Abstract

Active Pharmaceutical Ingredients (APIs) are developing rapidly, one of the recent development is cocrystal. Cocrystal is a crystalline structure composed of multicomponent, such as atom, ion, or molecule, that are bonded by non-covalent bond. Cocrystal is solid at ambient temperature in its pure form. Cocrystals are made by combining active ingredients with its coformer using variety of method, like solubilisation as well as grinding. Cocrystalization has proven to alter physicochemical properties of drugs, including solubility, dissolution, stability, compressibility, which also alter its bioavailability without the change on its pharmacological effects.

Abstract

PEMBENTUKAN KOKRISTALANTARAKALSIUM ATORVASTATIN DENGAN ISONIKOTINAMID DAN KARAKTERISASINYA. Penelitian ini bertujuan untuk meningkatkan kelarutan dan laju pelarutan atorvastatin dengan menggunakan metode kokristalisasi. Proses kokristalisasi dilakukan dengan metode solvent evaporation, solvent-drop grinding pada atorvastatin dan koformer (isonikotinamid) masing-masing dengan perbandingan 1:1 (satu mol atorvastatin dengan satu mol isonikotinamid digerus selama 15 menit sambil ditambahkan beberapa tetes metanol. Penambahan methanol berfungsi untuk mempercepat pembentukan kokristal. Kokristal dikarakterisasi menggunakan difraktometer sinar-x,mikroskop polarisasi, Scanning Electron Microscope (SEM), spektrometer infra merah dan Differential Scanning Calorimeter (DSC). Difraktogram dari kokristal menunjukkan intensitas puncak yang lebih rendah dibandingkan atorvastatin standar yang menunjukan telah terbentuk habit kristal baru. Hasil spektrometer infra merah, menunjukkan tidak adanya interaksi kimiawi dan perubahan struktur saat dimodifikasi menjadi kokristal. Thermogram DSC menunjukkan adanya perubahan titik leleh berbeda yang menandai adanya bentuk kristalin baru. Demikian juga pengamatan di bawah mikroskop polarisasi dan SEM menunjukkan bentuk kristal yang relatif baru dibandingkan dengan atorvastatin murninya. Hasil uji kelarutan dan laju pelarutan kokristal atorvastatin-isonikotinamida menunjukkan peningkatan dibandingkan dengan kelarutan dan laju pelarutan atorvastatin sendiri.

Abstract

Gel mata merupakan pengembangan dari sediaan mata konvensional yang sudah ada, seperti salep mata dan tetes mata. Tujuan dari penelitian ini yaitu menentukan kualitas sediaan dan membuat profil permeasi in vitro gel kloramfenikol. Menentukan kualitas sediaan gel mata kloramfenikol dengan melakukan pengamatan selama 28 hari pada pengujian organoleptis, pH, Viskositas, kadar kloramfenikol dalam sediaan dan uji sterilitas. Profil permeasi in vitro gel mata kloramfenikol dilakukan uji sel difusi Franz dengan membran kornea mata kelinci selama 8 jam. Hasil pengamatan sediaan gel mata kloramfenikol pada pengujian organoleptis, pH, Viskositas, kadar kloramfenikol dalam sediaan dan uji sterilitas, menunjukkan hasil yang baik, dan hasil uji difusi gel mata kloramfenikol menunjukkan kadar terpermeasi sebesar 1,513% selama 8 jam. Kata kunci: gel mata, kloramfenikol, sel difusi franz

Abstract

Pembuatan bentuk sediaan farmasi memerlukan eksipien. Eksipien yang digunakan dalam suatu formula obat berasal dari bahan alam atau sintesis. Buah Okra (Abelmoschus esculentus) mempunyai kandungan mucilago yang dapat diperoleh melalui proses ekstrasi. Mucilago yang berasal dari tanaman dalam berbagai penelitian memiliki fungsi sebagai eksipien farmasi. Mucilago Okra dapat dimanfaatkan sebagai pengemulsi, pensuspensi, pengikat tablet, matrix tablet dalam pembuatan tablet lepas lambat.

Abstract

Vitamin E (Tocopherol acetate) is used in both oral and topical dosage.  The aim of  the research was to study  different concentration of Carrageenan, as gelling agent in Tocopherol acetate Emulgels.  This experimental study was initiated by emulgel formulation using different concentration of carrageenan as gelling agent, as much as 0,5% (F1), 0,75% (F2), 0,85% (F3), 0,95 % (F4), 1% (F5), 1,125% (F6), 1,25% (F7), 1,5% (F8) and 2% (F9).  Tween 20 and Span 20 were used as emulsifiers (1 and 1,5 %),  Liquid paraffin as oily phase (7,5%), Propylen glycol as humectant (10%), propyl and methyl paraben as antimicrobial preservative (0,01% and 0,03%). The physical investigation of emulgel were included pH, spreading test, viscosity and freeze thawtest. The results for promising formula F5 showed that physical parameters of emulgels were in the range of requirement for topical dosage form. Keywords: Tocopherol acetate (Vitamin E), emulgels, carrageenan

