Abstract

This study was conducted to validate a commercial testosterone enzyme-linked immunosorbent assay (ELISA) kits (DRG EIA-1559) inanalytic and biological manner for measuring serum testosterone concentrations in kacang goats. This study used 18 healthy kacang goats, six bucks ( 2 years), six kids ( 6 months), and six does ( 2 years). Blood samples were collected from jugular vein and prepared as serum. Two validation tests were performed, an analytical validation comprises a parallelism, accuracy, precision and sensitivity and a biological validation by comparing testosterone concentration from bucks, kids, and does. Testosterone concentrations were measured using ELISA technique. Data of analytical validation were analyzed descriptively and test of equality of slope was performed to see the parallelism between samples and standard curves. Analysis of variance (ANOVA) was used for biological validation data. Results of parallelism showed that sample curve was parallel to the standard curve. Accuracy, precision (% CV of intra-and inter-assay) and sensitivity of the assay were 99.65±4.27%, 10%, 15% and 0.083 ng/ml, respectively. Results of biological validation showed that the assay used were accurately measured testosterone which testosterone concentrations in bucks were significantly higher compared to kids and does (P 0.05). In conclusion, a commercial testosterone ELISA kits (DRG EIA-1559) is a reliable assay for measuring serum testosterone concentration in kacang goats. Abstract

The purpose of this research was find out the effect of age and ejaculation frequency on quality of Aceh bull spermatozoa.. This research was conducted at Frozen Semen Prossesing Laboratorium, Saree, Aceh Besar from Januari to April 2013. Six healthy aceh bulls were divided into two groups of age, those were 3.0-4.0 years old; and 4.0-5.0 years old. Three numbers of bull at each group were assumed as the replication. Semen was collected once a week using artificial vagina and collected until third semen ejaculation. After collecting, volume, pH, color, consistency, concentration, motility, mortality, and abnormality of spermatozoa were immediately measured. This data was analyzed by analysis of variance (ANOVA) two-way pattern, followed by Duncant test. The result showed that the evaluation of bull semen at 3.0-4.0 years old and 4.0-5.0 years old affect significantly (P<0.05) on semen volume and spermatozoa concentration and had no contribute significantly on motility, mortality and abnormality of spermatozoa,while ejaculation frequency effect significantly (P<0.05) on semen volume, concentration, and abnormality spermatozoa, but had no effect significantly on motility and mortality (P>0.05). It is concluded that aged and ejaculation frequency effect significantly on quality of aceh bull fresh spermatozoa.Key words: ejaculation frequency, quality of spermatozoa, aceh bull, age

Abstract

The aim of this study was to find out correlation between cat’s welfare and caring management in a number of cats kept at p etshops inBekasi from January to February 2013. This study used a questionnaire with ratings based on five aspects of animal welfare (feeding, healthy,comfortable, fear, and normal behavior) and through direct observation. Data was analyzed using Correlation Test Chi Square with 95%confidence interval. The result showed that there is the corelation between animal welfare and caring management of cats at pet shop in Bekasi,West Java.____________________________________________________________________________________________________________________Key words: animal welfare, cats, pet shops

Abstract

The objective of this research is to know Aceh cattle follicle dynamic under environmental heat stress condition. This research was conducted on December 2012 until August 2013. Animals used in this research were 20 Aceh cattle aged 5-8 years old, weighing of 150-250 kg, and had at least 2 regular cycles. All cattle used were clinically in good body condition score. The cattle were divided into two groups of 10, used in two different period of times (December-January and July-August) to examine the effect of environmental heat to their follicle dynamic. Each group is separated into two different keeping management; one group was kept in pens while the others in the pasture, 5 cattle in each management group. Research timing was based on information acquired from Indrapuri Agency for Meteorology, Climatology, and Geophysics (BMKG) which predicted that extreme weather would last from July to August while December to January would be relatively normal in Aceh region. All cattle?s estrous cycle were synchronized by 5 mg/mL of PGF2? intramuscularly (Lutalyse?, Pharmacia Upjohn Company, Pfizer Inc.). Ultrasonography examination was performed to monitor ovary follicle?s growth and dynamic during one cycle. Days during ovulation marked by standing heat was regarded as Day 0 of estrous cycle. Follicular dynamic examination during estrous cycle on December-January and also on July-August, both penned or pastured cattle showed the follicle waves that was 3 follicle growth waves. The size of follicle growth on first wave (1st DF) on cattle kept in pen on July-August and December-January were relatively similar. The size of follicle growth on first wave for cattle kept in pasture on July-August and December-January were relatively similar. To conclude, Aceh cattle?s follicle dynamic is not change during environment heat stress condition and is not affected by different cattle-keeping management.

