Abstract

AbstrakPenelitian Keanekaragaman Coleoptera dan Araneida di ekosistem Perkebunan Kakao Rakyat Kabupaten Kolaka Timur telah dilaksanakan pada bulan Oktober 2014 – Juni  2015. Tujuannya untuk mengevaluasi kepadatan relatif dan keanekaragaman Coleoptera dan Araneida permukaan tanah. Pengambilan sampel dilakukan di empat kondisi sanitasi kebun yang berbeda, yaitu tanpa sanitasi (TS), sanitasi bersih (SB), sanitasi pohon (SP), dan sanitasi lahan/seresah (SS), dengan luas masing-masing kebun sebesar satu hektar. Penelitian bersifat deskriptif - kuantitatif, dengan metode purposive, dan pengambilan sampel melalui penangkapan langsung dengan tangan, umpan serangga, dan perangkap. Hasilnya, dari ordo Coleoptera diperoleh enam famili yaitu Scarabaeidae, Scolytidae, Staphylinidae, Nitidulidae, Hydrophilidae, Dermestidae dan dari ordo Araneida juga diperoleh enam famili yaitu Salticidae, Thomisidae, Araneidae, Lycosidae, Arachnidae, dan Oxyopidae. Kepadatan relatif tertinggi ordo Coleoptera terdapat di kebun sanitasi bersih, yaitu famili Scolytidae 34,20%, dengan indeks keragaman -0,16. Kepadatan relatif tertinggi dari ordo Araneida juga terdapat di kebun sanitasi bersih yaitu famili Lycosidae 5,20% dengan indeks keragaman -0,018. Kata kunci : Kepadatan relatif, Indeks keanekaragaman, Coleoptera, Araneida  AbstractThe research diversity of Coleoptera and Araneida on the ground of Cocoa smallholder Plantation at East Kolaka was held in October 2014 - June 2015. The objective is to evaluate the relative density and diversity of Coleoptera and ground Araneida. Sampling was carried out in four different gardens sanitary conditions, i.e. without sanitation (TS), clean sanitary (SB), tree sanitary (SP), and land sanitary (SS) with each garden area of one hectare. The study was descriptive, using purposive sampling through direct capture by hand, insect baits and traps. The result, of the order coleoptera obtained six families (Scarabaeidae, Scolytidae, Staphylinidae, Nitidulidae, hydrophilidae, Dermestidae) and six families are Salticidae, Thomisidae, Araneidae, Lycosidae, Arachnidae, dan Oxyopidae of the order Araneida. In the cocoa field with clean sanitation, the highest relative density of the order coleoptera exhibited by the family Scolytidae 34.20%, with a diversity index -0.16 and the highest relative density of the order araneida exhibited by the familiy Lycosidae 5.20% with a diversity index -0.018. Keywords: Relative density, Diversity index, Coleoptera, Araneida

Abstract

Screening of indigenous bacteria from sago waste based on amylolytic and cellulolytic activity was done to obtain bacterial isolate having double activity, i.e. could to hydrolize of starch (amylolytic) and cellulose (cellulolytic). Screening amylolytic and cellulolytic bacteria was done based on amylolytic and cellulolytic activity on agar media. Determination of amylolytic activity on starch agar media was based on the presence of clear zone around the bacterial colony upon flooding with lugol’s iodine solution. Cellulolytic activity was determine based on the presence of clear zone around the bacterial colony on Carboxy methyl cellulose (CMC) agar upon flooding with congo red solution. Presence of a clear zone around the colony indicated starch and cellulose hydrolysis. The diameters of clear zone produced on CMC and starch agar were measured and used as an indication of the amylolytic and cellulolytic activities of the bacteria. The results of the screening based on amylolytic and cellulolytic activity showed that a number of 21 bacterial isolates that having both activities. LCA2 was the bacterial isolate with the highest amylolytic and cellulolytic activity as revealed by the size of clearing zone on both types of agar plates. The diameters of clear zone on starch and CMC agar were 4,98   and 3,65 cm2, respectively. Therefore, LCA2 isolate was bacterial isolate that potent for biconvertion sago hampas into value-added products. Keywords : Bacteria, Amylolytic, Cellulolytic, Sago waste.

Abstract

The aim of this research is to determine the distribution, diversity, evenness and similarity of Cerambycidae bettle based on the total of species and individual at different altitude in mount of Mekongga area in Tinukari village Wawo sub district, north Kolaka regency in South East Sulawesi province. This research was used a survey method, catching Cerambycidae bettle by using sweep net, light trap, malaise trap, artocarpus trap and yellow pan trap by TCBG-LIPI mekongga team. Mounting and identification in laboratory entomology, sector zoology, biology research center Tndonesian of science, Cibinong Bogor, west java. The result show that there are 3 subfamilies of Cerambycidae bettle; Laminae (54 species), Cerambycidae (21 species) and Prioninae (1 species). The highest diversity index, was found at 0-500 asl (H=3,389) and the evennes high category was found at 1000-1500 asl (H=1). The highest similarity index of species was found at 0-500 and 500-1000 asl (47,225%).   Key words : Cerambycidae Bettle, Diversity, Mekongga Mount.

