Abstract

Schinus terebinthifolius Raddi is known to have medicinal benefit to cure various deseases. This experiment was conducted to analyse the chemical components of the essential oil of S. terebinthifolius Raddi, because the medicinal effects is strongly related to the chemical components. Chemical components analyses of the essential oil from Schinus terebinthifolius Raddi. was conducted using the combination method of Gas Chromatography and Mass Spectrometry (GCMS). Chromatograms represented 47 components in the essential oil of S. terebinthifolium leaves with 17 major components (> 1% content). They were 3-carene, -pinene, -pinene, -phellandrene (type 1), -phellandrene (type 2), D-limonene, sabinene, p-simene, -simene, -elemene, isocaryophillene, -cubebene, 4aR-(4a,7,8a)-decahydro-4a-methyl-1-methyilene-7-(1-methylethenyl) naphtalena (type 1), 4aR-(4a,7,8a)-decahydro-4a-methyl-1-methylene-7-(1-methylethenyl) naphtalena (type 2), (1,4a,8a)-1,2,4a,5,6,8a)-hexahydro-4,7-dimethyl-1-(1-methylethyl)-naphtalene, -cadinene and 1ar-(1a,4a,7,7a, 7b) -decahydro-1,1,7-trimethyl-4-methylene-1H-sicloprope azulen-7-ol. Abstract

The polycyclic aromatic hydrocarbons (PAHs) are a class of organic compounds, composed two or more fused aromatic (benzene) rings, occurred in the environment due to incomplete combustion of organic matters such as forest fires, volcanic eruption, burning of fossil fuels and incorrect process of meat cooking (grilling, smoking, roasting). Healt concerns are focussed on the metabolite transformation of PAHs, which may have mutagenic, carcinogenic and terratogenic activity. Due to their hydophobicity, it is probable the compounds enter human food chain and accumulated in human lipid tissues. Therefore, environment pollution by PAHs must be considered. PAHs gain entry to human body through several routes including respiration, absorpsion through skin surface ang through food consumption that have been polluted with PAHs. In this study concentrations of PAHs (pyrene, perilene, benzo(a)anthracene, benzo(k)fluorantrene and benzo(a)pyrene) in traditional smoked meat, liquid smoked meat and roasted meat are determined. Determination of PAHs used gas chromatograph with OV 17 2% as a stationary phase, nitrogen as carrier gas and flame ionization detector. The results showed that PAHs concentrations in traditional smoked meat were higher than liquid smoked meat (41.19 ppb and 1.03 – 9.26 ppb). In the roasted meat, lipid concentration and roasting time influence PAHs concentration level.Keywords : polycyclic aromatic hydrocarbon, smoked meat, liquid smoke

Abstract

Kidney stone disease is the second major causal leading to kidney failure. Treatment to the disease could be by surgery, ultravibration and either traditional or modern medecines. Dendrophthoe falcata (L.f) Ettingsh grow, as a parasite, on the mindi (Melia azeradach), a plant has yet been used to cure kidney disorder caused by calcium kidney stone. Dendrophthoe falcata (L.f) Ettingsh leaves powder was extracted by ethanol (70%) and re-extracted by ethyl acetate. The compound content in water and ethyl acetate fractions were analyzed by thin layer chromatography (TLC) using cellulose as the stationary phase and a mixture of butanol: acetic acid: water (4:1:5) as mobile phase. The TLC data showed that water and ethyl acetate fractions contained flavonoid compounds. Kidney stone was analyzed using Infra Red Spectrophotometer to identify its chemical contents. The result showed that the kidney stone consisted of calcium and magnesium carbonate/phosphate. The Fast Neutron Activation analysis was utilized to determine the solubility of kidney stone. The result exhibited that the maximum solubility of kidney stone was achieved in water fraction with the concentration of 20 % or in ethyl acetate fraction with the concentration of 40 %. The solubility of kidney stone in water fraction was higher than that in ethyl acetate fraction.Key word : Kidney stone , Dendrophthoe falcata , Water fraction, Ethyl acetat fraction.