Abstract

Penelitian ini bertujuan mengetahui efek injeksi inhibin B terhadap penurunan konsentrasi follicle stimulating hormone (FSH) di dalamserum pada tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 24 ekor tikus putih berjenis kelamin jantan dengan strain Wistar berumur 4 bulan dengan bobot badan 150-200 g. Tikus-tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu KK0, KP1, KP2, dan KP3, masing-masing kelompok terdiri atas 6 ekor. Kelompok KK0 merupakan kelompok kontrol hanya diinjeksi dengan phosphate buffer saline (PBS), sedangkan kelompok KP1, KP2, dan KP3 diinjeksi dengan inhibin B dengan dosis berturut-turut 25, 50, dan 100 pg/ekor. Injeksi inhibin B dilakukan secara intraperitoneum sebanyak 5 kali selama 48 hari dengan interval waktu 12 hari. Injeksi pertama inhibin B dilarutkan dengan0,05 ml PBS dan 0,05 ml Freud?s complete adjuvant (FCA). Injeksi kedua sampai kelima, inhibin B dilarutkan dengan 0,05 ml PBS dan 0,05 ml Freud?s incomplete adjuvant (FICA). Pada hari ke-6 setelah injeksi inhibin B terakhir, tikus dikorbankan secara dislocatio cervicalis,lalu darah dikoleksi langsung dari jantung dan didiamkan hingga didapatkan serum untuk pemeriksaan konsentrasi FSH dengan menggunakan metode enzyme-linked immunosorbent assay (ELISA). Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan konsentrasi FSH secara nyata (P 0,05) bila dibandingkan dengan kelompok kontrol. Berdasarkan hal tersebut, inhibin B berpeluang untuk dikembangkan sebagai kandidat kontrasepsi pria hormon berbasis peptida.

Abstract

The objective of this study was to characterize the morphology of ovary from slaughtered Aceh cow and measuring the concentration of estradiol in the follicular fluid to examine the potential utilizing for in-vitro fertilization (IVF) development in Aceh cattle. In total, 30 pairs of ovaries were collected from 30 Aceh cattle slaughtered at Banda Aceh abattoir. Morphological characteristics of ovaries observed were the size of ovaries, weight of ovaries, number of follicles, the diameter of follicles, corpus luteal (CL), and volume of follicular fluid. The follicular fluid was aspirated using a disposable syringe and analyzed the estradiol concentrations using enzyme-linked immunosorbent assay (ELISA). The follicles were grouped into 3 categories based on the diameter (Ø) of follicles: small follicles (Ø 1 - 5 mm), medium follicles (Ø 5 - 8 mm), and large follicles (Ø 8 mm). The morphological characteristic of ovaries varied between right and left ovaries in which right ovaries showed bigger size and weight compared to the left ovaries (P 0.05), whereas the number of follicles and follicular fluid did not significantly different (P 0.05). The concentrations of estradiol in follicular fluid of large and medium follicles were significantly higher compared to small follicles (P 0.05), whereas the concentration of estradiol between large and medium follicles, did not significant different (P 0.05). In conclusion, the follicles from abattoir can be used as potential oocytes resources to support IVF development for Aceh cattle in the future. The follicular fluid provided from these follicles can be an alternative for in vitro maturation media of oocytes due to containing high estradiol concentrations.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates ?-galactose, ?-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

Penelitian ini bertujuan mengetahui peningkatan level steroid dan persentase kebuntingan sapi aceh terhadap induksi ovulasi dengan metode presynch-ovsynch. Dalam penelitian ini digunakan sepuluh ekor sapi aceh betina dengan status tidak bunting, minimal dua bulan pascapartus, sudah pernah beranak, dan sehat secara klinis. Sapi dibagi atas dua kelompok, yang masing-masing terdiri atas lima ekor sapi. Kelompok pertama (K1) disinkronisasi berahi dengan metode presynch-ovsynch. Pada kelompok kedua (K2), disinkronisasi berahi menggunakan 5 ml PGF2? secara intramuskulus dengan pola penyuntikan ganda dengan interval 12 hari. Setelah  48 jam akhir perlakuan, sapi pada K1 dan K2 diinseminasi menggunakan semen beku fertil. Observasi berahi dilakukan setelah penyuntikan terakhir. Koleksi darah untuk pemeriksaan level estradiol dilakukan segera setelah inseminasi dilakukan sedangkan koleksi darah untuk pemeriksaan progesteron dilakukan pada hari ke-7 pasca-inseminasi. Level steroid diukur menggunakan teknik enzyme-linked immunosorbent assay (ELISA). Pemeriksaan kebuntingan dilakukan 90 hari pasca-inseminasi menggunakan teknik palpasi rektal. Seluruh sapi menunjukkan gejala berahi setelah perlakuan. Level estradiol dan progesteron pada K1 vs K2 masing-masing adalah 294,98±110,48 vs 392,76±11,6 pg/ml (P 0,05) dan 23,85±15,14vs 12,69±5,64ng/ml (P 0,05). Persentase kebuntingan pada K1 vs K2 masing-masing adalah 60,0 vs 0,0%. Dari hasil penelitian disimpulkan bahwa metode presynch-ovsynch tidak dapat meningkatkan level steroid tetapi dapat meningkatkan persentase kebuntingan pada sapi aceh.

