Abstract

The aim of the investigation is to obtain a good preparations of effervescent granules of ethanol extract of pepino fruit (Solanum muricatum Aiton) based on parameter of flowability and repose angle, density, moisture content, loss on drying and dispersibility. Maceration method and ethanol 96% as the solvent is use to obtain the extract of pepino fruit (Solanum muricatum Aiton). There are three formula design of effervescent granules formula 1,2 and 3 each of which in variation of  binder PVP K30 2%, 4% and 6% (w/w). The granules is obtain by wet granulation method in which ethanol extract of pepino fruit as the active ingredient. Results of evaluation showed that angle of repose  range from 33.425-34.352o while flowability range from 5.84-6.66 g/second. Bulk density range from 0.519-0.570 g/ml, tap density 0.552-0.53 g/ml, true density 0.2005-1.22 g/ml. Uji porositas berkisar 6.36-44.609 % while the dispersibility range from 1.407-1.477 gram/second. Based on the results can be concluded that all granules possess good properties pharmaceutically.

Abstract

TB is an infectious disease caused by the bacterium Mycobacterium tuberculosis. Empirically the plants that have long been used in medicine are Curcuma mangga  rhizome, which shows pharmacological activity as an anti Mycobacterium tuberculosis drug. The objective of the research is to determine the ability of hexane and methanol extracts of Curcuma mangga in inhibiting the growth of Mycobacterium tuberculosis Strain H37Rv and MDR. The extraction of Curcuma mangga rhizome was conducted by maceration methode using hexane and methanol as the solvents, respectively.  Both of extracts were made in 1000 dan 500 ppm concentration, then tested for anti Mycobacterium tuberculosis activity using MODS (Microscopic Observation Drug Suspectibility) method. Determination of anti M.tuberculosis activity was based on microscopic observation on Mycobacterium tuberculosis colony in cord formation. The results showed that hexane and methanol extract in 1000 ppm concentration inhibited the growth of Mycobacterium tuberculosis Strain H37Rv dan MDR. However in 500 ppm only n-hexane  extract showing the inhibiton activity. In conclusion, n-hexane extract have the ability to inhibit the growth of Mycobacterium tuberculosis Strain H37Rv dan MDR.

Abstract

Dengue hemorrhagic fever (DHF) threatens people in the rainy season. DHF is one of the major public health problems in Indonesia. These problems encourage an effort to eradicate  Ae. aegypti larvae by using natural larvicide. One of the plants that can be used as natural larvicides is avocado seeds (Persea americana Seed) which contain saponin. This study aims to determine the larvacidal activity of avocado seed extract sandgranules with LC50 of 8.87 mg.mL-1 against Ae. aegypti larvae. This study is an experimental research with a complete random design in a unidirectional pattern. The independent variable in this study is the variation concentration of Explotab as a disintegrant agent ie 2, 3, and 4%. Sand granules of avocado seed extract are made by wet granulation method. Results of the larvacidal activity test of  avocado seed extract sand granules were analyzed by calculating the percentage of larvae mortality and statistical analysis using SPSS. The results of this study is the larvacidal activity of pure avocado seed extract with LC50 8.87 mg. L-1 is greater than the avocado seed extract sand granules. The addition of Explotab did not affect the total larvae mortality, but there was a significant difference in larvae mortality at 5 hours of sand granules exposure with explotab concentration of 3% and 4% (a significance value <0.05).

Abstract

Arumanis mango leaves (Mangifera indica L.) has antibacterial activity and antifungal, evidently. Escherichia coli and staphylococcus aureus are opprtunistic pathogen bacteria that can cause gastrointestinal and skin infection. Purification of extract was done to eliminate the presence of ballast substances that cannot produce therapeutic effects. This study aims to acknowledge the antibacterial activity of purified mango arumanis leaves extract (PMALE) against E. coli and S. aureus, flavonoid compound content by TLC. Arumanis Mango leaves powder was extracted with 96% ethanol solvent by maceration method, followed purification the extract was done with hot water-ethylacetate solvent by liquid-liquid extraction method. Antibacterial activity test was done by agar diffusion method against E. coli and S. aureus bacteria with the same serial concentration of 6.25; 12.5; 25; 50; 75 and 100% and Thin Layer Chromatography (TLC) tests for detection flavonoid compounds, the stationary phase used silica gel 60F 254 and the mobile phase used n-hexane-ethylacetate (1:3). The results of PMALE had antibacterial activity at lower concentrations 6,25% against both of E. coli and S. aureus. PMALE had flavonoid compounds with Rf values of 0.81.

