Abstract

Geography and climate of Palu and Sigi Regency has been markedly suitable for dragon fruit supported by the available land in the areas.  Unfortunately, development of the commodity has been restricted by less available qualified seedlings and unskill farmers.  Dragon fruit seedling production can be achieved through cuttings and tissue culture acclimatized planlets.  This program aimed to transfer the technology on cultivation, seedling production through cuttings and tissue culture planlets of dragon fruit, which is turn can increase the income of the farmers.  The method used to solve the problems justified were:  extension, training and accompaniment on cultivation, seedling production through cuttings and tissue culture planlets acclimatized of dragon fruit.  The extension, training and accompaniment were conducted both in class and on farm.  The results of this program:  The farmers or the program partners understand and become skillful in:  dragon fruit cultivation; producing dragon fruit seedlings through cutting and tissue culture planlets acclimatized.  The farmers got more than 250 qualified seedlings from tissue culture propagation and 200 seedlings obtained from cuttings grown in a nursery, and the farmers have willingness to be nursery man.

Abstract

The development of Central Sulawesi superior commodity,which is Tulo-5, the drough-resistant cultivars, through tissue culture technique can be used as an alternative, in addition to conventional breeding. This study aims to determine the growth of jackfruit shoots in  various concentrations of benzylamino purine in vitro. The research has been conducted at the Laboratory of Plant Biotechnology, Agriculture Faculty of Tadulako University, Palu.  The research began on December 2017 and ended up on February 2018. This research used Completely Randomised Design of one-factor treatment which is BAP concentrations consist of 4 stage which is1,5 ppm BAP, 2,0 ppm BAP, 2,5 ppm BAP, 3,0 ppm BAP. The result showed that 6 weeks after planting, BAP concentration had significant effect on shoots time to emerge and shoots number but no significant effect on leaf number.  The result of Tukey’s HSD test showed that concentration2,5 ppm gave the fastest shoot emergence with an average of 2,875 days after planting, in concentration 2,0 ppm gave the highest number of shoots with an average of 2.125 shoots per explant, and for the highest number of leaves found in the concentration of 2,0 ppm with the average number of leaves is 1 strand per explant.

Abstract

This research aimed to findout efficient sterilization method and MS media to germinate dragonfruit seeds in vitro, conducting at Biotechnology Laboratory, Agriculture Faculty, Tadulako University, using factorial completely randomized design. The first factor was sterilization technique (S), i.e: S1 (The sliced fruit was washed with sterile distilled water three times, then rinsed in Bayclin (household bleach containing 5.25% NaOCl) 5% for 15 minutes, followed by washing in sterile distilled water three times. S2 (The seeds were taken from the fruit meat, then rinsed in 5% Bayclin for 15 minutes, then washed with sterile distilled water three times. S3 (The seeds were taken from the fruit meat, then rinsed in 15% Bayclin for 15 minutes, then washed with sterile distilled water three times. The second factor was the strength of MS media (M), i.e full strength MS (M1) and half strength MS for macro and micro nutrients (M2). Each treatment combination consisted of 35 seeds, and was replicated three times. Germination responses were observed as time to germinate, germination percentage and percentage of opened cotyledone seedlings. All datas were subjected to Analysis of Variance and the mean differences among the treatments were analyzed using Honest Significant Difference (HSD) at the level of 1%. The results showed that removing seed pulp, prior to rinsing the seeds in 15% Bayclin for 15 minutes followed by washing in sterile aquadest three times, and cultured in half MS produce the fastest and highest seed germination of 99.05% with 92.38% opened cotyledon seedlings after 2 weeks in culture.   Key Words: Dragonfruit, germination, in vitro, MS strength, sterilization.
