Abstract

?-amilase is an enzyme that catalyzes the hydrolysis of starch polysaccharides into oligosaccharides to then produce glucose. Inhibitions of the ?-amylase enzyme is an effective strategy in modulating blood sugar levels in diabetics. The aglycon curculigoside A has similar structure to chalcon which is able to inhibit the ?-amilase enzyme for the treatment of type 2 diabetes. The aim of this study was to determine the interaction of the curculigoside A aglycone compound and its derivatives with the ?-amylase enzyme by using the docking simulation method. Docking simulations were carried out by using AutoDock 4.2 and ?-amilase protein (PDB ID: 1B2Y) as macro molecule. The docking results showed that the aglycone curculigoside A and its derivatives able to interacted into the active site of the ?-amylase enzyme. Three best ligands according to the simulation and prediction tests were compound 10, compound 23, and compound 41 has formed hydrogen bonding to Asp197, Glu233 and Asp300 residues with free bonding energy of -7.29, -7.22, dan -7.84 kcal/mol, respectively. In conclusion, Three best ligands has the same pattern of hydrogen bonds to the native ligand AC1 (6-methyl-5-(4,5,6-trihydroxy-3-hydroxymethyl-cyclohex-2-enylamino)-tetrahydro-pyran-2,3,4-triol) via amino acids redisues of ?-amylase that play a role in the substrate by hydrolysis process.Keywords: aglycon curculigoside A, ?-amylase, diabetes, docking

Abstract

Testing activity sun screen of several species from family Zingiberaceae with a spectrophotometric method have been. Study purpose is to obtain data on the sunscreen activity of ginger extract Z. officinale var. Officinale, Z. officinale var. Rubrum and Zingiber zerumbet SM and their combination with the parameters of % transmission of erythema, % transmission of pigmentation and SPF. The method used an experimental laboratory. Evaluation of the activity of ethanol extract of Z. officinale var. Officinale, Z. officinale var. Rubrum and Zingiber zerumbet SM made by maceration method using 70% ethanol. The test is based on the ability of the absorb the rays ultraviolet. Research carried out by the respective concentrations of ethanol extract of Z. officinale var. Officinale, Z. officinale var. Rubrum and Zingiber zerumbet SM of 200 ppm , 400 ppm , 600 ppm, 800 ppm and 1000 ppm. Based on percent transmission of erythema and percent transmission of pigmentation at a concentration of 1000 ppm, respectively ethanol extract Z. officinale var. Officinale, Z. officinale var . Rubrum and Zingiber zerumbet SM could be used as a sunscreen but still weak to be used as sunscreen ingredients. The result showed that combined Z. officinale var. Officinale, Z. officinale var. Rubrum and Zingiber zerumbet SM at a concentration of 500 ppm obtained percentage value of 1% transmission of erythema and 3,9% of pigmentatied transmission and categories as sunblock with SPF value is 50 protection.

Abstract

Has conducted research of antioxidant activity from kersen leaves (Muntingia calabura Linn) using DPPH (1,1-diphenyl-1-picrilhydrazil) and FRAP (ferric reducing antioxidant power). Kersen leaves extracted by maceration using ethanol 70%. Phytochemical identification of kersen leaves contain phenolic, flavonoids and saponins compounds. The test results of antioxidant activity using DPPH method with series concentrations of 2, 4, 6, 8, and 10 ppm obtain IC50 value 6.8249 ug / ml. Testing of the ethanol extract of antioxidants obtained by FRAP method IC50 83,149 µM. Based on these data the ethanol extract of kersen leaves has good antioxidant activity.