Abstract

The Effectiveness Test of Cucumber Seeds Suspension Formula (Cucumis sativus L) towards Roundworms (Ascaris lumbricoides) as in Vitro Anthelmintic, It has been conducted research to know cucumber seeds powder (Cucumis sativus L) can be made into form of suspension stock and to know stock of cucumber seeds suspension (Cucumis sativus L) can give effect of anthelmintic toward roundwoms Ascaris lumbricoides. The research was conducted by using experimental method. Powder of cucumber seeds (Cucumis sativus L) were made into suspension stock with 3 concentratios such as 5%, 10%, 15% concetrations and suspension without powder of cucumber seeds (Cucumis sativus L) with different addition subtance concentration. Test animal used was roundworm (Ascaris lumbricoides) that was taken by pig?s intestine. Treatment group consisted into five categories such as 125 mg positive control of pamoat pirantel (Combantrin®), negative control of suspension without powder of cucumber seeds (Cucumis sativus L), and category of cucumber seeds suspension (Cucumis sativus.L) as 5%, 10%, and 15%. Each group was soaked with 40 ml test ingredient into petri dish. Then it was observed and writte the number of died worms in every minute. 100% observation of died worm was group of positive control at 405 minutes, negative control at 1710 minutes, 5% concetration of cucumber seeds suspension stock at 540 minutes, 10%concentration at 465 minutes and 15% concentration at 420 minutes. The result of research showed that cucumber seeds (Cucumis sativus L) can be made into suspension stock and effective as anhelmintic toward roundworms Ascaris lumbricoides.

Abstract

Rheumatoid arthritis is an chronic autoimmune disease, which is characterized by inflammation and degradation of the joints and cartilages. Rheumatoid therapy is currently using immunosuppressant drugs, nonsteroidal anti-inflammatory drugs and corticosteroids. Treatment of chronic rheumatoid can trigger side effects. Syzygium polyanthum leaves contain alkaloid compounds, flavonoids, saponins, tannins and essential oils consist of citral and eugenol which are thought to be potential as antirheumatoid. This study aims to determine the effect antirheumatoid arthritis and effective dose of ethanol extracts of  Syzygium polyanthum  leaf. This study used 15 male rats. Treatment started by measuring foot volume using a pletismometer and then inducting CFA intraplantary as much as 0,1 mL on day 1 and left until day 16th. On the day 17th a foot volume and arthritis index was measured and then treated until the day 30th. The animals were divided into 5 groups: negative control (Na. CMC 1%), positive control (methylprednisolone dose 1.646 mg / kg BW), ethanol extractof Syzygium polyanthum  leaf at a dose of 75 mg / kg, 150 mg / kg and 300 mg / kg. On the day 31st, foot volume and arthritis index measurements were taken again. The results showed a decrease in foot volume and arthritis index after given of the extract. Foot volume difference data were analyzed using the One Way Anova test followed by the Post Hoc LSD test. The difference between the arthritis index data was analyzed using Kruskal-Wallis followed by the Mann-Whitney test. Statistical results show that positive control has the same effect as extract (P> 0.05). Based on the results of statistical analysis of inflammation of the foot volume and the arthritis index shows that the ethanol extract of Syzygium polyanthum  leaf had an effect as an antirheumatoid arthritis

Abstract

Angiogenesis is a process of forming new capillaries found in the blood vessel system. Angiogenesis plays an important role in tumor growth and metastasis which includes several biological processes. Cancer cells require a supply of nutrients through the blood vessels to grow and develop so that the cancer cells secrete growth substances to stimulate the formation of new blood vessels to meet their nutritional supply. Robusta coffee beans (C.robusta) contain compounds that function as strong antioxidants namely Chlorogenic acid which is a polyphenol group compound that is thought to have antiangiogenesis effects. This study aims to look at the antiangiogenesis potential of robusta coffee bean ethanol extract (C.robusta) with the Chorio Allantoic Membrane (CAM) method and determine the concentration of robusta coffee bean extract (C.robusta) which has the potential as an antiangiogenesis. Antiangiogenesis test using the Chorio Allantoic Membrane (CAM) method using 9-day-old chicken eggs. In this study eggs were divided into 4 groups, group I was the untreated group, groups II, III, IV were groups that were given ethanol extract of robusta coffee beans (C.robusta) with concentrations of 6000 ?g/ml, 5500 µg/ml respectively. and 5000 µg/ml. The eggs are re-incubated for 72 hours. In this study, a macroscopic semi-quantitative score was observed. The score data obtained were analyzed statistically by the One Way Anova test. Based on the Saphiro Wilk normality test results, the results of observations of antiangiogenesis in ethanol extracts of Robusta (Coffea robusta) concentrations of 6000 ?g/ml, 5500 ?g/ml, and 5000 ?g/ml obtained a significance value of 0.248 (p>0.05). Continuing the One Way Anova test in the antiangiogenesis test obtained a significance value of 0.007 (p<0.05). Then proceed with the Post Hoc Test. In the test group the concentration of 6000 µg/ml, 5500 µg/ml, and 5000 µg/ml had a value of p>0.05 which means that there were no differences between groups in inhibiting angiogenesis. Based on the results of the study it can be concluded that the ethanol extract of robusta coffee beans (C. robusta) 6000 ?g/ml, 5500 ?g/ml, and 5000 ?g/ml has antiangiogenesis effect on Chorio Allantoic Membrane (CAM) of chicken embryo.

