ethyl acetate of lime extract more effective than that of ethanol extract was to inhibit and killed clinical isolate bacterial.Keywords: lime peel (Citrus aurantifolia Swingle), antibacterial activity, clinical isolate bacteria, MIC and MBC.

Abstract

The objective of this reasearch was to determine the ornamental plants Lidah mertua (Sanseviera trifasciata.sp), Puring (Codiaeum variegatum), Palem Kuning (Chrysalidocarpus lutecsens) ability to absorb lead (Pb) concentration. This research is descriftive explorative research design with completely randomized design with 9 times treatments and 3 repetitions. The independent variables were lidah mertua (Sanseviera trifasciata.sp) Puring (Codiaeum variegatum), Palem Kuning (Chrysalidocarpus lutecsens), and the subjec variable is measurement of lead (Pb) concentration in the leaves. Data analysis techniques using qualitative and quantitative analysis. Quantitative data was obtained by calculating the lead (Pb) concentration in ornamental plants such as: Lidah mertua (Sanseviera trifasciata.sp) Puring (Codiaeum variegatum), Palem Kuning (Chrysalidocarpus lutecsens) by using AAS (Atomic Absorption Spectrophotometer) test.  Based on reasearch finding, there several finding of oranamental plants ability in absorb lead (Pb): lidah mertua (Sanseviera trifasciata.sp ): 0,06 ppm; 0,1 ppm; 0,15 ppm, puring (Codiaeum variegatum):  0,04 ppm; 0,09 ppm; 0,2 ppm and palem kuning (Chrysalidocarpus lutecsens): 0,02 ppm; 0,04 ppm; 0,11 ppm. Statistical test states that the type of plant and the traffic density does not affect the ability of plants to absorb lead (Pb).Keywords: Lead (Pb), lidah mertua (Sanseviera trifasciata.sp), Puring (Codiaeum variegatum), Palem Kuning (Chrysalidocarpus lutecsens).

Abstract

This research is aimed at determining the effect of POGIL model teaching on the understanding of Natural Science Concept, Skill of Sciences Process, and Critical Thinking Capability of student in photosynthesis. This research is a experiment with nonequivalent control group design. The sample were selected randomly and the hypothesis was analized with Manova and t-test. The rsearch concludes that (1) there an effect of POGIL model teaching in understanding Natural Sciences concept (2) there an effect of POGIL model teaching on Skill of Sciences Process (3) there an effect of POGIL model teaching on the capability of critical thinking (4) there is simultaneous effect of the application of POGIL model teaching on the Natural Sciences concept, Skill of Sciences Process and in the capability of critical thinking.Keywords: POGIL, understanding Natural Sciences concept, Skill of Sciences Process and the capability of critical thinking

Abstract

Production of chrysant flowers (Chrysanthemum morifolium) is halted due to seedling supplay, that it is necessary to produce seedling on tissue culture. This study aims to determine the effect of BAP and 2.4 D for callus formation of chrysanthemum. The research using completely randomized design (CRD). The treatment was consistsed of two factors. BAP (0,0 mg/l, 1,0 mg/l, 1.5 mg/l and 2,0 mg/l) and concentration of 2,4 D (0,0 mg/l, 2,0 mg/l, 3,0 mg/l, 4,0 mg/l). Dependent variables measured were days of callus occurrence, callus colour and texture of callus. Result showed that growth regulators BAP and 2.4 D influenced the timing of the callus formation. Most rapid callus formation appeared at concentration of 2,0 mg/ l 2,4 D and longest callus formation appeared at concentrations of 0,0 mg/l 2,4 D. Callus did not appear on treatment of 0,0 mg/l BAP. The combination of BAP and 2.4 D did not influence on colour and texture of callus. Highest callus score was obtained in the treatment combination of D0B1, D0B0,5 and D0B2. The lowest callus score obtained in the treatment D4B0.Keyword: BAP, 2,4 D, Chrysanthemum morifolium, explants, Callus.

