Abstract

The aim of the study was to determine the changing of optical density of antibody of mice (Mus musculus) immunized with 40 μg of fraction 3 of goat zona pellucida (gZP3) as an anti fertility agent.  Each of the mice injected three times, once as the first immunization and twice as booster in 14 days interval respectively. The first immunization, gZP3 suspension was diluted with Complete Freund’s Adjuvant (CFA), meanwhile the booster gZP3 suspension was diluted with Incomplete Freund’s Adjuvant (IFA) 1:1. Blood samples collection for  serum were carried out four times, i.e. before first immunization, before first booster, before second booster and after second booster. The measurement of antibody mice in serum using Elisa technique indicated the increasing (p<0.05) of optical density serum from after the first immunization until before the second booster injection. It could be concluded that gZP3 was effective as an immuno contraceptive substance. Key Words : fraction-3 of goat zona pellucida, pregnancy rate, litter size.

Abstract

Sorghum species as forage crop may cause problem, because under certain condition it produces durrin, which will further breakdown to form prussic acid (HCN). Prussic acid are potentially toxic to ruminants, and the fatal concentration is 500 ppm. The aims of this research were to find out the role of cutting age as well as nitrogen (N) fertilization on the quality of sweet sorghum forage. The method were three cutting age (30, 40 and 60 days) and four level N fertilization (0, 100, 200 and 300 kg N/hectare). The quality of forage were observed of the content: Crude Protein (CP), Crude Fiber (CF) and HCN. The design applied was Factorial Randomized Block Design. The data were statistically evaluated by analysis of variances and significances beetwen means were tested by using honestly significant difference method. The result showed crops with moderate cutting ages (40 days) produce good quality of forage (CP = 13.65 %, CF = 28.17 %, HCN = 222–253 ppm) and optimum vegetative growth. Key words: forage, durrin, prussic acid, vegetative growth

Abstract

The aims of this research were finding the polymorphism phenomenon of diminazene aceturate in etawa breed goats by modus analysis of T1/2 b. The five  infected trypanosome goats and five healthies goats ware injected  5 mg/kg body weight diminazene aceturate by intramuscular administrated. The whole blood samples was obtained from sampling each 10, 30, 50, 70, 90, 120, 240, 720, 1440, 2880, 5760, 10080 minutes. The analytes in goats plasma was analysis by high performance liquid chromatography. The result showed that the pharmacogenetics profile of diminazene aceturate was bimode. Two mode being at  83.43 hours-1 and 122.5 hours –1. Key words: diminazene aceturate, bimodus, elimination half life

Abstract

The objective of the study was to know the repeated of AAIn to the same species of animal were not induces AAId and total immunoglobulin productions.  This study used 10 male rabbits divided into 4 groups as series trial.  Each rabbit in groups I, II, III and IV was injected subcutaneously with 0.5 ml of 1:10 antibody anti-inhibin per animal one time; Group II rabbit are injected twice by using the same dosage antibody anti-inhibin single dose, twice, thrice, fourfold every two week respectively. The antibody anti-idiotypic was measured by using indirect ELISA.  While the level of total immunoglobulins by using direct ELISA.  The result of the study showed that repeated injections of rabbit antibody anti-inhibin were not stimulated antibody anti-idiotypic in the same species and the total immunoglobulins of rabbit were not decreased. Key words: anti-inhibin, anti-idiotipic.

Abstract

Nine Ongole Crossbred steers were used in this research., average 1-1,5 years old and their body weight between 180-200 kgs.  The aim of this research was to investigate effects of  using different level  of chicken manure in haylase complete feed on N retained of Ongole Crossbreed  Steers ( PO ). Three different proportion of manure chicken in the same proportion of maize stalk 40% and concentrate 60% on dry matter basis which iso nitrogen (average crude protein 12%), R1: maize stalk 40% : concentrate 60%   (0% chicken manure), R2: maize stalk 40% : concentrate 60% (5% chicken manure), R3: maize stalk 40% : concentrate 60% (10% chicken manure).  Concentrates were consisted of rice polishing, wheat pollard, coconut cake, kapok seed cake, waste of cassava, mollases, urea and mineral. The research was in vivo by urine collection method.  Urine collection was done during 7 days on the last week collection period. During urine collection added H2SO4 10% to N fixation and then kept in the refrigerator. Urine samples were analyzed urine nitrogen by proximate analysis. Statistic analysis was Randomized Block Design (3x3) with body weight as covariance. The result showed that  there were not significantly difference (p>0,05) between each treatment on N retained of Ongole crossbreed steers. The lowest part of N retained was R3 and the highest was R2.  It was concluded that proportion of maize stalk 40% : concentrate 60% until 5% chicken manure can used as NPN source in haylase complete feed to Ongole crossbreed steers because there were not negative effect on N balance. It was suggested to use R2 for ruminant because it almost has the same value on N retained and more economic than R1. Key words: Complete feed, haylage, chicken manure, maize stalk