Abstract

The experiment was conducted to compare estrous performance between and estradiol level kacang and Nubian goats estrous induction by prostaglandin F2 alpha (PGF2?). Eight female goats were divided into two groups, groups 1 (K1) consisted five Nubian goats and group 2 (K2) included three kacang goats with criterias as follows: age 1.5-3.0 years old, not pregnant, at least two months postpartum, and healthy clinically. All goats were estrous synchronized by double injection 7,5 mg PGF2? intramuscularly with 10 days interval. Goats that appeared estrus symptoms were mated naturally to a male goat. Determination of estrous intensity was performed using scoring method. Blood samples were taken at the first time the goats were mated (day 0). The measurement of estradiol concentration was done by enzyme linked immunosorbent assay (ELISA) method. Data of intensity, onset, duration, and estradiol level were analyzed using t test. The results showed that the rate of intensity, onset, duration, and estradiol level of estrous at KI vs KII were 2.08±0.52 vs 1.85±0.67, 56.00±34.11 vs 40.80±18.20 hours, and 24.00±26.15 vs 33.80±9.10 hours, and 103.51±42.49 vs 67.95±38.44 pg/ml (P>0,05). It can be concluded there was no difference of estrous performance and estradiol level between kacang and Nubian which estrous induction with PGF2?. The relationship between the intensity of estrus estradiol level in the Nubian goats was 0.995 while the kacang goats was 0.890.

Abstract

Spermatoza yang melalui proses pembekuan seringkali mengalami kerusakan karena terbentuknya kristal es. Pembentukan kristal es dapat menyebabkan penurunan kualitas spermatozoa pasca thawing terutama penurunan motilitas. Penambahan krioprotektan dalam pengencer dapat meminimalisir kerusakan sel. Salah satu jenis krioprotektan yang sering digunakan pada pembekuan semen adalah gliserol. Penelitian ini bertujuan untuk melihat pengaruh penambahan gliserol dalam pengencer Tris Kuning Telur (TKT) terhadap kualitas spermatozoa (motilitas, spermatozoa hidup, dan MPU) sapi aceh  yang disimpan dalam nitrogen cair. Desain penelitian menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 3 perlakuan dan 6 pengulangan. Pengamatan kualitas spermatozoa dilakukan setelah pembekuan di dalam nitrogen cair. Data dianalisis dengan uji Analysis of Variance (ANOVA) pola satu arah. Hasil kualitas spermatozoa pada masing-masing konsentrasi gliserol (3%, 5%, dan 7%) berturut-turut untuk hasil motilitas sebesar: 17,17±12,17%, 51,17±11,13%, dan  33,33±8,76%; hasil spermatozoa hidup sebesar: 22,50±15,41%, 56,33±8,98%, dan 42,50±6,35%; hasil MPU sebesar: 31,42±17,90%, 61,50±10,82%, dan 42,50±12,14%. Berdasarkan hasil penelitian dapat disimpulkan bahwa penambahan gliserol dalam pengencer Tris Kuning Telur dapat mempertahankan kualitas spermatozoa (motilitas, spermatozoa hidup, dan MPU) sapi aceh setelah pembekuan. Penambahan gliserol dengan konsentrasi 5% dalam pengencer Tris Kuning Telur paling baik dalam mempertahankan kualitas spermatozoa sapi aceh dibandingkan dengan konsentrasi 3% dan 7% (Freezing spermatozoa are often experiencing the damage by the formation of ice crystal. The formation of ice crystal leads to the quality reduction of post-thawing spermatozoa especially reducing motility. The addition of cryoprotectant in the diluent can minimize cell damage. One of the most common cryoprotectant type that was used in sement freezing is glycerol. This research aims to examine the influence of the addition of glycerol in tris-yolk, or known as Tris Egg Yolk (TKT) to the quality of spermatozoa (motility, live spermatozoa, and MPU) of aceh cattles stored in liquid nitrogen. The research design used in this research is a completely randomized design, that consists of three treatments and six repetitions. Quality of spermatozoa observation were performed after freezing process in the liquid nitrogen. Data were analyzed by Analysis of Variance (ANOVA) with one tail pattern. The result of spermatozoa quality in each concentration of glycerol (3%, 5%, and 7%) respectively for the motitity as follows: 17.17±12.17%; 51.17±11.13%; and  33.33±8.76%; result of live spermatozoa as follows: 22.50±15.41%; 56.33±8.98%; and 42.50±6.35%; result of MPU as follows: 31.42±17.90%; 61.50±10.82%; and 42.50±12.14%. Based on the result, the conclusion is that the addition of glycerol in the tris-yolk can maintain the quality of spermatozoa (motility, live spermatozoa, and MPU) of aceh cattle after freezing. The addition of glycerol with 5% concentration in the tris-yolk dilator is the best concentration to maintain the quality of spermatozoa aceh cattle, compared to the 3% and 7% concentration)
