Abstract

Plant stress due to NaCl compound will have an impact on the reduction of plant growth and yield.  Tolerant varieties are required  to increase plant production in coastal areas since these areas are closely related to salinity.  This study aimed to (1) determine the appropriate NaCl concentration for saline tolerance in maize stress selection, (2) determine the response of maize plant to salinity stress in nutrient culture, and (3) determine the response of new hybrid maize having adaptibility in coastal area.  Two experiments were run from November 2017 until May 2018. The first experiment was conducted in a greenhouse at the Laboratory of Agronomy, Faculty of Agriculture University of Bengkulu  to determine the tolerance response of 25 new hybrid maize to salinity in nutrient culture.  The second experiment was conducted on the coastal areas in Beringin Raya Village, Muarabangkahulu District, Bengkulu City to determine the response of new hybrid maize having adaptibility in a coastal areas.  The concentration of  NaCl at which level the LC50 was determined was found to be in 150 mM. This concentration was then used to test the tolerance of 25 new hybrid maize to NaCl stress.  The hybrids of CT18, CT19, CT22, CT25, CT31, CT33, CT34, CT40, CT47, and  CT50 showed good responses to the concentration treatment  of 150 mM NaCl based on plant fresh weight and shoot dry weight. The hybrids of CT17,CT19, and CT20 showed good adaptibility on coastal area based on the length of cob, diameter of cob, seed weight per cob, and cob weight per plant.  Among those three hybrids, CT17 and CT20 resulted low fresh weight and low shoot dry weight.  The factors other than NaCl in coastal area maybe affect better response. However, CT19 proved as a hybrid which was consistently in a good response to salinity stress both in nutrient culture and in coastal growing area.

Abstract

Ralstonia solanacearum has been known to cause bacterial wilt disease on chili pepper.  Despite many reports on the potential use of endophytic fungi to induce plant resistance, its utilization to suppress bacterial wilt disease of chili has not been widely reported.  The aims of this research was to screen potential endophytic fungi that may increase chili resistance against bacterial wilt disease.  Selection of endophytic fungi was done using in vivo and antibiosis test. Strains of fungi were considered the most potent in suppressing the development of bacterial wilt in chilli were identified. Ten out of 62 isolates of endophytic fungi gave the highest suppression on chilli’s bacterial wilt disease.  Most strains of endophytic fungi were able to suppress the development of bacterial wilt disease, but not always positively correlated to the vegetative and generative growth of chilli. Based on the level of disease intensity and the growth of plants were obtained three strains of endophytic fungi that considered potentially suppress the incidence of bacterial wilt disease.  The three isolates was identified as Fusarium solani f.sp. phaseoli (AC-2.13 and AC-4.4) and Trichoderma asperellum (AC-3.18) using morphology and molecular characters. Although all three selected isolates were able to suppress bacterial wilt disease in this study, but application of F. solani f.sp. phaseoli should be considered in practical use since it is generally known as the causal agent of root rot disease of beans

Abstract

The utilization of starter cultures is vital for dairy fermentation industries. Lactobacillus paracasei M104 and Pediococcus pentosaceus M103 have been isolated from goat milk and were demonstrated to acidify goat milk during fermentation. This study was conducted to compare the physicochemical and microbial qualities of goat milk fermented using these starter cultures either as a single starter or combined in goat milk. The treatments were L. paracasei M104 (Lp) and P. pentosaceus M103 (Pp) as single starter culture fermentation and combined (Lp/Pp) fermentation at a ratio of 1:1 at 6% inoculation (v/v). For control treatment, goat milk was fermented using yogurt starter, i.e., a combination culture of Streptococcus thermophilus and L. bulgaricus (St/Lb) at a ratio of 1:1 at 6% (v/v). The fermentation was performed by incubating the inoculated goat milk at 37°C for 18 h. The pH and degree of acidity were measured every 2 h during the fermentation process, whereas the physicochemical (pH, degree of acidity, lactose, protein, and viscosity) and microbial (total lactic acid bacteria/LAB) qualities were measured after the fermentation process. Data analysis revealed that the different starter cultures had no significant effect (P > 0.05) on the pH and degree of acidity, lactose and protein content, viscosity, and total LAB content. The goat milk fermented in this study had an average pH of 4.41 ± 0.11 and a degree of acidity of 1.26 ± 0.05%, a lactose content of 5.47± 0.80%, a protein content of 6.69± 1.52%, a viscosity of 1217 ± 395 cP, and a total LAB content of 9.57 ± 1.49 log CFU/mL. In conclusion, goat milk fermented using both single and combined starter cultures of L. paracasei M104 and P. pentosaceus M103 exhibited similar physicochemical and microbial qualities and required longer fermentation time compared with the control.

Abstract

DOI: 10.17014/ijog.v6i4.130Activated carbon made from Tanjung Tabalong coal was investigated its absorption capability to organic and inorganic elements in industrial waste. Coal was carbonized at low temperature of 600C to produce semicoke, and then was activated at temperature of 700C with activation time of 120 minutes with water steam flow. The absorption capability of activated carbon to chemical oxygen demand (COD) was performed using 2.5 and 9.0 g activated carbon for 250 ml and 300 ml COD waste respectively. The agitation time of each experiment were 30, 60, and 90 minutes. Atomic absorption spectrophotometer (AAS) was used to analyze the COD waste. The result shows that 2.5 g activated carbon could absorb COD waste ranging from 6.9-67.5 %, while the utilization of 9 g could absorb COD waste ranging from 88.9 - 100 %. The more activated carbon and the longer time of agitation used in this experiment, the more the absorption of COD waste.

Abstract

http://dx.doi.org/10.17014/ijog.vol3no3.20083Gold ore mining at Waluran Subregency, Sukabumi Regency was carried out by an underground mining method, and only rock that contains high enough gold ore would be taken (selective mining). The gold ore from the mining was processed with a direct amalgamation method, so it produced low gold concentrate, but the concentrate of mercury discarded to the river was high enough. Monitoring result of water and sedimentation in Ciliunggunung River in 2004, showed that the river was contamined by mercury (Hg), and the contamination was above the value limit. This happened especially in August 2005 where the Hg content was about 0.218 mg/l on the Ciliunggunung River in CLG.07. The chemical analysis result of heavy metals for the water of the Ciliunggunung River in CLG.07 was known to contain Fe above the standard drinking water criteria, meanwhile Mn, Cu, Pb, and As were still under the maximum drinking water criteria value. To decrease the mercury contamination, it is suggested the process of gold ore from the amalgamation direct method to be changed to the amalgamation indirect method. The indirect gold ore amalgamation process consists of: removing fine particles by washing, grinding ore, and the amalgamation phase. The total impact of the indirect amalgamation method will increase the tying of gold by mercury, so the losing of mercury will be decreased and the gain of gold will be optimum.