Abstract

Medicinal plants spread out in Indonesia, however only a part of them have been clinically investigated that exhibited therapeutic effecs. This research was to isolate a new compounds in Usnea dasypoga. A new twentyfour-membered macrolactone composed of three units of unknown fatty acid (2R*,3R*,4S*,7S* or 2S* 3S* 4R*, 7R*)-3,7-dihydroxy-2,4-dimethyloctanoic acid forming a C3-symmetrical lactide was isolated from Indonesian lichen Usnea dasypoga Rohl. The result shown that those compound had been confirmed with the spectroscopic data.Keywords: Natural products isolation, three lactide, lichen

Abstract

A brief review on turmeric from the stand-point of chemical content are presented. Recent studies on turmeric showed that at least 7 compounds of curcumin derivatives have been found, including cyclocurcumin the newer curcumin derivative.Keywords : curcumin, curcumin derivatives.

Abstract

Optimation of a method is an obligatory step in order one to obtain a valid and accountable data, especially when one deal with biological samples. Biological samples usually contain minute substances which sometime elicit great errors when someone try to determine its concentration. Therefore, in order to reduce those errors, optimation of a method is mandatory to be carried out. The aim of this experiment was to determine malonic dialdehyde (MDA), a product of lipid peroxidation. The optimation carried out in this experiment include the selection of wavelength of maximum absorbance, the amount of sodium thiobarbiturate, pH, the incubation period, standard curve linearity, limit of detection and quantification, accuracy and precision. The result showed that the optimum conditions of spectrofluorometric method of MDA were as follow: the excitation and emission wavelengths was 522 and 575 nm, respectively, pH of 1 and incubation period of 135 minutes. These conditions yielded correlation coefficient of standard curve of 0.9993, limit detection and quantification of 3.5146 x 10-4 mg/ml and 1.0544 x 10-3 mg/ml, respectively, an average recovery of 85.01 % and variation coefficient of 1.84 %.Key words: optimation, spectrofluorometric, sperm, malonic dialdehyde, MDA

Abstract

The leaves of Dewa (Gynura procumbens (Lour) Merr), is utilized in traditional medicine as anticancer, had been reported to contain flavonoid derivatives and supposed to have cytotoxic properties. Hence, these particular substances might be useful to be developed as anticancer in the future. Research on ethanol extract of Gynura procumbens (Lour) Merr leaves as anticarcinogen has been carried out, using toxicity assay model to vero, myeloma and lymphocyte cells. This research applied varied concentrations and adriamycin as a positive control. Observation to the cell growth inhibition was carried out for 4 days. The result showed that the dominant substance (the third spot of paper chromatogram) inhibit the growth of vero and myelome cells but lymphocyte cell. The LC50 of this substance to myelome, vero and lymphocyte cells were < 82.8 g/ml, 103.86 g /ml and > 2650 g/ml, respectively, whereas the LC50 of adriamycin to the cell lines were < 0.015 g /ml. Therefore, substances obtained from Gynura procumbens (Lour) Merr leaves might be developed against cancer.Key word: Gynura procumbens (Lour) Merr, mieloma

Abstract

Superoxide radical scavenger property of curcumin and its derivatives was investigated using Nitro Blue Tetrazolium (NBT) method. Curcumin was modified at the active methyleen groups with aryl group (4-aryl curcumin), which supposed to be related to its anti-inflamatory activity. The experiment were carried out by mixing curcumin solution at five different concentrations and superoxide radical. After incubating for 60 minutes, NBT2+ (15,29 M) was added and measured at max of 687 nm, so as the curcumin derivatives. The result showed that superoxide radical made bathochromic shift of curcumin. Bathochromic shift was higher when aryl group was attached at active methyleen C-4 of curcumin. This shift made the max of curcumin and its derivatives moving close to max of diformasan (result of NBT2+ reduction by superoxide radical), producing an overlapping spectra. The increasing concentration of curcumin and its derivatives made the increasing max of diformasan absorbance. Thus, the method of NBT2+ reduction, in fact, unable to estimate the scavenger property of curcumin and its derivatives.Key words: curcumin derivatives, superoxide radical