Abstract

This study aims to characterize sequences of 18S rRNA gene fragment from Pokea (Batissa violacea celebensis Martens, 1897) and its role in differentiated Pokea with other Bivalvia. The method used is a series of PCR (Polymerase Chain Reaction) reaction used to determine the sequence of 18S rRNA gene fragments Pokea. Analysis of the data using program NCBI (National Center for Biotechnology Information), Clustal X, Phydit (Phylogenetik editor), and TreeViewX for characterization 18S rRNA gene sequence to construct a phylogenetik tree of Pokea. The results showed that character of 18S rRNA gene fragments Pokea, namely: size 827 bp, including the family Corbicullidae because it has the closest kinship with Corbicula fluminea, as well as having restriction enzyme sites XhoI, PstI, BamHI, and DraI.   Keywords: Morphology, characterization, 18S rRNA gene, Pokea (Batissa Violacea celebensis Martens, 1897).

Abstract

Research on the Study and utilization of Traditional medicine plants by Moronene ethnic in the Rau-Rau village Southeast Sulawesi were aims to: 1) inventory these kinds of medicinal plants are utilized including the identification of scientific and local names, 2) study the knowledge society in the utilization of plants as medicine, metods processing and the efficacy, 3) study the  knowledge kinds of disease and how to use of medicinal plants. With methods survey exploration conducted interviews to expert treatment (sandro) and the community. There are 51  spesies plant  in 27 family growing in the backyard, in the gardens and in the forests around the settlement. The organ  medicinal plant species (leaves, stems, bark, rhizome, tuber, fruit and SAP) are uses for tradisional medicines to cure about 36 kids of diseases. Abstract

The aim of this research was to know the dragonfly (Odonata) spesies around River and Moramo Swamp, Sumber Sari Village, Moramo District, South Konawe Regency, South-East Sulawesi. Dragonfly captured on three site namely river, swamp I and swamp II/ Moramo swamp. This research used descriptive method. There are 28 species of dragonfly which are include of 8 family namely Lindeniidae, Libellulidae, Megapodagrionidae, Lestidae, Coenagrionidae, Calopterygidae, Chlorocyphidae and Plactynemididae. Suborder Epiprocta found 13,33% family and 50% suborder Zygoptera of all families. There are 12 species in river, 15 species in swamp I and 13 species in swamp II/Moramo swamp. The Calopterygidae, Megapodagrionidae and Platycnemididae only found in river, whereas Lestidae (Lestes concinus Hagen) found in swamp only. The dragonfly spesies found in river were diffirent from swamp I and swamp II/Moramo swamp.   Key word:  Dragonfly, Odonata, Moramo Swamp, River,Stocktaking

Abstract

Isolation and identification of mycorrhiza fungal that associated with Spathoglottis plicata Blumeand Phalaenopsis amabilis L. orchid root have be done. The descriptive research consist of threesteps that were isolation, purification and identification. Slice of roots were inoculated on PotatoDextro Agar (PDA) media for 2 – 5 days. The fungal colony were purified then identified basedon colony and morphology characters. The observed parameter were fungal colony, hypha andspore type. The result showed that there were 3 isolate found from root of terrestrial orchidSpathoglottis plicata Blume i.e SA1.1 was Chaetoium belonging to Ascomycetes class; SA2.2was Beltrania belonging to Ascomycetes class too and SA2.3 was Rhizoctonia belonging toDeuteromycetes class. Meanwhile the identification result of mycorrhiza fungal from root ofepiphytic orchid Phalaenopsis amabilis L. showed that 2 isolate i.e PA1.2 and PA1.3 both of themRhizoctonia belonging to Deuteromycetes class.Keywords : orchid mycorrhiza, Spathoglottis plicata Blume, Phalaenopsis amabilis L., orchid root

Abstract

The use of plants as traditional medicine has been known since long time ago by people in Lipu. This process has been inherited from one to next generation, nevertheless. nowdays, it has started to be extincted. So, it should be better to indentify the species of plant used as traditional medicine by people in Lipu. The research aims to investigate thekinds of plant which uses as traditional medicine. The part of plant used and how to process and the adventage of the plant. The research used explorative survey method. The result of the research there are 54 species from 31 family of plants used as traditional medicine by the people. The parts used of plant are leaf, stem/bark, root, flower, fruit, seed, tuber,rhizome and sap. The processing of the plants by refine, boil, squeeze, parch, pour boiling water, rasp and without processing. And the ability of the plants are can treat kinds of disease.

