Abstract

The steroidal contraceptive agents are highly predictable in preventing pregnancy, however, all currently used synthetic progestogens have certain potential side effects. These side effects may be due to either the direct actions of the synthetic steroids or the indirect actions through the womens own endocrine system. The objective of the study is to evaluate potential alterations of the endocrine functions in women using Norplant compared to injectable contraceptive method. The study was conducted in a cohort study by recruiting 25 women in each group receiving either Norplant or Depoprovera contraceptive method voluntarily. Six months after entering the study 10 ml of blood was taken from the cubits] vein and follicle stimulating hormone (FSH) , luteini zing hormone (LH), prolactine (PRL), progesterone (P), oestradiol (E2), total testosterone (r), free testosterone (FT) and sex hormone binding globulin (SHBG) were measured using radioimmunoassay. The mean of SHBG level in Norplant (29.44 ± 9.85 nmol/l) is statistically, lower (p421.001) compared to Depoprovera acceptors (50.64 ± 21.29 nmol/l), however, they were still in the normal range. The means of total testosterone and free testosterone level were higher in Norplant group, however, they were not statistically different (p = 0,391 and 0.182 respectively). No androgenicity symptoms were clinically found in both groups. Either Norplant or Depoprovera lowering the secretion of progestogen and oestrogen, indicated that the alterations of endocrine function occured more frequently in the level of target organ (ovary) rather than in the level of hypophysis or hypothalamus. These findings were also confirmed by the mean levels of FSH, LH and prolactine which were also in the normal range. It is concluded, that there were no potential alterations of the endocrine functions in women using either Norplant or Deprovera especially in androgenicity. The influence of both progestogen contraceptive methods were greater on the level of target organ.Key words : contraception-Norplant-Depoprovera -radioimmuno assay-blood

Abstract

Key Words : atherosclerosis - lipid - lipoprotein - apolipoprotein - coronary heart disease

Abstract

Rurnah Sakit Umum Pusat Dr: Sardjito, Yogyakarta.

Abstract

The objective of this study is to provide evidence on whether the subfomical organ is a neuroendocrine gland, by which the neurosecretion process to be carried out in this organ. This investigation involved the use of light microscopy by using an immunohistochemistry as well as histochemistry methods and transmission electron microscopy. Eighty adult male and female albino rats of the Lembaga Makanan Rakyat strain (Jakarta) weighing between 150-250 g, and 9 rats of Wistar strain, weighing between 200-300 g, were studied.According to morphology and by using immunohistochemistry and histochemistry methods, the results revealed that the subfomical organ was not a neuroendocrine gland that secreted a neurohormone or neurosecretory substances through a neurosecretion process.Key Words: subfomical organ-neurosecretion process - transmission electron microscopy - inununohistochemistry technics - histochemistry technics.

Abstract

The humoral IgA response to bacterial polysaccharide antigen in rats was investigated in vivo. Rats were immunized with pneumococcal polysaccharide type-3 (PPS-3) via different routes, i.e. in the Peyers patch (iPP), in the colon (I.C.), peritoneal cavity (LP.), and intravenous (I.V.). The development of specific antibody-forming cells (AFC) and their isotypes in the intestinal mucosa, gut-associated lymphoid tissue. (GALT), mesenteric lymph nodes (MLN) and spleen were studied by immunohistochemistry. The serum antibody levels were measured by enzyme-linked " lll unosorbent assay (ELISA).The results showed that iPP immunization evoked high numbers of anti-PPS-3 AFC of the IgA isotype in the mucosa of the small intestine and in the Peyers patch. On the contrary, the I.C. route did not elicit a mucosal response, though a few AFC were found in the MLN and spleen. Following I.P. priming, a specific IgA response was found, especially in the MLN and spleen, and low response was detected in the villi. A high response was found in the parathymic lymph nodes (PTLN). The I.V. immunization gave rise to the development of AFC in the spleen. particularly of the 1gM isotype. There was no mucosal responses to PPS-3 antigen in the colon arise irrespective of the route of immunization.Key-words : IgA-colon-antibody-forming cells- pneumococcal polysaccharide antigen -immunohistochemistry