Abstract

Skin constantly exposed by the environmental stresses, such as pollutants, solar radiation, heat, and cold, which cause the free radicals that cause premature aging. This bad effects of free radicals can be reduced by natural antioxidants such as Beluntas china (Gynura pseudochina (L.) DC) extract. The antioxidants use of beluntas china extract for skin will be more optimal if it is applied by appropriate form, emulgel, which has stronger consistency and longer contact time than gel. The aim of this research was to formulate an antioxidant emulgel from beluntas china extract, which is effective, stable, and safe with the best gelling agent. The extract was formulated by variety types of gelling agents which were carrageenan, hydroxypropyl methylcellulose (HPMC), and carbomer 934. Then, the physical stability was evaluated, including organoleptic, homogeneity, pH, viscosity, and freeze thaw. The results showed that formulation of carbomer 934 1% 2xIC50 extract concentration give the best physical stability evaluation for 60 days storage time.Keywords: antioxidant, beluntas china extract, emulgel, free radicals, skin

Abstract

Cellulose nanocrystals is a cellulose derivates which has been widely researched and observed as an chemical agent. Different with cellulose that has been widely used as pharmaceutical excipient especially in solid dosage form, cellulose in nanocrystals form is not available in pharmaceutical grade. Cellulose nanocrystals have different characteristics and quality which is depend on its preparation including sourcing, isolation procedure, and hydrolysis reaction involved. This difference is very important to deeply observed its impact in pharmaceutical dosage form with different active ingredients. In addition, cellulose nanocrystals should meet FDA requirement as pharmaceutical excipient. This review  describe cellulose nanocrystals preparation and its characteristics, its application to active pharmaceutical ingredients, and its properties in order to meet FDA requirement.Keywords: Cellulose, nanocrystals, pharmaceutical excipient

Abstract

 Tetes mata sulfasetamidayang mengandung natrium sulfasetamida 10%, 15%, dan 30% telah dibuat dan disterilkan. Metode sterilisasi yang digunakan adalah uap air mengalir 98-100 oC, penyaring bakteri, dan autoklaf 120-121 oC selama 15 menit. Tujuan penelitian ini adalah mendapatkan sediaan yang paling stabil selama penyimpanan 28 hari. Semua sediaan diamati kejernihan, pH dan konsentrasi natrium sulfasetamida.Semua sediaan mengalami kenaikan pH. Kekeruhan terjadi pada sediaan yang disterilkan dengan autoklaf. Sediaan paling stabil adalah tetes mata yang mengandung natrium sulfasetamid 10% yang disterilisasi dengan penyaring bakteri.

Abstract

Tanaman seledri (Apium graveolens L.) mengandung berbagai senyawa metabolit sekunder antara lain flavonoid, polifenol, dan kuinon. Tanaman seledri selain untuk bumbu masakan dan sayuran, telah lama digunakan sebagai obat tradisonal untuk penurun tekanan darah tinggi (hipertensi), diuretik, dan hematuria. Penelitian ini bertujuan untuk mengungkap khasiat lain dari tanaman seledri sebagai antikalkuli atau peluruh batu ginjal. Pengujian efek antikalkuli (bagian dari uji preklinis) dilakukan secara in vitro yaitu dengan menguji tingkat kelarutan komponen batu ginjal (kalsium oksalat atau magnesium ammonium fosfat) sebagai solut (100 mg serbuk batu) dalam berbagai variasi konsentrasi sediaan cair seledri sebagai solven dibandingkan dengan solven air (volumen= 10 mL, suhu= 37 oC, waktu= 4 dan 24 jam). Hasil penelitian menunjukkan bahwa cairan infusa seledri pada konsentrasi 1,3; 3,3; dan 5,0% dapat melarutkan komponen kalsium dan magnesium batu ginjal dengan tingkat kelarutan yang secara signifikan lebih besar dibandingkan kelarutan dalam air sebagai kontrol negatif (konsentrasi 5%, Ca: 4,657 vs 199 ppm, Mg: 9,912 vs 9,37 ppm). Sementara fraksi air dari ekstrak metanol seledri juga menunjukkan daya larut yang signifikan terhadap baik kalsium maupun magnesium komponen batu ginjal pada konsentrasi pada 0,5% dibandingkan air (Ca: 3,7 vs 1,5 ppm dan Mg: 25,9 vs 14,5 ppm). Hasil tersebut dapat disimpulkan bahwa herba seledri memiliki potensi sebagai peluruh batu ginjal dengan mekanisme melarutkan kristal komponen batu ginjal. Abstract

Bawang Tiwai (Eleutherine americana (L.) Merr bulbus. is a typical plant from Kutai tribes which commonly used as an anti-bacterial and food coloring. The stability test is performed to determine the stability of the extract. Stability test was conducted in two solvents are distilled water and ethanol include the effects of temperature (25°C, 50°C, 80°C), pH (3, 5, 7), an oxidant for 6 hours, conditions of storage at room temperature and the temperature of the refrigerator for 24 hours and storage for 2 weeks. The absorbance of extract in ethanol and aquadest was decreasing due to the change of temperature, time, the presence of oxidator but due to pH changes, the absorbance of extract was increasing. Meanwhile, on the storage condition, showed the highest absorbance on dark, and room temperature storage compared to other condition.Key words: Eleutherine americana (L.)
