Abstract

The aims of this study is to know the effectiveness of ethanol extract of red betel leaf (Piper crocatum) against mortality of Boophilus microplus Larvae in Vitro, as well as knowing lethal concentration 50 (LC50) and lethal concentration 90 (LC90). Method that used in the research was completely randomized design. There were five treatments and each treatment was done in five replications. This research used ten Boophilus microplus larvae in each treatment for all replication. The observation and recording of mortality was done at 1, 2, 3, 4, and 5 hours. Boophilus microplus larvae were declared dead if there was no movement. The obtained data was analyzed using ANOVA factorial. The result of this research show that ethanol extract of red betel leaf (Piper crocatum) has effectiveness against mortality of Boophlius microplus larvae in Vitro. In the extract 10% concentration, there is effectiveness that almost the same as Noticks®. The higher concentration of the extract, the higher the effectiveness against mortality lavae.In probit analysis show that LC50 achieved by concentration 5,4% or 2.700mg/50ml and LC90 achieved by concentration 11,3% or 5.650mg/50ml.

Abstract

Research of gastrointestinal protozoan in cats at Surabaya are still limited. Protozoa infection of the gastrointestinal tract can cause diarrhea and even zoonosis. This research aimed to identify and determine the prevalence of protozoan in cats in animal hostptal and animal clinic in Surabaya. Ninety  fecal samples were collected from 2 animal clinic and one animal hospital. Samples were  examined e wet mount (native, sedimentation, and floatation) and (Ziehl Nellsen) stain. Protozoa was identified by using a light microscope with 400x and 1000x magnification. The result showed 68,89% of samples were positively infected by gastrointestinal protozoa. The protozoa were Blastocystis sp, Cryptosporidium sp, Giardia sp, and Eimeria sp. By chi square test, there was  not significant differences the prevalence of gastrointestinal protozoan in cat between sex, age, breed, and diarrhea status

Abstract

The aim of this research is to identify the prevalence of gastrointestinal track protozoa in local chicken (Gallus domesticus) located in Kramat Village, District of Bangkalan, Bangkalan Regency using fecal examination. The number of sample used were 140 including 70 samples from rice fields location and 70 samples from fisheries location. The result showed that 54 (38.6%) local chickens were infected by species of Eimeria; E. acervulina (2.5%), E. brunetti (22.8%), E. maxima (46.8%), E. mitis (1.3%), E. necatrix (22.8%), E. praecox (2.5%), and E. tenella (1.3%). The result was made of 16 (22.9%) local chickens in rice fields location and 38 (54.3%) local chickens in fisheries location. The infection of Eimeria sp. on male local chickens were 24 (34.3%) while on the female local chickens were 30 (42.9%). Chi Square Test showed that there was a highly significant difference toward the prevalence in rice fields and fishery locations (p<0.01), but there was no significant difference toward the prevalence of male and female local chickens (p>0.05).

Abstract

The purpose of this research to know cross reaction of Haemonchus contortus protein with anti-Fasciola gigantica serum by using the Western Blot technique. The result can be used as Haemonchosis serologic diagnostic. This research was conducted using Haemonchus contortus worms obtained from abomasum, especially cow worms and Fasciola gigantica worms obtained from the bile duct and gall bladder. The worms were crushed and added with Phosphate Buffer Saline (PBS) to make homogenates and then centrifuged at 3500 rpm for 15 minutes. Homogenate Fasciola gigantica was immunized to mice to get anti-Fasciola gigantica serum at a dose of 200 ?g/mice and booster 3 times with a span of two weeks. Homogenates were carried out using the SDS-PAGE technique to analyze proteins using a brilliant blue dye. Cross reaction of Haemonchus contortus protein with anti-Fasciola gigantica serum byusing Western blot technique and obtained 3 protein bands are 94.9, 52.7, 46.7 kDa.

Abstract

Fasciola gigantica is a worm which could infect breeding animal and human. One of the plants that can be used as worm drug is Pumpkin seed (Cucurbita moschata Durch).Pumpkin seed (Cucurbita moschata Durch) has been used in traditional anthelmintic medicine. This research was conducted to determine the anthelmintic effect of Pumpkin seed (Cucurbita moschata Durch) infusion on lethal death time of Fasciola gigantica in vitro. This research used Fasciola gigantica 25-75 mm in length without differentiating their sex. The concentrations of Pumpkin seed (Cucurbita moschata Durch) for treatment of Fasciola gigantica were 25%, 50% and 75%. The negative control used NaCl physiology (0.9 %). ANOVA factorial test showed significant difference among all of the experiment groups (p <0.05). Duncan multiple range test was seen the best treatment was the shortest lethal death time in concentration of 75%.Six hours of the treatment was the best time for making death of the worm. The Pumpkin seed (Cucurbita moschata Durch) in 75% dose was the best dose for making death of the worm. In 6th hours of the treatment in 75% dose administering had the best for making death of Fasciola gigantica. As for the reason, pumpkin seed (Cucurbita moschata Durch) can be use as anthelmintic.