Abstract

This study examined the effects of Moringa oleifera (M oleifera) extract on the level of GABA and the function of GABA A receptors in mice brain. The mouse were divided into the control groups were given physiological saline orally, M oleifera groups received doses of 100; 200 and 400 mg/kg M oleifera extract (orally) respectively. 60 min after administration of M oleifera extract or saline, all of the mice brain was measurement GABA level by ELISA and GABA A receptors expression by Western blott analysis.  Moringa oleifera extract at dose 400 mg/kg BW significantly increased GABA levels and enhancement expression of GABA A receptors in the brain of mice. These results suggest that the anticonvulsant effect of M oleifera extract at dose 400 mg/kg is due to elevate of GABA levels bound to GABA A receptor. Keywords: Gamma Amino Butiric Acid, Anticonvulsant, Moringa oleifera

Abstract

The dermatitis-test and indirect-ELISA were the diagnostic technique that often used to diagnose toxocariasis. Therefore, both of these techniques were not efficient because they need a long time and have low specificity, especially if crude protein was used as an antigen. Double antibody sandwich-ELISA was used to develop ELISA technique to get the high sensitivity and specificity tests. For this technique needed a polyclonal antibody as coating.  This research used nine males Angora rabbit were divided into three groups. The first group was injected by adjuvant as a control. The second were injected by L2 immunogen of T. cati. The third were injected by the adult worm immunogen. There were two steps of injections, as follow: initiation step with CFA added to homogenate and the second step was booster immunization that was done two weeks after initiation, with IFA added to homogenate. The booster was done three times with two weeks interval. Sera was taken two weeks after the final booster. Antibody titers were 0 for the control, 40,960 for L2 immunization, and 20,480 for T. cati adult immunization. The positive optical density was 0.540 ± 0.050 sera with L2 homogenate immunization that statistically did not significantly difference (p>0.05) with T. cati adult homogenate immunization, was 0.528 ± 0.034.Therefore both of these results showed high significantly difference (p<0.01) with the control, was 0.208±0.004. Key words: polyclonal antibody, toxocariasis, sensitivity, specificity

Abstract

A study has been performed to find out the effect of  Acorus calamus Linn. rhizome extract on decreasing  the blood cortisol concentration in albino male rats which stimulated by electro shock. 30 Wistar albino male rats, weight 150-175 grams were, divided into 6 groups which received orally respectively PGA 3%, PGA 3% by electro shock, Acorus calamus Linn. rhizome extract 500 mg/kg BB, 1000 mg/kg BB, and 1500 mg/kg BB with electro shock, and diazepam 0,2 mg/kg BB as control also with electro shock. The blood samples were taken in a certain time to measure the blood cortisol concentration by fluorescent polarized immunoassay method. The data was  analyzed by Simple Anova, continued by HSD 5%, concluded that Acorus calamus Linn. rhizome extract showed the effect on decreasing blood cortisol concentration, and the biggest effect showed by the dose of 1500 mg/kg BB. Key words: Acorus calamus Linn. rhizome;  blood cortisol; albino male rats

Abstract

Objectives of the research were to know the influence of Zn and Cu suplemented at bioprocess by yeast of Saccharomyces cerevisiae to the content of nutrien fermented product and its change (dry matter, crude protein, crude fat, and crude fibre) and also the change of colony amount and pH.  The research was conducted experimentally with Completely Randomized Design with four treatment and each repeated by six times. Treatments consist of: S0 = ( 150 g of substrat + 0.2% Saccharomyces cerevisiae); S1 = ( 150 g of substrat + 0.2% Saccharomyces cerevisiae + 215.9 ml ZnCl2); S2 = ( 150 g of substrat + 0.2% Saccharomyces cerevisiae + 47.22 ml CuSO4); and S3=   (150 g of substrat + 0.2% Saccharomyces cerevisiae + 22.9 ml ZnCl2 + 23.5 ml CuSO4).  Substrats consist of corn and soy bean meal with 20% crude protein.  Result of  the research shows there is a high significant (p<0.05)  increasing of dry matter, crude protein, and crude fat; and also colony amount and pH at the treatments which are suplemented with mineral of Zn and Cu compare to treatments without of suplemented.  The crude fibre was decreases but not significant (p>0.05) with treatment without mineral suplemented. Abstract

The aim of this present study was to investigate the progressive motility and membrane integrity of cat epididymal spermatozoa during storage at 4°C.  Spermatozoa was collected by flushing technique with physiological saline and diluted in Tris egg yolk extender for three days.  The result showed that mean concentration of spermatozoa from cauda epididymal was 108,40 ± 81,49.106spz/ml.  The percentage of progressive motility and membrane integrity of spermatozoa on day-0 (time of collection) was 74,50 ± 3,50 % and  80,82 ± 2,00 %, respectively.  The percentage of progressive motility on day-1 was 62.5 ± 5.59 % and significantly higher(P<0,05)  than day-2 (50.5 ± 5.68 %) and  day-3 (25.5 ± 9.60 %).  There was also significantly different on the percentage of membrane integrity on day-1(71.54 ± 2.50 %), day-2 (64.16 ± 4.15 %) and day-3 (50.03 ± 4.16 %) of storage at 4°C.  In conclusion ,the spermatozoa derived from cat cauda epididymal could be used for artificial insemination after 2 days of storage at  4°C. Key words: epididymal sperm, storage, cat