Abstract

Abstrak Penelitian ini bertujuan untuk mengetahui kandungan karbon serasah daun Kuma (Palaquium luzoniense Vid.) di kawasan Hutan Lindung Nanga-Nanga Papalia Kota Kendari Sulawesi Tenggara. Penelitian ini merupakan penelitian eksplorasi dengan menentukan 3 stasiun pengambilan sampel berdasarkan pengamatan ketersediaan tumbuhan Kuma (Palaquium luzoniense Vid.) dengan ketinggian yang berbeda yakni Titik I (ketinggian ±223 mpdL), Titik II (ketinggian ±246 mdpL) dan Titik III (ketinggian ±276 mdpL). Pengambilan serasah daun Kuma (Palaquium luzoniense Vid.) dilakukan secara purposive sampling. Kandungan karbon diperoleh dengan cara mengalikan nilai kadar karbon dan nilai biomassa serasah daun. Hasil perhitungan kandungan karbon sebesar 5501,565 gr/m2/bulan. Kata kunci: Kuma (Palaquium luzoniense Vid.), Serasah Daun, Kandungan Karbon, Hutan Lindung Nanga-Nanga   Abstract This study aimed to determine the carbon content of Kuma leaf litter (Palaquium luzoniense Vid.) in Nanga-Nanga Papalia Protected Forest area at Kendari Southeast Sulawesi. This study was an exploration research by determined three station sampling based on observation of availability Kuma (Palaquium luzoniense Vid.) species  with differet hight s.i.e first point (altitude of ± 223 mpdL), second point (altitude of ± 246 mdpL) and third point (altitude ± 276 mdpL). Sampling of  Kuma leaf litter (Palaquium luzoniense Vid.) used purposive sampling. The carbon content is obtained by multiplying %carbon content and biomass values. Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui struktur populasi dan kandungan logam berat gastropoda (Telescopium telescopium) di Taman Nasional Rawa Aopa Watumohai. Penelitian ini dilaksanakan pada bulan Januari sampai Maret 2018. Lokasi pengambilan sampel penelitian terletak pada mangrove yang tumbuh di Sungai Lanowulu dan Roraya di TNRAW. Penelitian ini menggunakan metode purposive sampel. Penelitian ini menggunakan plot kuadrat ukuran 1 m2 diletakkan di setiap stasiun. Jumlah individu dari T. telescopium dihitung berat dan panjangnya. Selain itu, kandungan logam berat Timbal (Pb) dan Cadmium (Cd) di dalam daging T. telescopium dihitung menggunakan Atomic Absorption Spectroscopy (AAS) di Laboratorium Fakultas Perikanan dan Ilmu Kelautan Universitas Halu Oleo. Struktur populasi T. telescopium ditentukan dengan analisis histogram, distribusi ukurannya ditentukan dengan analisis koefisien variasi (CV). Hasil penelitian menunjukkan struktur populasi gastropoda membentuk kurva normal dengan ukuran kecil. Kandungan logam Pb dan Cd di dalam sedimen dan air laut dari Sungai Roraya lebih tinggi secara signifikan dibandingkan dengan Sungai Lanowulu, disisi lain, kandungan logam berat Pb dan Cd dalam daging T. telescopium tidak berbeda signifikan. Dengan demikian, kandungan logam berat Pb dan Cd pada air laut, sedimen dan daging T. telescopium di sungai Lanowulu dan sungai Roraya berada di atas baku mutu sesuai dengan Kepmen LH. No. 51. 2004. Kata kunci: Gastropoda, Mangrove, Logam Berat, TNRAW. ABSTRACTThis study aims todetermine the population structure and heavy metals contents of gastropods (Telescopiumtelescopium) living in RawaAopaWatumohai National RAWN Park. This research was conducted from January until March 2018. The sampling location of research was in mangroves grown along the Lanowulu and Roraya rives at RAWN Park. This research uses purposive sampling method.  This study used the plot quadrate of 1 m2 widethat placed in each station. The number of individuals of.T. telecopium was counted, while their fresh weight and length of T.telescopiumwere measured. In addition, the content of heavy metals of Lead (Pb) and Cadmium (Cd) into meat of T. telescopium were measured by using Atomic Absorption spectroscopy (AAS) in Laboratory at Faculty of Fisheries andMarine Science at Halu Oleo University. The population structure of T. telescopium was determined by histrogram analysis, its size distribution was determined by coefficient of variation (CV) analysis. The results showed that population structure of gastropods was normal curve with small ranges of size distribution. The content of Pb and Cd into sediment and sea water of Rorayariver were significantly higher as compared than that in Lanowulu river. On the other hand, the content of heavy metals Pb and Cd into meat of T. telescopium was not significantly different. However, the heavy metals contents of Pb and Cd into sea water, sediment and meat of T. telescopiumboth in Lanowuluand Roraya riverswere above the quality standard according to Ministry of Environment No. 51/LH/2004. Keywords : Gastropods, mangroves, Lanowulu and Roraya rivers, heavy metals, Rawa Aopa Watumohai National Park
