Abstract

ABSTRAK   Pelepasan natrium diklofenak dari Sediaan Transdermal Patch Tipe Matriks dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 telah diteliti. Pada penelitian ini sediaan transdermal patch tipe matriks  yang mengandung natrium diklofenak dibuat dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 pada perbandingan yang berbeda ( 9:1 (Formula I); 8:2 (Formula II); dan 7:3 (Formula III)). Persentase (%) moisture content dihitung dari perbedaan berat relatif pada penimbangan akhir. Uji homogenitas permukaan patch dilakukan dengan menggunakan metode Scanning Electron Microscopy (SEM). Uji disolusi dengan media larutan dapar fosfat salin pH 7,4 ± 0.05 sebanyak 500,0 mL yang terlebih dahulu dipanaskan sampai mencapai temperatur percobaan 37 ± 0.5°C diaduk pada kecepatan 50 rpm. Hasil dianalisis menggunakan SPSS dengan metode ANOVA one way dengan tingkat kepercayaan 95% (a = 0,05). Berdasarkan penelitian ini, penggunaan polimer kombinasi antara etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 dapat meningkatkan fluks pelepasan natrium diklofenak dari sediaan patch natrium diklofenak. Perbandingan 7:3 dari etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30  adalah kombinasi yang terbaik untuk sediaan patch natrium diklofenak karena konsistensi bentuk fisik serta nilai fluks pelepasan dan penetrasi yang dihasilkan lebih besar dari perbandingan lainnya.   Kata Kunci :   Pelepasan Sodium diklofenak, plasticizer, kombinasi matrix dari EC N-20  dan PVP K-30     ABSTRACT   The sodium diclofenac realeased from transdermal patch of combination of ethyl cellulose (EC) N-20 nd polyvinyl pyrollidon (PVP) K-30 was investigated. In this study, matrix-type of transdermal patchntaining diclofenac sodium were prepared using polymeric combination of EC N-20  and PVP K-30 in various  ratios ( 9:1 (Formula I); 8:2 (Formula II); and 7:3 (Formula III)). The percentage of moisture content was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using fluorescence microscope and scanning electron microscope (SEM). Released test using dissolution apparatus were carried out in 500 mL of phosphate buffer saline pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. Results were analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0,05).  It can be concluded that the combination of EC N-20 and PVP K-30 at ratio 7:3 was the best choice for manufacturing transdermal patch based on physicochemical and the release profile.   Keywords:    Diclofenac sodium released, plasticizer, combination matrix of EC N-20 and PVP K-30

Persentase kadar air dihitung sebagai perbedaan antara berat awal dan akhir. Homogenitas dari permukaan Patch di determinasi menggunakan Mikroskop flourescence dan SEM. Uji pelepasan dilakukan menggunakan alat uji dissolusi dalam 500 mL larutan salin fosfat buffer pH 7.4 ± 0.05 pada suhu 37 ± 0.5°C dengan kecepatan pengadukan 50 rpm. Hasil di analisis menggunakan program statistik SPSS analisis one way variance (ANAVA) dengan tingkat kepercayaan 95% (α = 0,05), menunjukkan bahwa kombinasi dari EC N–20 and PVP K–30 pada rasio 6:4 dengan plasticizer PEG 400 5% merupakan pilihan terbaik untuk industri Patch transdemal berdasarkan fisikokimia dan profil pelepasan.

Abstract

A research has been conducted on the antibacterial activity of the santol leaves extract (S.koetjape Merr.) against S.aureus and E.coli. The santol leaves extracts were prepared by maceration used methanol as solvent and the methanol extract was then fractionated. The antibacterial activity was determined by measured the diameter of inhibition zones in difusi method. The result showed that santol leaves extracts and fractions has antibacterial activity against S.aureus and E.coli. The effective concentration as an antibacterial of methanol extracts is at 10%, and of n-hexane fractions is at 5%. Keywords : S.koetjape Merr, Santol leaves, Antibacteria   Abstrak Telah dilakukan penelitian uji aktivitas antibakteri ekstrak daun kecapi (S.koetjape Merr.) terhadap S.aureus dan E.coli. Ekstrak daun kecapi dibuat secara maserasi dengan pelarut metanol, lalu difraksinasi. Uji aktivitas antibakteri ekstrak daun kecapi diukur berdasarkan luas daerah zoa bunuh pertumbuhan bakteri dengan metode difusi agar. Hasil uji menunjukkan bahwa ekstrak dan fraksi daun kecapi memiliki aktivitas antibakteri terhadap bakteri S.aureus dan E.coli. Konsentrasi efektif antibakteri ekstrak metanol adalah 10% dan fraksi n-heksana adalah 5%.  Kata Kunci : S.koetjape Merr, Daun Kecapi, Antibakteri

Abstract

Each plant is host to one or more endophytes contain bacteria and fungi. Endophytic fungi could potentially have an activity and produce secondary metabolites same as its host. Yacon leaf (Smallanthus sonchifolius) has been known to have antimicrobial activity that allow the endophytic fungi also has potential as antimicrobial. The purpose of this research was to isolated the endophytic fungi from Yacon leaf which has activity as antifungal and can produce secondary metabolites same as its host. The method that used to test the activity of endophytic fungi are using direct planting method, whereas the identification of secondary metabolites using Thin Layer Chromatography (TLC) and spray reagent. The results were obtained that the endophytic fungi has an activity as antifungal against Candida utilis, Candida albicans and Malassezia Purpur, along with the presence of secondary metabolites that same as its host.Key words: endophytic fungi, Smallanthus sonchifolius, antifungal, Thin Layer Chromatography (TLC)ABSTRAKSetiap tanaman mengandung satu atau lebih mikroba endofit yang terdiri dari bakteri dan jamur. Jamur endofit berpotensi memiliki aktivitas dan menghasilkan metabolit sekunder yang sama seperti inangnya. Daun Yakon (Smallanthus sonchifolius) telah diketahui memiliki aktivitas sebagai antimikroba sehingga memungkinkan adanya jamur endofit yang juga berpotensi sebagai antimikroba. Tujuan dari penelitian ini adalah didapatkan isolat jamur endofit dari daun Yakon yang memiliki aktivitas sebagai antijamur dan dapat menghasilkan metabolit sekunder yang sama seperti inangnya. Metode yang digunakan untuk menguji aktivitas isolat jamur endofit adalah menggunakan metode tanam langsung, sedangkan identifikasi metabolit sekunder menggunakan metode Kromatografi Lapis Tipis (KLT) dan pereaksi semprot. Hasil penelitian yang diperoleh adalah adanya isolat jamur endofit yang memiliki aktivitas sebagai antijamur terhadap jamur Candida utilis, Candida albicans, dan Malassezia purpur, serta adanya metabolit sekunder yang sama seperti inangnya.Kata kunci: Jamur endofit, Smallanthus sonchifolius, antijamur, Kromatografi Lapis Tipis (KLT)
