Abstract

Desensitization therapy on “galah” shrimp type I-allergy have been studied on male Swiss Webster mice. Induction of type I allergy was done by intradermic injection of 30% of “galah” shrimp extracts an allergen twice within 14 days. Evaluation of allergic respons was evaluated based on active cutaneous anaphylactic reaction. Desensitization therapy was done by injecting the allergen intradermically daily in 3 weeks with gradual increased of the doses. Based on tolerance level to the cutaneos anaphylactic reaction, the result showed that allergic mice were cured, while the therapy to nonallergic mice induced the same allergic reaction.Keywords : Type I- allergic reaction, ”galah” shrimp, active cutaneous anaphylactic reaction, desentization.

Abstract

Aqueous extract of the leaves of Nerium indicum Mill. is traditionally used to treat cancer. The extract has been consumed for relatively a long time, consequently major organs are often intoxicated. Therefore, this study was aimed to determine sub chronic toxicity of the aqueous extract on male mice. Toxic symptoms, major organs intoxicated and haematological changes due to treatment of aqueous extract of the leaves of N. indicum are determined.Twenty male mice (185-250 g, strain wistar) are divided into 4 groups [I, treated with aquadest (3 ml/200 g BW); II, treated with water extract (5%, 3ml/200 g BB); III, treated with water extract (10%, 3 ml/200 g BB); and IV, treated with water extract (20%, 3 ml/200 g BB)]. Toxic symptoms are observed after 3 hours of treatment; body weight, haematological measurement, food intake, urine chemistry, SGPT, ureum, and histopathological pictures are also observed. The results of the study do not show any toxic symptoms, the body weight and food intake of the mice increase (I-IV). There are no major organs intoxicated; however a slight increase of leukocyte (WBC, white blood cell) count is clearly showed by group IV.It is concluded that consuming aqueous extract (20%, the highest concentration) of N. indicum for 30 days does not elicit toxic effect in mice. Abstract

Glutathione S-transferase (GST) is a detoxifying enzyme which plays an important role in glutathione conjugation reaction with electrophilic xenobiotic on phase two metabolism. In this research, the effect of oral administration of PGV-0 on rat kidney’s GST activity was investigated.To represent GST activity isolated from rat kidney, 1-chloro-2,4-dinitro benzen (CDNB) was used as a substrate on conjugation with glutathione (GSH) spectrophotometrically. The activity of rat kidney’s GSTs from untreated rat was used as control, Based on data, it can be concluded that PGV-0 showed an inhibitory effect on rat kidney’s GSTs. Abstract

The total protein of Mirabilis jalapa L leaves had the ability to cleave supercoiled DNA and showed toxicity on HeLa and Raji cell-lines. The 30 kD protein (MJ-30) purified by cationic exchange chromatography possessed activites as RIP e.g DNA supercoiled cleavage and RNA N-glycosidase activity. The aim of this study was to prepare pure MJ-30 and to observe the stability of MJ-30 .MJ-30 of M.jalapa L leaves was purified using the combination of ammonium sulphate fractionation and cationic exchange chromatography with NaCl gradient elution. MJ-30 was subjected for its stability to assays against pH, temperature, and storage period.Stability assay showed that MJ-30 is stabil at pH 5-6, then the activity decreased as the pH increased. This protein was stable at 300 – 550C, and the activity decreased when the temperature increased. Storage at 40C, MJ-30 was stable until 12 days but the activity was decreasing. However, at 300C, MJ-30 was stable for 3 days only. Glycerol addition to the MJ-30 solution has made the activity stable for 18 days at 40C and 300C storage.Keyword : Protein MJ-30, Leaves of Mirabilis jalapa L, stability, pH, temperature, storage

Abstract

Solid dispersions of furosemide were prepared by melting and solvent methods using polyethylene glycol (PEG) 6000, talc and PEG – talc as dispersion carrier in order to improve furosemide dissolution. The increase of dissolution and physical properties of its powder was evaluated. Only furosemide solid dispersions with combination of PEG – talc 1 : 3 showed a good physical properties. The assay of the effect of PEG – talc ratio on furosemide dissolution showed that the increase of ratio of PEG – talc increased significantly dissolution rate.Key Word : furosemide, solid dispersion, dissolution

