Resistance Â to Â chemotherapy Â is Â believed Â to Â cause Â treatment Â failure Â of Â the Â patientÂ cancer. Secang (Caesalpinia sappan L.) has been proven to possess anticancer activity on someÂ cancer cell lines. The aimed of this study to develop ethanolic extract of secang wood (EES)Â as Â chemosensitizing Â agent Â through Â apoptotic Â induction Â on Â breast Â cancer Â MCF-7 Â cells.Â Extraction Â of Â secang Â was Â done Â by Â using Â maceration Â with Â 70 Â % Â ethanol. Â Single Â andÂ combinatorial treatment of EES and doxorubicin on MCF-7 breast cancer cells were analyzedÂ by using MTT assay to determine the IC50 value and combination index (CI) to evaluate theÂ combinatorial effect. Apoptosis was analyzed with flowcytometry (annexin V). Â EES showed aÂ dose-dependent cytotoxicity (IC50 value of 37 Âµg/ml), while combinatorial treatment showedÂ that Â 7 Â concentrations Â was Â found Â to Â be Â synergist Â with Â doxorubicin Â on Â MCF-7 Â cells.Â Combinatorial treatment also triggered apoptotic instead of single treatment. Based on thisÂ result, Â we Â conclude Â that Â ethanolic Â extract Â of Â secang Â wood Â is Â potential Â as Â chemosensitizingÂ agent in breast cancer.Keyword: Caesalpinia sappan L, MCF-7 cells, doxorubicin, apoptosis.