Abstract

Penelitian ini bertujuan untuk menentukan bioaktivitas dari daging buah dan kulit buah L.domesticum sebagai bahan aktif kosmetik. Proses penyiapan ekstrak dilakukan dengan metode maserasi dengan pelarut etanol 96 % dan etil asetat, sehingga diperoleh ekstrak etanolik daging buah (BLE), ekstrak etanolik kulit buah (KLE), ektrak etil asetat daging buah (BLEA) dan ekstrak etil asetat kulit buah (KLEA). Selanjutnya ditentukan golongan senyawa dengan kromatografi lapis tipis dan penentuan kandungan fenolik dan flavonoid total dengan metode kolorimetri. Aktivitas antioksidan diukur menggunakan pengujian peredaman radikal 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching (BCB) dan Ferric reducing antioxidant power (FRAP). Efek penghambatan enzim pendegradasi kulit (antiaging) dilakukan dengan mengukur penghambatan enzim elastase dan kolagenase. Efek penghambatan tyrosinase juga diukur menggunakan mushroom tyrosinase. Berdasarkan profil kromatografi lapis tipis (KLT), menunjukkan bahwa daging buah dan kulit buah Lansat mengandung senyawa golongan fenolik, flavonoid, terpenoid dan steroid. Kandungan flavonoid total terbesar terdapat pada KLE (4,76±0,26 %b/b Ekuivalen Kuersetin atau EK) dan kandungan fenolik total terbesar terdapat pada BLEA (6,4±0,15 %b/b Ekuivalen Asam Galat atau EAG). Hasil aktivitas antioksidan pada masing-masing ekstrak dengan menggunakan DPPH: BLEA>KLE>BLE>KLEA; BCB: KLEA>BLEA>KLE>BLE; dan FRAP: BLEA>KLE> BLE>KLEA. KLE dan KLEA memberikan hasil yang baik dalam menurunkan aktivitas dari enzim elastase dan enzim kolagenase. Selain itu, pengujian anti tyrosinase menunjukkan bahwa BLE memiliki aktivitas anti-tyrosinase terbaik dibanding ekstrak lainnya. Berdasarkan dari hasil tersebut dapat disimpulkan bahwa ekstrak buah dan kulit buah langsat memiliki potensi sebagai bahan aktif kosmetik untuk anti aging dan anti tyrosinase.

Abstract

Daun beroma (Cajanus cajan (L.) Milps) berpotensi sebagai antioksidan dikaitkan dengan kandungan senyawa kimia yaitu flavanoid dan fenolik. Aktivitas antioksidan dari ekstrak etanol 70% daun Cajanus cajan (L.) Milps dilakukan menggunakan metode 1,1- diphenyl-2-picrylhydrazyl (DPPH) dan 2,2’-azino-bis- [3- ethylbenzothiazoline sulphonate] (ABTS). Nilai IC50 pada pengujian metode DPPH yaitu 86,34 mg/ml dan IC50 pada metode ABTS yaitu 20,53 mg/ml. Berdasarkan nilai IC50 tersebut, dapat disimpulkan bahwa ekstrak etanol daun Cajanus cajan (L.) Milps mempunyai aktivitas antioksidan yang sangat kuat.

Abstract

Kemangi (Ocimum canum Sims L.) is one of the plants producing essential oils. Kemangi essential oil is taken from the stem and leaves. This study aims to determine the precentage of the yield of kemangi leaf oil and to identify the content of the chemical compounds of essential oils found in kemangi leaves based on the comparison of the place of growth and duration of distillation. Kemangi leaf samples were obtained from the Bontoala sub-district and from Tinggimoncong sub-district of South Sulawesi. The extraction method of essential oils of kemangi leaves is done by the method of distilation/hydrodestilation using three variations of distillation time, namely 6 hours, 12 hours and 18 hours. Based on the results of the study, it can be concluded that the kemangi leaf essential oil which has the highest yield is from Tinggimoncong sub-district of South Sulawesi, which is 3.325 % with distilation time for 18 hours, GC-MS identification results also show that kemangi leaves essential oil from Tinggimoncong sub-district of South Sulawesi has 93 components of chemical compounds with 5 main components, namely: 5-heptene-2-one, 6-methyl, 1,6-octadien-3-ol, 3,7-dimethyl, ?-Terpeniol, 2,6-octadienal, 3,7-dimethy (E), eugenoland also has a component of minor compounds, namely: citral, lynalil acetate, eucalyptol, geraniol, verbenol, carveol, and trans-isoeugenol.