Abstract

Curcuma zedoaria Rosc. rhizomes has been used as a traditional cancer medicine since a long time ago, but the so far researches performed on the pharmacological mechanism were lacking. This present research has been done to determine the effect of ethanolic extract of Curcuma zedoaria Rosc. rhizomes on lung cancer of female mice previously induced by Benzo[a]pyrene (B[a]P).Newborn mice were induced by B[a]P and then were separated between male and female mice. On 30th day after birth, female mice were given the ethanolic extract of Curcuma zedoaria Rosc. rhizomes and divided into five groups. Three groups were given 200, 500 and 750 mg/kgBB extract, a group was given solvent (DMSO) as negative control, and another group was given nothing as positive control.The results of this present research shown that the ethanolic extract of Curcuma zedoaria Rosc. rhizomes has proved to posses inhibition effect on lung cancer growth in female mice, at dose of 250 mg/kgBB (49,63%), 500 mg/kgBB (73,33%) and 750 mg/kgBB (77,78%).Keywords : Newborn Mice, Benzo[a]piren, Curcuma zedoaria Rosc.

Abstract

Ribosome-inactivating protein (RIPs), merupakan sekelompok protein toksik dalam tanaman yang mempunyai anktivitas RNA-glycosidase yang mampu menghambat sintesis protein pada mamalia. RIP mempunyai kemampuan memotong DNA superkoil. Adanya aktivitas tersebut, menjadikan RIP sebagai kandidat yang potensial dalam terapi kanker. Secara tradisional, ekstrak air dari buah P. macrocarpa telah digunakan dalam pengobatan kanker. Untuk itu, tujuan dari penelitian ini adalah untuk mengidentifikasi keberadaan RIP dalam P. macrocarpa, yang kemungkinan merupakan senyawa yang bertanggung jawab dalam aktivitasnya tersebut. Daging buah dan biji P. macrocarpa diektraksi dengan buffer natrium fosfat 5mM pH 7.2 yang mengandung sodium klorid 0,14M. Fraksi protein diperoleh dengan pengendapan menggunakan ammonium sulfat. Aktivitas RIP ditentukan menggunakan metode pemotongan DNA superkoil. Hasil menunjukkan bahwa ekstrak gubal biji P. macrocarpa mempunyai kemampuan memotong DNA superkoil menjadi bentuk nik sirkularnya. Aktivitas tersebut nampak tergantung pada konsentrasi. Aktivitas fraksi protein dari biji terlihat lebih tinggi dibandingkan dengan ekstrak gubal, seperti ditunjukkan dengan munculnya bentuk linier pada konsentrasi protein sebesar 0.3μg/ml. Aktivitas memotong DNA superkoil tersebut tidak muncul pada crude extract dari daging buah. Sehingga dapat disimpulkan bahwa biji P. macrocarpa mengandung protein sejenis RIP. Abstract

PGV-0 is a curcumin derivate, a synthetic compound which may be a candidate of a new drug. This substance has a potent inflammatory effect with a very low toxicity.One of the first step which must be searched for a candidate of drug, is to perform a stability study. There are many degraded drugs causing the adverse reactions. Most of them could be the initiator in forming an antigen at anaphylactic reaction or allergic reaction. Further more some of the degraded drugs are very toxic. So, the study of degradation or the stability investigations of a new drug should be carried out.The PGV-0 stability in a buffer solution at pH 10,0 had been studied by an accelerated temperature method. The temperatures were held at 50°, 55° and 60° C. The intact PGV-0 has been analysed by HPLC. The results then were used to define the shelf-life, the half life and the activation energy of the degradation of PGV-0.It was evident that PGV-0 was unstable in aqueous solution, the shelf-life was only 45.3 hours, the half-life was 299 hours and the activation energy was 14,2 kkal mol-1. Because the PGV-0 was not stable in aqueous solution, it is suggested that this substance should be made into solid dosage forms instead of the liquid dosage forms.Key word : PGV-0, stability, solution
