Abstract

Bacterial endophytes in trees are potential source to obtain new antibiotic producers that have not been widely studied until now. This study aims to obtain bacterial endophytes isolates in the tissue of twig trees in National Park Banten, Indonesia Germplasm Garden (The Bogor Botanical Gardens), Cilacap Nusakambangan Island, Kaliurang National Park, Meru Betiri National Park Jember, and Baluran National Park, Situbondo. This selection was based on the growth ability in liquid nutrient, GY (glucose and yeast-extract), and antibiotic-3, and the ability to obstruct indicator microbes, such as Bacillus subtilis, Candida albicans, and Fusarium oxysporum. The selection result is obtained eight isolates that have inhibitory effect more than 4, such as BIN-1, KLP-1, OOH-1, STG-1, CMB-2, STL-1, MTG-1, and PPH-1. Based on chromatography method on various eluents, it is obtained three superior isolates, i.e OOH-1 ,STG-1, and CMB-2. Thus, this shows that bacterial endophytes in trees constitute potential sources for screening new antibiotic producers.

Abstract

Endophytic microbes are potential sources of antibiotics. Some numbers of endophytic bacteria were isolated from plants in Ujung Kulon, Kaliurang, Meru Betiri and Baluran National Park, Bogor Botanical Garden, and Nusakambangan forest, Indonesia. Previous studies have been conducted to examine and obtain endophytic bacteria isolates from the selected plants, which resulted in three selected isolates, namely OOH-1, STG-1, and CMB-2. This research was conducted to determine the molecular identity of OOH-1 and STG-1 isolates, as well as to identify antibiotic compounds produced by STG-1 isolate. Molecular identifcation of selected isolates was based on 16S rRNA gene analysis and amplifed using primers 27F and 1492R. A phylogeny tree was then constructed by comparing the resulting sequences with data from Gene Bank using the BLAST-N program. The identifcation showed that STG-1 isolate had a 99% similarity with Pseudomonas brenneri strain SFML 97-391, and OOH-1 isolate had a 99% similarity with Enterobacter xiangfangensis. Identifcation of antibiotic compounds was done by purifcation and separation of the compounds. Antibiotic activity was also examined based on Lethal Concentration (LC50) on Fusarium oxysporum with a LC50 of 0.01–0.02% against Fusarium oxysporum.

Abstract

Perkembangan penelitian arkeologi ternyata tidak pernah lepas dari berbagai disiplin ilmu, yang kesemuanya bertujuan untuk mempercepat pemecahan masalah arkeologi yang ada. Penggunaan ilmu lain termasuk pengembangan teknik modern dalam arkeologi, meliputi juga konsep dari lingkungan ilmu-ilmu eksakta.Analisa pollen merupakan salah satu kegiatan yang berhubungan dengan kegiatan multidisipliner dalam penelitian arkeologi. Kegiatan ini adalah usaha yang menyangkut penerapan ilmu dari lingkungan biologi-geologi kearah arkeologi. Penerapan analisa pollen di negara-negara maju sudah lebih dahulu dikerjakan. Dengan pengembangan analisa pollen dalam penelitian arkeologi di Indonesia, akan membuat penelitian arkeologi setapak lebih maju dan diharapkan sesuai dengan negara maju yang lain.
