Abstract

Tree seed  for planting program are not always available in sufficient quantity in right time. Seed storage efforts at the optimum conditions for plant propagation material are needed to meet those needs. Referring to the theory, this experiment was carried out to investigate the appropriate room of starage, container of storage, and periods of storage to maintain the germination of kesambi seeds. Experimental design used was split plot design with three factors such as room of storage (ambient room, temperature 27-30 0C and relative humidity 60-70%, air conditioned room, temperature 18-20 OC). Container of storage (plastic bag andblacu bag)and periods of storage (1 month , 3 months, and 5 months) . Every unit of treatment contain 25 seed and replication 4 times. The results of this research indicated that the viability of the kesambi seed can be maintained for 3  month using blacu bag as sinby an average g blacu bag as container with    the germanition percentage and  speed of ngeminiton by an average of (75% and 4.14% and 4.14 KN /etmal ) and the moisture content 7.79%

Abstract

Pongamia pinnata is an arboreal legume with high conservation value (Conference of The Parties to The Convention on Biological Diversity at the IX/2 nd  on plants of high conservation volue criteria). Biotechnology breeding program with the selection  of legume crops that have a high nitrogen use efficiency becomes an important issue to improve productivity and quality of P. pinnata.Thus the important symbiotic effectiveness test done to improve the ability of N fixation as the main element forming a plant tissues . The purpose of this study was to obtain Rhizobium strain from different P. pinannta habitats which has the highest effectiveness compared with strain Rhizobium sp. Commercial (USDA 122) and controls to P. pinnata seedlings. The parameters used are the number of nodules, plant height, root length and diameter . From this activity obtained 3 strains of Ambon and 2 strains of Banyuwangi .  Strains selected on the basis of colony growth speed of Rhizobium sp.in vitro.The average number of nodules, plant height,  root length and diameter of P. pinnata that are inoculated with strain  AMI. AM2, AM3, Bal and Ba2 from seram, Ambon higher than from Baluran , Banyuwangi. On average, the highest number of nodules occurred in simbiose of P.  pinnata) Ambon  Rhizobium AM3  (50 ± 0,66) eguels P. pinnata Ambon –Rhizobium USDA 122 (50 ± 0,88); the best plant height occurred in P. pinnata Ambon-Rhizobium Ba2 (72cm ± 1,14) ; the best root length  (25 cm ± 0.33) and the best stem diameter (4.0 mm  ±  0.33) occurred in P. pinnata Ambon-Rhizobium AM2. The highest symbiose effectiveness value of P. pinnata-Rhizobium occureed in P. pinnata origin from Ambon with Rhizobium strain AM3 (32,45).

Abstract

Observation of seed production and hybrid seed yield were assessed in hybridization orchard of Acacia mangium x Acacia auriculiformis on the first and second flowering season . The hybridization orchard was established in 2006 using 6 selected plus trees from the first-generation of progeny test of A. mangium and 12 selected plus trees from the first-generation of progeny test of A.auriculiformis. Crosses were conducted using 2 spikes for each cross. Based on synchronization in flowering time, 6 and 17 crossing combinations were made resulting in 23 and 89 hybrid families in the first and second flowering seasons respectively. The average  number of pods and seeds in the first flowering season were 1.9 pods/spike and 2.0 soundseed/pod respectively, while those in the second flowering time were increased to 2.6 pods/spike and 2.4 soundseeds/pot respectively. The percentage of poorseeds almost the same on the first and second flowering seasons, namely 28 .7%  and 32.0% respectively. Verification using SCAR marker indicated that almost all of collected seeds were hybrid seeds.

Abstract

FTA  card offers a simple and fast method for retrieval of DNA samples at room temperature and storage of DNA in the short and long term. This will fasiliate the detection and identification of plant pathogens rapidly; increasing the number of samples can be collected, stored and transported in the field, especially from remote locations. The purpose of this study is to investigate  the suitability of FTA cards as a new method for sampling DNA from multiple infected of  plant tissues such as leaves, bark and roots. The results of this study shows that the FTA card suitable for use as one method of smpling because its quick and accurate. Series of PCR using ITSIF/ITS4 and ITS3/ITS4 primer pairs were done to detect the fungal DNA from the FTA card Nested PCR has been done using species – specific primers to identify the fungus without cloning. Optimization of methods to maximize the use of FTA cards as a method of sampling and DNA extraction which is simple, fast, accurate and safe for the environment is still needed.

Abstract

A  research was done at natural tropical forest in forest concession of PT : Suka Jaya Makmur (SJM).Ketapang. West Kalimantan . The objective of the study is germplasm area in natural Forest. The aims of this floristic study were to investigate the potensial and distribution of tengkawang tree species. Tengkawang kernel is one of the fomous Non –Wood Forest Product (NWFP), it is rescources of on importance vegetable fat. The inventory was conducted using line plot method in each area of 26 Hectares. 130 sample plots were made up systematically within 13 line plot. The sample plot had a square shape with nested sampling area measured 20 mx 20 m for tree stage, 10 m x 10 m for pole stage, 5 m x 5 m for sapling stage and 2 m x 2 m for seeding stage. The result reveal that the germplasm consisted of 75 species were dominated by industralwood species . The germpalsm are was dominated by Shorea leprosula with importance value (31.01%). Eugenia cymosa (26.32%).Litsea Firma (18.11%) and Shorea laevis (16.02%) respectively . Some tengkawang trees were found in the study area with important value were varied were Shorea pinnanga (9.43%) . Shorea macrophylla (4.11%). Shorea macistopterix (0.63%) respectively but the famous of tengkawang tree (Shorea stenoptera)was not found in the study area. Therefore, block 35 & 36 PT SJM germplasm area unrepresentative for tengkawang genetic concervation plat.