Abstract

The research aim to identify the mechanical properties of  Areca nut fiber composite.. Pinangs fiber composite board uses polyester resin as a binder. Areca nut fiber that used in this research is fiber obtained from pinang which is old and not used anymore. The sample was made by varying the concentration of fiber used (30%, 40%, 50% 60% and 70%). From the results of tests conducted, there are differences in mechanical properties of pine fiber composite material on tensile test and press tests performed. In tensile test, composite of pinang fiber with 50% fiber concentration is the fiber with the best mechanical properties because it has the highest tensile strength, maximum length and maximum load, that is 16.35 MPa, 2.06 mm and 3147.45 N. Meanwhile, Composite betel fiber with a concentration of 30% has the highest broken modulus (MOR) at 30.43 kg / cm2, and elastic module of 1980.2 MPa

Abstract

This is aimed todeterminethe effect of 2,4-D and BAP onshoot regeneration of peanut (Arachis hypogaea L.) Kelinci variety. This research was conducted in tissue culture laboratory of BBI-PPH Narmada. Competely randomized design (CDR) was applied on this rescarch which involved 2 factorial concentration of (0-4 mg/L) 2,4-D and (0-2 mg/L) BAP for both shoot regeneration on MS medium speed of the emerging shoots, number of shoots, theroots appear, and the amount ofroots. Statistical analysis showed that 2,4-D and BAP affect the speed of the emerging shoots, number of shoots, rootsand the number of days appear roots, and there is the effect of the combination of 2,4-D and BAP. Fastest emerging shoots obtained on MS medium with the addition of 2mg/LBAP with an average of 90.33days and the highest number of shoots contained on MS medium with the addition of 1mg/LBAP with an average of 4.33 shoots. While the earliest root formation was found on MS medium with the addition of 1mg/L2,4-D.

Abstract

ABSTRAKTrehalose/T6P berperan penting bagi tahapan kehidupan tanaman dari tahap embryo, pertumbuahn vegetative, pertumbuhan generatif, hingga sanesensi melalui keterlibatannya dalam menentukan pembelahan sel, bentuk sel, arsitektur tanaman dan respon terhadap stress biotik dan abiotik. Mekanisme trehalose/T6P dalam peran tersebut melibatkan SnRK1 dan bZIP11 dengan mengintegrasikan antara ketersediaan dan penggunaan sumber karbon yang mempengaruhi pemrograman ekspresi gen-gen yang trerkait dalam anabolisme dan katabolisme serta gen-gen yang terlibat dalam respon tanaman terhadap stres biotik dan abiotik. Selain itu, protein-protein dalam lintasan metabolisme trehalose diduga terlibat juga dalam pemrograman dan aktivitas enzim-enzim target. Dengan mengetahui peran krusial metabolisme trehalose, manusia dapat memanfaatkan lintasan ini untuk kepentingan pertanian.   The Roles of Trehalose Metabolism at Entire Plant Life Abstract Trehalose/T6P has important role at entire plant stage of life starting from embryonic stage, vegetative growth, generative growth and senescence. The roles include cell devision, cell shape, plant architectecture, and plant responses to biotic and abiotic stress. Syncrunizing between energy demand and energy availability is the main mode of the roles through reprogramming of gene expression responsible for anabolism and/or catabolism processes depending on given situation including biotic and abiotic stress. Moreover, proteins of trehalose metabolism are suggested to affect enzyme activity of target proteins. SnRK1 and bZIP11 involve in all those modes. Understanding crusial roles of trehalose metabolism in plant therefore is necessary in order to get benefit from this pathway for agricultural issue.

Peran krusial metabolisme trehalose, manusia dapat memanfaatkan lintasanini untuk kepentingan pertanian.

Abstract

The present study aims at expressing and partially purifying PtpB in active form. To achieve this, Mtb PtpB gene has been cloned into pET30a vector and overexpressed in Escherichia coli BL 21(DE3) under IPTG induction in the form of an inclusion body. Following resolubilization by urea and dialysis, the resulted PtpB has been shown to be active against para-Nitrophenyl phosphate. It is concluded that the resulted PtpB has had been recovered from inclusion body to give the active form of the enzyme, and thus the success in overexpressing PtpB provides the required material to investigate the biochemical properties of the pathogen virulence factor further.
