Abstract

Ganoderma  philipii is a fungi causing root rot disease that have spread widely in Acacia mangium plantation. Study on mode of disease spreading is necessary to develop ways of controlling the spread of the disease. Somatic incompatibility test was carried out using fungal cultures that has been identified as G. philippii from two sites of different rotation. Each location showing different type of incompatibility. Compatible reaction was observed at Logas of first rotation indicating that the isolates are not genotypically distinct and may come from closely related clones. At this site the spread of root rot occurred by root to root contact. Incompatible reaction was observed at Deras of second rotation indicating that G. philipii isolate are genotypically distinct individuals and of different clones, eventhough they share same morphological characters. Genetic diversity found in G. philippii suggested that sexual reproduction of basidiocarps in an important factor and is strongly implicated as one of the main modes of dispersal as well as root to root contact.

Abstract

Identification of wood decay fungi based on morphological character of isolates is very helpful where identification of fruit body morphology is ambiguous. Nine isolates of Phlebiopsis sp.1 obtained from the isolation root of Eucalyptus pellita in permanent plots with root rot disease have the potential as biological control for G. philippii. The verification based on ITS sequences of rDNA showed that these isolates were closely related to P. gigantea and P. flavidoalba. The characterisation of morphological was performed on isolates grown on Malt Extract Agar media with sawdust. Based on the macroscopic observation, the isolates were categorized into three different morphotypes: (1). Cottony mycelium from the beginning to the advanced stage, one isolate (Pb5); (2). Cottony at the beginning and then turned into farinaceous to floccose on the advanced stage, 7 isolates (Pb1, Pb2, Pb4, PB6, PB8, Pb9, Pb10); (3). Absent at the beginning and then turned into zonate on the advanced stage, one isolate (Pb11). Morphotype 2 is the dominant group of Phlebiopsis sp.1 isolates, with fine tufts of white mycelium which turned brown on the surface of the colonies. Morphotype 3 has very different morphological characters than other 8 isolates, it has very characteristic concentric circle with different color and texture of each circle. The growth rate of mycelium ranged between 10.70 - 10.85 cm²/day for morphotype 1 and 2; 1.95 cm²/day for morphotype 3.

Abstract

The incidence of rot disese has inflicted substantial damage to some plantation of Acacia mangium in Sumatera and Kalimantan. As soil-borne pathogen, Ganoderma is commonly found in the tropic and in Indonesia the disease also couse significacant losses to rubber and soil palm plantation. To help control the disease, appropriate identification of the fungi cousing the disease and other field characteritics are critical. Morphological characteistics applied in this study has proved to be effective to identify the pathogen. The disease is predominantly caused by G. philippii. The result of morphological identicication was verified with the DNA sequence of the same fungus and confirm the accuracy of the morphological characteristics. Other fungi that attack root of Acacia mangium are G. australe and Phellinus spp.

Abstract

Root rot caused by Ganoderma philippii (Bres. & Henn. ex Sacc.) Bres., is an important disease in Acacia plantation. A strategy to control this disease is currently being developed, particularly on the application of biological control agent (BCA). Species specific primers for rapid identification of potential fungi as BCA were developed. This study aimed to obtain the best DNA condition and the best primers for species specific identification. DNA with 20x dilution is the best condition for amplification of the ITS fragment thus used for rapid species identification. The best primer set to detect Cerrena sp. is CrF1/CrR1, PbF2/PbR2 is the best primer set for Phlebiopsis sp. 1. There was no specific primer suitable to detect Phlebia sp. 1 and Phlebia sp. 2 only but Pl-2F1/Pl-2R4 is the best primer set for Phlebia spp.

Abstract

The purpose of this study is describing the mathematical problem solving ability using the TGT (Teams Games Tournament) learning model for the students of SMA Muhammadiyah 1 Purwokerto. This study uses qualitative research methods using the Miles and Huberman model which includes data reduction, data display, and conclusion (verification/conclusion drawing). The subjects of this study are students of X IPA 1 SMA Muhammadiyah 1 Purwokerto. By using purposive sampling technique, the students of  X IPA 1 were in three groups: the group of low achieving students, the group of medium achieving students, and the group of high achieving students. In each group, three students were selected to become respondents and then analyzed. Data collection methods in this study use questionnaire, tests, interviews, and documentation. From the results of this study, obtained: 1) the students with low achievement have a poor problem-solving ability, 2) the students with medium achievement have a good problem-solving ability, 3) the students with high achievement have great problem-solving skills.