Abstract

Faloak (Sterculia populifolia DC.) is one of species of sterculiaceae found in East Nusa Tenggara which has potential as a medicinal plant mainly as an antimicrobial. This study aims to determine the correlation of physicochemical study of faloak bark extracts with variation of solvents in inhibiting of pathogenic bacteria. The sample was extracted by maceration method with different polarity level of solvents i.e acetone, acetone 70%, water, ethanol 96%, ethanol 70% and ethanol 50%. The results of extraction through maceration indicate the difference of yield recovery from each of the extraction solvents. The highest yield was obtained from 70% ethanol extract, while the lowest yield of acetone extract. The increase of solvent polarity in this study did not give effect to the amount of recovery of yield. It is also seen from the highest total phenolic content obtained from 70% acetone extract while the lowest in aquadest extract. The antibacterial activity of faloak bark extract on Salmonella typhi was tested using agar diffusion method with 1% of extract solution. Both of ethanol 96% and acetone extracts did not show any inhibitory activity. The largest inhibitory activity was demonstrated by 50% ethanol extract. The polarity level of the extract, the level of total phenolic content and the magnitude of rendement  did not show correlation of increased inhibitory activity on Salmonella thypi as well.

Abstract

Green Synthesis is a method to make silver nanoparticles (AgNPs) by utilizing natural materials as bioreductors. One of the natural materials that can reduce metal ions is Kaempferia galanga rhizome because it has a chemical component that can penetrate the metal. The compounds are ethyl paramethoxycinnamate (EPMC) which is used as sunscreen. This study aims to produce AgNPs conjugated with EPMC that can be used as raw material sunscreen. This study was conducted by extracting EPMS from the Kaempferia galanga rhizome and synthesizing the AgNPs conjugated with EPMC. The synthesis results were characterized by UV-Vis Spectrophotometry, FTIR, SEM, PSA and sunscreen activity. Extracted result was obtained crystalline is ethyl paramethoxycinnamate compound with yield 2,3%. The characterization of EPMC with UV-Vis spectrophotometer was obtained maximum wavelength of 308 nm and FTIR analysis result at wave number 1701,27 cm-1 indicated the presence of carbonyl group (C = O) and 1165,97 cm-1 presence of C-O and showed specific characteristic from EPMS. Characterization of AgNPs using Uv-Vis spectrophotometer was obtained maximum wavelength of 469 nm. Characterization of AgNPs using SEM are spherical and monodispers forms.  Then, particle size using PSA obtained an average diameter of 182 nm. AgNPs conjugated with EPMS has a sunscreen activity with sunblock category and successfully provide protection against UV exposure at low concentration (200 ppm) categorized as ultra protection with SPF value 36,4.

Abstract

This experiment aims to determine the correlation of total phenolic and flavonoid content of jati putih leaves fraction (Gmelina arborea Roxb.) towards Antioxidant activity .  Sample was extracted by maceration method using ethanol 70% to obtain the ethanol extract (EE), followed by liquid-liquid extraction method to obtain fraction of ethyl acetate (EA) and n-Hexane (EH). The phytochemical screening  and determination of total phenolic and flavonoid content were done by colorimetric method. Antioxidant activity were done by DPPH, FRAP and ABTS methods. Phytochemical screening showed positive results for flavonoids, phenolic and saponins.  The largest total phenolic content was found on EA (11,59 µg/ml ± 0,3 %b/b EAG) and the largest total flavonoid content was on EA (3,88 µg/ml ± 0,02 %b/b EK). The total phenolic and flavonoid content of Jati putih leaves has a correlation with antioxidant activity. The coefficient correlation of activity on reducingDPPH radical was 56,7% (total of phenolic content) and 57,8% (total of flavonoid content) and on iron reduction power in FRAP method  was 99,9% (total of phenolics and flavonoids content). The relationship with the activity in reducing radical ABTS obtained coefficient correlation of 57,0% and 58,1% for total phenolic and flavonoids contents, respectively.

Abstract

The leaf of kedondong hutan (Spondias pinnata (L.F.) Kurz.) Contain of flavonoid compound, which capable as antioxidants that function for preventing free radicals. The purpose of this study was to determine the antioxidant activity profile of ethanol extract of kedondong hutan leaf with DPPH, ABTS radical and iron reduction methode (FRAP). This study used experimental research with stages of sample collection, plant determination, simplicia preparation, maceration with 70% ethanol, phytochemical screening and antioxidant activity assay with UV-Vis spectrophotometry. The results obtained from the ethanol extract of kedondong hutan leaves had antioxidant activity on DPPH and ABTS with IC50 of 32.83 ppm and 45.84 ppm respectively. In FRAP method kedondong hutan leaves had antioxidant activity which is equivalent to quarcetin of 2936.7 µmol QR /g sample. Ethanol extract of kedondong hutan leaves has very strong antioxidant power against DPPH, ABTS radical and iron reduction.