Abstract

The purpose of this study was to determine the prevalence of gastrointesatinal parasites in cats at Surabaya shelter. Eighty two samples of cat feces used in this study were taken from Griya satwa shelter and Kiro cat shelter. Feces samples were examined using the native, sedimentation and floating methods. Feces samples gastrointestinal parasite were identified by using a microscope with 400x and 1000x magnifications. Faces samples that are diagnosed positively if there is at least one of the three methods. Feces samples has observed, obtained prevalence rate of 43.9% or 36 positive samples from eighty two samples. The results of chi square analysis showed that p > 0.05, which showed that the parasites taken in the two shelters had clear differences in the prevalence of the gastrointestinal tract parasites. The parasites observed were Isospora sp., Ancylostoma sp., Toxocara cati and Toxocara leonina.

Abstract

Broiler chicken farms could be a part of potential source infection Leucocytozoonosis and Plasmodiosis disease due to the death of livestock. Broiler farms in Tanjung Gunung, District Jombang near the fields, water, and trees. This could be increase the population of vectors infection Leucocytozoonosis and Plasmodiosis. The main purpose of this researchis to detect Leucocytozoonosis and Plasmodiosis infections on broiler farms in Tnjung Gunung Village, District Jombang using 50 broiler chickens by Purposive Sampling  of 2 bredeers. The result show rhat there are 6 positive samples infected Leucocytozoonosis, while Plasmodiosis infection could not be fond. Need cage sanitation to break life cycles of vector Leucocytozoonosis and Plasmodiosis.

Abstract

The aims of the research are to prove the anthelmintic effects of Ocimum sanctum Linn. leaves ethanol extract on Raillietina echinobothrida in vitro and to know the value of LC50 and LC90of Ocimum sanctum Linn. leaves ethanol extract. The research used completely randomized design. The research had five treatments and each treatment was done in four replications. The research used 5 Raillietina echinobothrida adult worms for each treatment and all replications. The observation and the recording were done at the first hour until the sixth hour of the research. The Raillietina echinobothrida adult worms were noted dead if there was no movement when they were disturbed by anatomic tweezer, the death of Raillietina echinobothrida were confirmed when they dipped mildly in warm water (50°C) and they showed no movements. The acquired data was analyzed using ANOVA and continued with LSD (5%). The results were Ocimum santum Linn. ethanol extract has the anthelmintic activity in 1.25%, 2.5%, 5% and 10% concentrations. The LC50 and LC90 was analyzed using probit analysis. The results of LC50 calculation were 7.8% at the third hour, 6% at the forth hour, 3.2% at the fifth hour, 2.5% at the sixth hour and the results of LC90 were 6.5% at the fifth hour, 5.2% at the sixth hour. The higher concentration of the extract, the higher anthelmintic activity worked.

Abstract

The aim of this research was to detect the presence of nematode worms that infects sacrificial cattle slaughtered duringEid al-Adha 1439 H in East Surabaya. This research used 24 sampels of sacrificial cattle and used digestive tract surgical method. Based on the result of the examination using a microscope with 100x magnification, there are positive samples infected with nematode worms. In caecum cattle consist of Oesophagostomum radiatum and Trichuris spp. In colon cattle consist of Oesophagostomum radiatum.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui daya anthelmintik dari ekstrak etanol daun kemangi (Ocimum americanum Linn.) terhadap mortalitas cacing Haemonchus contortus secara in vitro. Sediaan ekstrak dibuat dengan konsentrasi (berat/volume) 2,5%, 5%, 7,5%, dan 10%. Untuk kontrol positif digunakan PBS dan DMSO sedangkan untuk kontrol negatif digunakan Albendazole 10 mg/ml. Sebanyak 10 ekor cacing Haemonchus contortus betina direndam dengan berbagai perlakuan dalam cawan petri. Masing-masing perlakuan diulang sebanyak 4 kali. Pengamatan dilakukan setiap 1 jam sekali selama 6 jam untuk melihat apakah cacing masih hidup atau sudah mati. Hasil dari penelitian ini menunjukkan bahwa ekstrak daun kemangi memiliki efek anthelmintik terhadap cacing Haemonchus contortus secara in vitro. Makin tinggi konsentrasi ekstrak, maka daya anthelmintiknya juga semakin tinggi. Pada ekstrak konsentrasi 10% memiliki kemampuan anthelmintik yang hampir sama dengan Albendazole.