Abstract

Nigella plant genus has potential as anti-HIV. One species of Nigella, Nigella sativa has been reported to have HIV-1 protease enzyme inhibitory activity. This research aims to determine the compounds of the Nigella genus that have activity as HIV-1 protease enzyme inhibitory activity through molecular docking method by Autodock Vina and to compare interaction stability through molecular dynamics simulations by AMBER. The metabolite of the Nigella genus was obtained from the KnapSack website, and enzyme model was obtained from the Protein Data Bank (3NU3). The results of molecular docking found the lowest affinity energy of Nigella compound is Nigellidine 4-O-sulfite (-13.4 kcal/mol). Meanwhile, the affinity energy of the ligand native (Amprenavir) was -12.1 kcal/mol. The lowest affinity energy of Nigellidine 4-O-sulfite might be predicted to have potency as an HIV-1 Protease inhibitor. Molecular dynamics simulation showed Root Mean Square Fluctuation (RMSF) value of Nigellidine 4-O-sulfite with the amino acid active site is 0.4064 Å for ASP:25 and 0.5667 Å for ASP: 125. Whereas RMSF ligand native with the amino acid active site, ASP: 25 is 0.3647 Å and ASP: 125 is 0.3639 Å. The higher RMSF value of Nigellidine 4-O-sulfite describes the lower interaction stability than the ligand native.

Abstract

An effort to improve the solubility of albendazole (ABZ), an anthelmintic drug has been successfully carried out through the formation of multicomponent crystal with dl-malic acid (MAL). Construction of phase solubility curve of ABZ in MAL solution and crystal morphological observations after recrystallization in the acetone-ethanol (9:1) mixture were performed for initial prediction of multicomponent crystal formation. ABZ-MAL multicomponent crystal was prepared by wet grinding or also known as solvent-drop grinding (SDG) with acetone-ethanol (9:1) mixture as a solvent followed by characterization of the multicomponent crystal formation by powder X-ray diffraction and Fourier transform infrared (FTIR) methods. The solubility of ABZ-MAL multicomponent crystal was tested in water at ambient temperature and in pH 1.2, 4.5 and 6.8 of buffered solutions at 37°C. The phase solubility curve of the ABZ in the MAL solution showed type Bs. The ABZ-MAL mixture has a different crystalline morphology than pure ABZ and MAL after recrystallization in the acetone-ethanol mixture (9:1). The powder X-ray diffraction pattern and the FTIR spectrum of ABZ-MAL from SDG different from intact ABZ and MAL powder X-ray diffraction patterns and these results can indicate the ABZ-MAL multicomponent crystal formation. The ABZ-MAL multicomponent crystal has better solubility than pure ABZ in all media used. These results can be concluded that ABZ-MAL multicomponent crystal can be prepared by solvent-drop grinding method with acetone-ethanol (9:1) mixture as a solvent and can increase the solubility of albendazole.

Abstract

Prevalensi diabetes melitus berdasarkan hasil Riset Kesehatan Dasar yang tertinggi di Indonesia terdapat di provinsi DKI Jakarta. Ketidakpatuhan berobat pasien diabetes melitus tipe 2 merupakan salah satu faktor yang mengakibatkan rendahnya kontrol glukosa darah dan meningkatnya resiko komplikasi. Penelitian ini dilakukan untuk mengetahui profil tingkat kepatuhan penggunaan obat di Puskesmas Kecamatan Makasar  Jakarta Timur dan faktor-faktor yang mempengaruhi kepatuhan penggunaan obat pada pasien diabetes melitus tipe 2. Penelitian ini berdesain cross sectional yang dilaksanakan pada April 2019 sampai Juni 2019 dengan total 175 responden diabetes melitus tipe 2. Pengumpulan data dilakukan dengan teknik wawancara terstruktur menggunakan kuesioner kepatuhan MMAS-8. Data diolah dengan menggunakan software statistik dan dianalisa menggunakan analisa distribusi frekuensi dan uji chi square. Hasil penelitian menunjukkan bahwa 71 responden (40,6%) memiliki tingkat kepatuhan sedang dalam menggunakan obat antidiabetes. Alasan utama ketidakpatuhan responden adalah bosan (43,6%). Ada hubungan bermakna antara tingkat kepatuhan dan hasil tes gula darah. Untuk mengetahui lebih dalam hubungan antara tingkat kepatuhan dengan hasil tes gula darah maka perlu adanya penelitian lanjutan tentang hubungan studi prospektif hasil tes gula darah dengan tingkat kepatuhan.
