Abstract

This research was conducted in order to synthesize two derivatives of C-9154 antibiotic from vanillin and to evaluate their biological activities. The synthesis of these C-9154 derivatives was performed through alkylation of vanillin, preparation of the corresponding benzaldoxim, reduction of this benzaldoxim, treatment of the resulting substituted benzilamine with maleic anhydride and esterification of this C-9154 antibiotic derivative. The alkylation of vanillin was carried out using diethylsulfate and NaOH as the base to give 89% yield of ethyl vanillin. The oxim synthesis from ethyl vanillin was performed using hydroxylamine hydrochloride under alkaline condition at 50oC for 2 hours to yield 89% 4-ethoxy-3-methoxybenzaldoxim. Reduction of this benzaldoxim with metallic Na in absolute ethanol afforded 82% yield of 4-ethoxy-3-methoxybenzylamine. This benzylamine derivative was then reacted with maleic anhydride in benzene to give the acid-form of C-9154 antibiotic derivative (6) in 74% yield. Esterification of this acid conducted Jumina using absolute ethanol in the presence of concentrated sulfuric acid at reflux for 4 hours gave the ethyl ester-form of C-9154 antibiotic derivative (7) in 87% yield. Antimicrobial activity evaluation was conducted using Staphyllococcus aureus and Eschericia coli. Based on the minimum inhibition concentration (MIC) data determined using aqueous methanol as the reference, it was observed that the acid-form (6) and the ethyl ester-form (7) of C-9154 antibiotic derivative obtained respectively gave MIC values of 2000-3000 g/ml and 500-1000 g/ml respectively either towards Staphyllococcus aureus or Eschericia coli.Key words: antibiotics, vanillin, oxim, benzylamine, C-9154 derivative, antimicrobe.

Abstract

In order to apply a standardization method on herbal medicines or pharmaceuticals, a thin layer chromatographic-densitometry was used to determine apigenin as the active constituent of celery extract formulated in a capsule dosage form. Silica gel GF 254 and chloroform – methanol – water (70 – 30 – 6,5 v/v) were used as stationary and mobile phases, respectively. The areas of apigenin were evaluated at 340 mm using a TLC-Scanner.The limit of detetion was 3 g and the accuracy was 89,6% after first extraction prosedure applied on celery capsules. The apigenin content was 0,017 %.Key Words : Apium graveolens L., apigenin, TLC – densitometry.

Abstract

Fatty acids are part of fatty oils found in plants, animals and other organisms. Fatty oils are useful for human being, and they are also used as drug solvents, soaps, cosmetics and paint. Plant seeds usually contain fatty oil, protein and carbohydrate, useful for plants growth Durian seeds were just as common rubbish, therefore an investigation of fatty acids found in durian seeds were might be beneficial. Durian seeds were collected, dried, powdered, and was extracted with petroleum ether using Soxhlet apparatus. Physical and chemical parameters of Fatty oil i.e colour, acid value, saponification value, iodine value, and peroxide value were determined. Methyl ester of fatty acid composition produced by fatty oil transesterification was analyzed using a gas chromatography. The chemical parametrs of fatty oil are found to be: acid value (19,4 ± 0,17 ), saponification value (190,65 ± 0,84) , iodinevalue (81,7 ± 0,53) , and peroxide value (3,93 ± 0,12). Physical properties of fatty oil were : concentration of fatty oil was 6,12 ± 0,16 (%), w/w of dried seeds; transparenty yellow and durian odoured, the water content of durian seeds was 12,17 %. The composition of fatty acid containing in fatty oil was unsaturated fatty acid: oleic acid (14,95%) and linoleic acid (12,46%), and saturated fatty acid which is consist of stearic acid (45,84%) and palmitic acid, ( 26,75 %), relatively to pure stearic acid (E.Merck, pro analysis).Key Word: Durio zibethinus Murr, durian seed, fatty acids, identification

Abstract

Paracetamol is the most material drug used in Indonesia. But it’s difficull in processing of the tablet because of capping tendency. The effect of paracetamol reprecipitation by using etanol solven during fabrication process to physical characteristic of tablets of has been studied, and to know the effect of paracetamol. In this studies made five formulas : paracetamol was sprayed by etanol, paracetamol was added filler then sprayed by etanol, paracetamol was added disintegrant then sprayed by elanol, granulating the paracetamol then was sprayed by etanol and paracetamol was without sprayed by etanol. Results showed that granules had been sprayed by etanol before the granulation have characteristic of flow materials was better, the friability of the granules decreased and compressibility of the granules was better. Hardness of tablets relatively increased, the friability of the tablets relative decreased and the disintegration time of tabltets increased. Generally, reprecipitation of paracetamol during the fabrication process, the physical properties of granuls and tablet were better.Key words : paracetamol, reprecipitation, physical properties of tablets

Abstract

The compound 2,5-bis-(4’-hydroxy-3’-methoxybenzilidin) cyclopentanone or Pentagamavunon-0 (PGV-0) as the new anti-inflamatory drug candidate, is one of curcumin structure modification. Identification of metabolites in faeces could be strengthen the whole information about biotransformation of drug and could be use for studying on PGV-0 metabolites which are excreted through bile. Male Sprague Dawley derived rats were used in all experiments. PGV-0 was given intravenously using dose at 40 mg/kg BB. Then the faeces callected after 24 and 48 hours. The metabolite investigation devided into two parts, etil acetate fraction and water fraction, of faeces. The faeces which is excreted after 24 hours, found was more-polar metabolite according to PGV-0; it has shoft-yellow flourencese and having Rf value = 0,48 on TLC, using silica gel F254 as the stationary phase and etil acetate as the mobile phase. No PGV-0 glucuronide and sulfate metabolites were detectable in faeces, which are excreted on 24 or 48 hours.Key words : Pentagamavunon-0, biotransformation, intravenous, faeces

Abstract

Consumer perception and motivation study to drug information services has been performed in Jogjakarta. The result have shown that consumer motivation based on drive not the respon variable. Abstract

Garcinia is proven to have multi benefit in curing various deseases, some have edible fruit and edible oil. This experiment was a preliminary analysis, including phytochemistry screening, toxicity and antibacterial tests of stem-bark G. celebica and G. tetandra extracts. Ether, ethanol and water extracts of both Garcinia contained 19 different chemical components. Hexane, methanol and water extracts stem-bark of both Garcinia extracts were found to have toxicological effect on Artemisia salina larva with the highest LC50 of G. celebica water extract. Dichloromethane-methanol 1:1 extract of G. celebica acted as growth inhibition on Gram positive and Gram negative bacteri , while the extract of G. tetandra could only inhibit the growth of Gram negative bacterium. Comparing with erythromycin and novobiosin antibiotics, the inhibiton growth activity of the two Garcinia extracts were lower than those of antibiotics.Key words: Garcinia, phytochemical screening, toxicological effect, anti-bactery
